Using of organism Eisenia foetida in ecotoxicity tests

Pesticide products used for plant protection are one of the chemicals that are intentionally brought into the environment. In Czech Republic there are legally used only those pesticide products, which are registered by SRS (State Phytosanitary Administration). For registration of these products there is required in addition to assessing the likely negative effects on human and animal health, also information whether the pesticide products negatively not affect the environment, in particular avoiding the contamination of aqua system or not effect the non target organisms. This master's thesis discuss the use of organism Eisenia foetida (redworm) in the contact ecotoxicity tests by testing pesticide products. There were selected such pesticide products where there are not solid information about their impact on non-target soil organism (Eisenia foetida) and based on literature research was chosen ecotoxicity test by OECD 207 method, the acute toxicity test which is in place for 14 days. Two pesticide products Topsin M 500 SC (active ingredient thiophanate-methyl) and Perfekthion (active ingredient dimethoate) were tested. Ecotoxicological values were calculated and ecotoxicity of such peticide products for soil organism were avaulated.
Utilization of organism Daphnia magna in ecotoxicity tests

This thesis deals with ecotoxicity tests of selected substances used in agriculture and horticulture. It is focused on the determination of acute and chronic toxicity of selected herbicides. These herbicides are sold commercially, specifically Finalsan (active substance is nonanoic acid), the product of Neudorff GmbH KG company, and Dominator (active substance is glyphosate, isopropylamine salt) by Dow AgroSciences Ltd. Next tested substances are potassium nitrate and sodium nitrate - common agricultural fertilizers. Potassium dichromate was also tested. Potassium dichromate is very toxic for aquatic environment and it is used as the standard in acute tests. Tests were carried out in terms of effects on aquatic systems, due to very good solubility in water of selected substances. The tests were performed on the organism Daphnia magna.
The use of algal test in ecotoxicology

This diploma thesis deals with a use of algal test in ecotoxicology. A freshwater algae, Desmodesmus subspicatus, was used to asses the ecotoxicity of selected chemical substances. A spectrophotometry method, of VIS spectrum, was used in evaluation of the EC50. A correlation, between a number of algal cells and the wavelength absorbance at 683 nm, has been determined for this purpose. Following chemicals were tested, 2-[2-[(2,6-dichlorophenyl)amino]phenyl]acetic acid, (RS)-2-[4-(2-methylpropyl)phenyl]propanoic acid and N-(4-hydro-xyphenyl)acetamide. An EC50 value was determined for all these substances and also their ecotoxicity has been evaluated.
Ecotoxicological evaluation of sediments

Sediments of lotic and lentic water can be a reservoir of a number of contaminants, which can enter the aquatic ecosystem either due to the controlled discharge of waste water or leakage of various accidents or technological devices. Sediments can hereby affect not only the aquatic ecosystem, but also the terrestrial system in the case that they will be used as fertilizer or as a material for the recultivation. For this reason it is important to consider their potential ecotoxicity. Sediments were subjected to ecotoxicological bioassays and their potential ecotoxicity was assessed. The phytotoxicity assays on terrestrial plants Sinapis alba, Lactuca sativa and Allium cepa were used and the effects of aqueous extracts of sediments on the inhibition of growth their root were evaluated and the value of IC50 was determined. For the ecotoxicological assessment of sediments and determination of IC50 value also Lemna minor as a representative of aquatic plants was selected. For more objective assessment of the sediment ecotoxicity aquatic invertebrates as Artemia salina and Daphnia magna were selected and the value of LC50 or EC50 was determined.
The ecotoxicological evaluation of musk compounds

This master’s thesis is focused on evaluation of synthetic musk fragrances using ecotoxicity tests. Four musk fragrances were tested – Galaxolide, Tonalide, Musk ketone and Musk xylene. Four alternative ecotoxicity tests with living organisms, one standard ecotoxicity test and three fytotests were used for evaluation of ecotoxicity. Alternative tests were performed on crustaceans Daphnia magna and Thamnocephalus platyurus, brine shrimp Artemia salina and rotifer Brachionus calicyflorus. Root growth inhibition test of Sinapis alba, root growth inhibition test of Lactuca sativa, root growth inhibition test of Allium cepa and growth inhibition test of Lemna minor were performed in the terms of fytotests. On the basis of the results the values of EC50, IC50 and LC50 were determined and the ecotoxicity of musk compounds was evaluated.
Evaluation of ecotoxicity of selected pharmaceuticals

In recent years consumption of drugs and thus the quantity of their residues in natural ecosystems is increasing. It is necessary to deal with their possible negative effects on the environment. This thesis is focused on the ecotoxicological evaluation of pharmaceuticals. Diclofenac and ibuprofen from the group of non-steroidal anti-inflammatory substances, penicillin G and ampicillin from the group of antibiotics were tested. Mainly tests on aquatic organisms were used to evaluate of ecotoxicity, because the tested pharmaceuticals are predominantly occurred in the aquatic environment. Daphnia magna, Thamnocephalus platyurus, Artemia salina, Sinapis alba, Lemna minor, and Brachionus calyciflorus were used as testing organisms. Values of LC50, EC50 and IC50 were determinated and the ecotoxicity of pharmaceuticals was compared.
Assessment of the effect of formaldehyde resin monomers on the environment

Thousand of new substances with unknown environmental effect are produced and used daily. Many of them are deliberately or by negligence released and deposited into the environment where they could undergo different transport and degradation mechanisms. They are able to affect different types of organisms, as well as humans. For this reason, the awareness of the problems, associated with pollution of all environmental compartments is growing. The environmental impact of monomer formaldehyde resins, which are produced for commercial use, on the environment were studied in this research. For the purposes of ecotoxicological evaluation samples of melamine-formaldehyde and urea-formaldehyde resin were chosen and three toxicity tests were run: acute test with measurement of inhibition of the mobility of Daphnia magna, the acute test with luminescent bacteria Vibrio fischeri and acute test with measurement of inhibition of oxygen consumption by activated sludge. For more comprehensive assessment of the impact of these substances on the environment, biodegradability was also determined. Both tested substances showed minor hazardous impact to testing organisms and it can be concluded, that these substances should not present a significant risk to the environment.
The usability of algal test for evaluation of ecotoxicity

This diploma thesis deals with the evaluation of algal test methods and passes judgement on their availibility for ecotoxicological analysis of chosen chemical substances. By counting of algal cells and spectrophotometry was determined influence of potassium bichromate on growth of freshwater algae Desmodesmus subspicatus.
Ecotoxicological evaluation of the sludges from water treatment plants

Evaluation of the sewage sludges from different sewage water treatments will be performed by means of available ecotoxicity tests. The evaluation will be done in accordance with the valid legislation. On the basis of the obtained values the sludges will be classified into the respective grades of leachability and the dangerous property of the sewage sludges ecotoxicity will be evaluated.
Influence of mining on environment in respect to ecotoxicology

This thesis deals with an influence of mining on environment from the point of view of ecotoxicology. First focus in the thesis is on overall impact of coal mining on environment. The thesis is especially closely focusing on mine water, its formation and negative influence on aquatic ecosystem. In this connection the thesis mentions examples of ecotoxicological evaluation, so called acidic mining water. The thesis also mentions legislative scope that is related to the issues about protecting environment against impact from coal mining and subject dedicated to restoration of affected areas.
