Prevalence and diversity of Cryptosporidium infecting fur animals

The object of this thesis was evaluation of occurrence and prevalence of Cryptosporidium infection in fur animals, mainly American mink, fox and chinchilla. A total 370 individual specimens originated from mink (n = 340), fox (n = 18) and chinchilla (n = 12) were collected. While microscopy examination did not proved any presence of Cryptosporidium oocyst in fecal samples, molecular tolls based on amplification of small ribosomal subunit and 60 kDa glycoprotein of Cryptosporidium revealed three positive samples in minks. Following phylogeny analyses of both loci showed presence of C. ubiquitum of the XIIa1 family subtype in all positive samples. The XIIa1 family subtype was detected in Carnivores for the first time. No correlation between Cryptosporidium infection and presence of diarrhea was observed in this thesis.

This thesis deals with the microscopic analysis of honey and its use in the evaluation and testing of honey. The work is divided into theoretical and practical parts. The theoretical part is devoted to the analysis of pollen and physical and chemical methods of determining the type of honey. The practical part describes the results and compares them. 22 honey samples originating from beekeepers has been shown 17 beekeepers false claims about their type of honey. Of the 7 aldulterated honeys, 4 samples were violated by foreign particles from the environment. Further honeys from the market containing suspicious particles and very critically low pollen grains in 1 g of honey, which points to a adulteration of honey. Unifloral honeys from Olomouc were found honey from satureja (S/2) and lavandula (F/1) containing neither one pollen grain of these plants.

The bovine mammary gland is the main production unit of dairy cattle. Due to its exploitation is often threatened its integrity and homeostasis. Therefore, mastitis are most common health problem of dairy cattle. Significant defensive elements of bovinne mammary gland are leukocytes: neutrophils, monocytes, macrophages and lymphocytes. The structural characteristics of these cells are closely related to the functional parameters such as migration and phagocytosis. Therefore, the work is focused on the structure and ultrastructure of these cells in the blood stream and in the ductal system of the mammary gland. The work also describes the pathological structure of these cells, including terminal stages of cell's life: necrosis and apoptosis.
