The role of cytoskeleton in auxin transport

Auxins are a class of plant hormones (phytohormones) with their most frequently endogenously occurring representative indol-3-acetic acid (IAA). Because of their influence on division and elongation of cells, auxins play an important role in many developmental and physiological processes such as embryo development, vascular tissue patterning and tropisms. These effects are often mediated by polar auxin transport, which results in a wide variety of auxin concentrations in cells and entire tissues. Transport of auxin from cell to cell is partly mediated by diffusion, the prevalence of auxin transport is however mediated by auxin carriers located on plasma membrane (PM). Among such carriers belong AUX1/LAX (AUXIN RESISTANT 1/LIKE AUX1) transporter family, which helps with auxin influx and families of PIN-FORMED (PIN) and ABCB/PGP (ATP-binding cassette subfamily B/P-glycoprotein) transporters, which take part in auxin efflux. These proteins are in various degrees dependent on a system of membrane vesicles, trafficking along actin cytoskeleton, which ensures among others cycling of these vesicles among PM and endosomal cell space. Cellular localization and functional characterization of TTL proteins of Arabidopsis

This thesis focuses on the study of TTL protein family in the model species Arabidopsis thaliana. It sums up available published data on this so far poorly studied genes and presents newly gained experimental data. Main emphasis is given to TTLs cellular localization in possible relationship to their physiological function. Presented thesis comprises of in silico analysis of TTL proteins, but also the creation and microscopy analysis of TTL-mRuby2 and TTL-GFP fusion proteins in transient and permanent transformants. Interactions of Mitochondria with Other Cellular Structures

In cells, mitochondria fulfil diverse set of roles, including a production of energy-rich molecules, which are necessary for proper functioning of the cell, calcium homeostasis, apoptosis and even biosynthesis of Fe-S centres, heme and steroids. To coordinate some of these processes with events occurring in the rest of the cell, mitochondria need to communicate with the other cellular structures through their physical contacts. Resulting intracellular platforms give rise to additional mitochondrial functions. This thesis summarizes current findings from the cells of mammalian model organisms and the budding yeast Saccharomyces cerevisiae about the interactions of this semiautonomous organelle with other cellular components and about the functions, which these interactions mediate.
Visualization of cell structures in leaf cells of Malus domestica as a tool for study of Malus-Venturia inaequalis interactions

Apple scab, the most serious disease of apple is caused by fungal pathogen Venturia inaequalis. Knowledge about the apple response to apple scab attack on the cellular and tissue level is insufficient. For studies of Malus-Venturia interaction on the cellular and tissue level, the establishment of methods for cell structures visualization in apple leaves is necessary. In this work, the experimental plant material grown in vitro and ex vitro was successfully established and the method of apple infection by conidia of V. inaequalis was optimized. Various methods of cell components visualization such as vital staining, in situ immunolocalization, transformation, environmental scanning electron microscopy and confocal microscopy, were tested. Cell structures, such as the cytoskeleton, the cell wall and the cuticle were visualized in apple leaves. Preliminary experiments following specific the changes of cell wall structures induced by V. inaequalis attack were performed. Further, changes of cuticle structure, the first barrier for penetration of pathogen to plant tissues during infection, were observed during the leaf ontogenesis. Powered by TCPDF (www.tcpdf.org)
Searching for mechanisms and functions of microtubular interactions with other plant cell structures

Microtubular cytoskeleton is involved in many processes in plant cells, including cell division, growth and development. Other proteins enable its functions by modulation of its dynamics and organization and by mediation of functional and structural interaction with other cell structures. Identification of the mediating proteins and the functions of these interactions under specific conditions were the main aims of the thesis. Membrane proteins interacting with microtubules were identified using biochemical methods. Surprisingly, the identified proteins co-sedimenting with microtubules were not members of the "classical" microtubule associated proteins (MAPs). There were enzymes, chaperones and plant specific proteins among them. For further studies, the identified microtubule-associated heat-shock protein 90 (Hsp90_MT) was chosen. Recombinant Hsp90_MT binds directly to microtubules and tubulin dimers in vitro. The ATP-binding pocket is not responsible for this association. In BY-2, Hsp90_MT co-localizes with phragmoplast and cortical microtubules and is involved in microtubule recovery after their depolymerization during cold treatment. Role of formins in the organization and dynamics of intracellular structures in Arabidopsis thaliana

On the basis of detailed phenotypic examination of fh1 and fh2 mutants we observed that the main housekeeping Arabidopsis thaliana formin AtFH1 (At3g25500) and its closest relative, AtFH2 (At2g43800) are involved in both actin filaments and microtubule dynamics. fh1 mutants showed increased sensitivity to the actin polymerization inhibitor Latrunculin B (LatB). Formin mutants had cotyledon pavement cells which exhibited more pronounced lobes compared to the wild type, and alterations in vascular tissue patterning were found. The double fh1 fh2 homozygote was not obtained, suggesting that at least one functional formin gene is required for proper gametophyte development. Methods used to observe and quantify both architecture and dynamics of the cortical cytoskeleton from confocal laser scanning microscopy (CLSM) and variable angle epifluorescence microscopy (VAEM) were standarized and allowed to find that mutants exhibited more abundant but less dynamic F- actin bundles and more dynamic microtubules than wild type seedlings, fh1 mutant phenotype observed in roots was further aggravated by a (heterozygous) fh2 mutation. This bachelor thesis discusses the phenomenon of cellular senescence in the context of cytoskeleton organization. Differences in the organization of cytoskeleton be- tween normal proliferative cells and senescent cells are being compared. Cellular cytoskele- ton is a very dynamic structure and influences the function of the cell within a tissue. This thesis gathers current evidence about senescence and cytoskeleton and indicates possible directions for future research. Sperm morphology and molecular mechanism associated with sperm fertilizing ability in mice.

The process of sperm capacitation and acrosome reaction (AR) are highly dynamic processes essential for the fertilization, including cytoskeleton proteins in the sperm head. The study of the distribution of actin, spectrin and α-tubulin in the head of representative mammalian sperm indicates that before and after the AR there were changes in the distribution of the cytoskeleton structures in the sperm head of all representatives, mainly in the apical part of the acrosome, but also in the equatorial and postacrosome part of the sperm head. The particularly well-studied actin cytoskeleton plays an important role during the capacitation and before and after the AR. Environmental estrogens can interfere with the function of endogenous hormones in very low concentrations. They can interact with specific receptors affecting several signalling pathways leading to sperm capacitation and AR. The effect of 17β-estradiol, estrone, estriol and synthetic 17α-ethynylestradiol was evaluated by the ratio of tyrosine phosphorylation and the state of the acrosome during in vitro capacitation in the sperm head. Role of cytoskeleton in plant cell morphogenesis

The cells are able to acquire variety of shapes, in which cytoskeleton plays an important role. Cytoskeleton influences deposition of cell wall materials, regulates vesicle movement in cell, participates in exocytosis and endocytosis. Cortical microtubules affect celulose accumulation in cell wall and determine direction of cell expansion, although the exact connection between microtubules and cellulose remains unclear. Actin promotes growth and contributes to its spatial regulation in both tip ang diffuse growing cells. Actin is important for secretion in expanding cells but its exact functions in cell growth regulation are not explained yet. Analysis of mutants, spectroscopic methods, cytoskeletal drugs, fluorescence proteins and other methods are used to better understand how actin and microtubule cytoskeleton are integrated during plant cell morphogenesis. Epidermal and trichome cells of Arabidopsis thaliana are a good model of research and they are used for most studies.
Cytoplasmic membrane and tonoplast dynamics during closing and opening of stomata

Stomata are epidermal structures mediating regulated contact of plant apoplast with surrounding environment via stomatal opening. Change of turgor plays crucial part in initiation of stomatal opening or closure. During stomatal movement, guard cell undergo considerable and repetitive changes in cell volume and consequently surface area over a period of minutes. Alteration in the surface of membrane must occurs due to limited stretching capability of the plasma membrane. It can be achieved through membrane invaginations and endocytosis of small vesicles. Microscopy and electrophysiology techniques have proven that both processes are happening in guard cells. These processes are controlled and managed by complex web of signal pathways in which actin and microtubular cytoskeletons, SNARE proteins, ion channels and others molecules have crucial parts. The aim of this work is to summarize current knowledge on the processes and mechanisms of these membranes surface changes and their molecular principle.
