Methodist School-Based Prevention and his Role in Practical Elementary School and Special Elementary School

The diploma thesis focuses at a theme of Methodology of school prevention and its role at the specialised primary school. The theoretical part is split into four main chapters which refers to the education system of children with need of special educative care in the Czech Republic. It defines socially pathological aspects including risk behaviour with its prevention. The last chapter describes primary prevention at schools using a minimal preventive program and work of a methodologist of school prevention. The practical part contains a quality-type research based on semistructural interviews with methodologists of school prevention at chosen specialised primary schools. The first task of my diploma thesis is to find out what advise parents need and ask for the most. The other task is to find out what aspects a methodologist of school prevention has to take into consideration during issuing a minimal preventive program. The questions have been asked in the way to find out how cooperation at the level of prevention between parents and specialised primary school works, and who else among school staff except of a methodologist participates in the issuing a minimal prevention program. I have contacted sixteen specialised primary schools in the South Bohemia region for the interview. Ten have replied, but only six of them were willing to cooperate further. Two of the schools are located in the regional city, the other four are from smaller cities of similar sizes. Interviews were carried out with methodologists of school prevention and questions were focused at the cooperation at the level of prevention between parents and specialised primary school, and possible influence of location of the school which could be reflected in the minimal prevention program.

Morphological properties, species specific differences, development, and function of egg envelopes are of importance for a better understanding of fertilization strategies as well as for captive reproduction. We reviewed morphology and the developmental stages of egg envelopes, mechanism of polyspermy block, and also the role of micropyle and cortical granules in polyspermy block for acipenserid eggs. The structure of the egg envelope is similar among the acipenserids, comprising an external envelope (thecal cells, basal lamina, and follicular epithelium), a five-layered oocyte envelope (adhesive layer, alveolar layer, epilayer, and zona radiata externa and interna) and a layer of oocyte matrix and cortical granules. The development of acipenserid egg envelope within the ovary comprises five stages, with further changes following fertilization. Moreover, we standardized the terminology used to describe the egg membranes which can minimize the confusions and be helpful for future work on acipenserids eggs as well as for other fish species. The sperm to egg ratio (required to fertilize eggs) and effects of pre-incubation of eggs in freshwater before fertilization were studied to standardized fertilization protocols for sterlet Acipenser ruthenus. Pre-incubation time had no effect on fertilization success at 430,000:1 and 43,000:1 sperm to egg ratios, while it was significant at the 4300:1 and 430:1 ratios. The use of adequate experimental suboptimal sperm to egg ratio revealed a positive effect of pre-incubation time, such that at the 430:1 ratio, 0.5 min pre-incubation increased the fertilization rate than 10 min. Transmission electron microscopy showed that pre-incubation of eggs in water for <10 min did not trigger a cortical reaction, suggesting that a low sperm to egg ratio 0.5 to 1 min pre-incubation of eggs in freshwater prior to fertilization can enhance fertilization rate of sterlet (minimally do not change fertilization). The effects of pre-incubation in seawater and the duration of egg receptivity were determined for fresh and over-ripe sea bass eggs. Our results revealed a significant effect of pre-incubation time for both the fresh (P < 0.01) and over-ripe eggs (P < 0.01). The fresh eggs had a higher fertilization success than over-ripe eggs. Fertilization success of eggs significantly declined for both of these treatments after 3 min of pre-incubation clearly indicated that sea bass eggs are able to be fertilized by sperm for up to 3 min after release into seawater. Effects of pre-incubation of eggs and activation medium on the percentage of eyed embryos for Ide Leuciscus idus were examined. At the eyed-egg stage, pre-incubation time was significant for the freshwater activation medium (P < 0.001), such that the percentage of eyed embryos declined across the pre-incubation time gradient. Activating medium had a significant effect on the percentage of eyed embryos for each pre-incubation time (P < 0.05). More precisely, freshwater produced the lowest percentage of eyed embryos at all pre-incubation times, whereas saline water and Woynarovich solution produced the highest percentage of eyed embryos at 0 s and 30 s before incubation. Examination of sperm traits showed no impact of activating medium on computer assisted sperm analysis parameters. Our results suggested that saline water or Woynarovich solution improve fertilization rate in Ide during fertilization. In conclusion, this thesis provides basic knowledge on gamete biology, role of egg receptivity period, effects of pre-incubation of eggs and different activating medium which can be useful to understand the fertilization strategies of different externally fertilizing fishes as well as standardize their fertilization protocol for captive reproduction.

Territorial Administration in the Czech Republic - the direct election of mayors

The starting point of the diploma thesis is a discussion of the direct election of mayors in the Czech Republic. The aim of the thesis in this context is to assess whether the direct election of mayors is appropriate in the normative framework of territorial administration of Czech municipalities and if so, under what circumstances. A partial aim of the work, intended to lead to the fulfilment of the main aim, is the description of the formal role of mayors in the administration of municipalities, the description of the manner of appointment of mayors into their positions, as well as the description of the anchoring of the position and election of mayors of municipalities in selected European countries. Another partial aim is the description of starting points for the direct election of mayors, stemming from the attitudes of mayors of selected municipalities. The subject of study will be the normative setting of the mayors position in the regional administration in the Czech Republic and simultaneously the manner of their election. Another subject of study will be the attitudes of various mayors in relation to the issue of their formal position and election. The data on the mayors attitudes will be obtained by the focus group technique. The criterion for the selection of respondents will be the municipality size. The purpose of the work is to generalize the data obtained and to assess it on the basis of the prevailing opinion.

Revealing phosphoproteins playing role in tobacco pollen activated in vitro

5 Abstract Tobacco mature pollen rehydrates in vivo on a stigma tissue, and develops into the rapidly-growing pollen tube. This rehydration process is accompanied by the de-repression of stored mRNA transcripts, resulting in the synthesis of novel proteins. Furthermore, such metabolic switch is also likely to be regulated on the level of post-translational modifications of the already-present proteins, namely via phosphorylation, since it was shown to play a significant regulatory role in numerous cellular processes. Since only a minor part of proteins is phosphorylated in a cell at a time, the employment of various enrichment techniques is usually of key importance. In this diploma project, metal oxide/hydroxide affinity chromatography (MOAC) with aluminium hydroxide matrix was applied in order to enrich phosphoproteins from the mature pollen and the 30-minute in vitro activated pollen crude protein extracts. The enriched fraction was separated by both 2D-GE and gel-free liquid chromatography (LC) approaches with subsequent mass spectrometric analyses. Collectively, 139 phosphoprotein candidates were identified. The Motivation as a Leadership

Abstract This bachelor thesis deals with the topic of Motivation as a way of keeping people. In the theoretical part is through the help of key words a reader familiar with the concepts of manager, management the role, management functions, leadership, incentive mechanism, participation, motivation the program benefits. After getting acquainted with these concepts, which are further developed and properly explained it is compiled by a questionnaire consisting of 13 questions dealing with motivation and benefits in the workplace. The questionnaire is submitted to the selected five managers. The three owners or co- owners private companies, the remaining two are employed in the public sector. With the questionnaire were brought to the attention of the three women and two men. The individual answers have been commenting and at the end summarized into a single conclusion. From this conclusion it follows that the motivational processes and benefits in individual organisations are set up similarly and that the best motivator is financial reward, or the public praise. Furthermore, it was found: managers prefer individual approach to leadership, a good team and the pleasant environment of the employees want rather than some of the other motivators. Last but not least can from the responses of most managers sense that you may want a the personal development of the employees, but only in a given field. Part of the managers do not agree with the further education of employees due to concerns about competition. The male part of the managers used a rather shorter sharper the answer to the question, the staff applies a rather authoritative way of leadership, while the female part of the managers was rather longer and more sentence. From the responses it is apparent that women apply to their employees rather democratic style of leadership. Differences between the Common law and the Continental law and their practical consequences

This Bachelors thesis analyses the differences between the Common law and the Continental law and their practical consequences and let to understand better how this two systems work in general and how crossed between each other in cases. How they could be used in the area of countries, which are under their influence and show to the readers how he or she can use the law in their own favour in practice. In the theoretical part is described the general definition of law the Common law and Continental law the features, history and development of both systems of law. Examined the role and functions of lawyers, judges and types of juries in each system. Represented the sources of Common law and Continental law. It is contain all necessary information for reader to make understand all questions which can appear. The practical part consist of Case Study of the Court Proceeding (Great Britain and Czech Republic). It examined courts and court proceeding in each system .The comparison between them and their lawyers. The results are analyzed and supplemented with comments. In conclusion of bachelors thesis discussed the whole summary of the work, which results were gotten and added comments and recommendations of author.

Regional Policy and its Role in Development of Pardubický Region

The Czech Republic through its European Union membership obtains a possibility at development projects to draw on financial support from the European funds. This diploma thesis deals with the regional policy in context of the European funds and the regional policy effect on the Pardubice Region development. The Regional policy (including its principles, tools and targets) and an evaluation of draw on possibilities of European subsidy instruments for regional development (in framework of Regional Operation Programmes) are described in the first part of the thesis. The second (main) part deals with an analysis of the projects realized in particular counties of the Pardubice Region during programme period 2007 2013. This analysis is realized with theoretical and practical experiences and based on available data and documents from Regional Council NUTS 2 Northeast. The main part of the thesis also contains a particular projects comparison based on information which had been obtained in interview with managers of the projects. This comparison gives information related to the impact on regional policy in the Pardubice Region.

The coaching and its importance for leadership

This bachelor thesis deals with the coaching and its importance for personal leadership. The first part of the research in literary sources clarifies the fundamental term of coaching and its application. Consecutively, it also explains different models of coaching. Next topic of the literary research is the figure of coach, his role and qualities, and also the characteristics of internal and external coach. The last part of the literary research is dedicated to the issues of personal leadership and management. The core of this thesis uses the pen and paper interview with managers of the ČEZ company to examine the quality of coaching and personal leadership in one of the biggest companies in the Czech Republic, how managers perceive additional training courses, and also to survey the overall leading structure of ČEZ. In the conclusion I summarize the acquired findings.

Domestication of the horse and its spread cultural world

The horse was given to the people with the help of a long and complex process called domestication. He played in our history a very important role. Thanks to him, the human population is widespread almost all over the world. First, the horse was a source of food and later began to use the horse's ability to carry heavy loads or carry riders across the landscape. These animals were participants of all world wars, even the largest ones. Thanks riding became effective military. Evidence of this process can be found in caves, on ceramic products, paintings, but also in books. Today horses as working animals largely replaced the machine, but also horses remain in the care of a man for his leisure. This work deals with the domestication of the horse from its inception, practically up to the present.
