Title: Procedurally Generated Volumetric Cloudscapes for Unity Author: Jan Koblı́žek Department: Department of Software and Computer Science Education Supervisor: Mgr. Martin Kahoun, Department of Software and Computer Science Edu- cation Abstract: The traditional approach to cloud rendering in computer games is based on static sky- boxes or a set of static textures. Volumetric clouds used to be too computationally expensive, but with advances in GPU performance, they were successfully used in recent gaming titles. This thesis presents an implementation of real-time volumetric clouds for the Unity game engine. Clouds are described by multiple textures (both 3-dimensional and 2-dimensional) and rendered using a ray marching algorithm. The resulting implementation allows three types of low altitude clouds - cumulus, stra- tocumulus and stratus. The user can seamlessly transition between different coverages, times of the day, and animate clouds based of the speed and direction of the wind. Stimuli - responsive nanoparticles with boronic ester linkages as a potential carrier for drug delivery

This thesis deals with the post-polymerization functionalization of a pH-responsive poly(styrene)-b-poly(4-vinyl pyridine)-b-poly(ethylene oxide) triblock terpolymer with stimuli responsive and diol-binding 2-bromomethyl-4-fluorophenylboronic acid and 2- bromomethyl-phenylboronic acid suitable for biomedical applications. A reproducible method of quaternization of poly(4-vinyl pyridine) block with prescriptible quaternization ratio was developed and the reaction yield was analyzed by 11 B, 1 H NMR and infrared spectroscopy. A reproducible self-assembly protocol for stable nanoparticles from functionalized stimuli-responsive triblock terpolymer was found, and the nanoparticles were studied by static, dynamic and electrophoretic light scattering, and cryogenic transmission electron microscopy. Fluorescence and UV-VIS spectroscopy measurements were performed for description of boron-ester linkages formation and hydrolysis with model drug Alizarin as a study of encapsulation and release reactions together with sugar sensing reactions for potential drug delivery.
Phase-resolved Brillouin light scattering: development and applications

Spinové vlny mají potenciál být použity jako nová platforma pro přenos a zpracování dat, protože mohou dosáhnout vlnových délek v rozsahu nanometrů a frekvencí v rozsahu terahertzů. K tomu, aby bylo možné navrhnout zařízení a logické obvody založené na spinových vlnách, je zapotřebí získat informace o prostorovém rozložení intenzity spinové vlny a pokud je to možné, také o jejich fázi. To lze měřit pomocí fázově rozlišeného fokuso-vaného Brillouinova rozptylu světla (µ -BLS). Předložená práce se zabývá rozšířením stávající optické sestavy o možnost měření fáze, kde doposud bylo možné měřit pouze intenzitu. Toto rozšíření sestavy je důkladně popsáno a charakterizováno. Schopnosti optické sestavy jsou demonstrovány ve studii šíření spinových vln skrz Néelovu doménovou stěnu. Získané 2D mapy intenzity spinových vln ukazují, že propagace přes doménovou stěnu je ovlivněna topologicky vynucenou kruhovou Blochovou čarou ve středu doménové stěny a že režim propagace závisí na frekvenci spinových vln. V prvním režimu propagace se vytvoří dva svazky spinových vlny šířící se kolem kruhové Blochovy čáry, zatímco ve druhém režimu se spinové vlny šíří pouze středem. Fázově rozlišené µ-BLS měření odhaluje fázový po- sun spinových vln pro oba režimy. Mikromagnetické modelování spinových vln ukazuje rozrušení jejich fázových vlnoploch, které je třeba brát v úvahu při interpretaci měření a navrhování potenciálních zařízení. Mikromagnetické simulace ukazují, že vnější magnetické pole může být použito k pohybu kruhové Blochovy čáry ve stěně domény, a tedy k manipulaci spinových vln.
Beating Intracellular Bacterial Infections with Polymeric Nanobead-Based Interventions: Development, Structure Characterization, and Analysis

One hundred years after the discovery of antimicrobials and antibiotics, intracellular bacterial pathogens remain a major cause of global morbidity and mortality. This is due to the complex and intricate ability of these pathogens to undergo intracellular replication while evading host cell immune defense. Bacterial agents such as Legionella pneumophila, Francisella tularensis, and Mycobacterium tuberculosis, as the causative agents of Legionnaires' disease, pulmonary tularemia, and tuberculosis (TB), respectively, contribute to this burden. Moreover, these agents are weaponizable pathogens due to their aerosolizability. TB represents a global health problem, although a potentially curative therapy has been available for approximately 50 years; this intracellular disease affects approximately 1 in 3 people worldwide, with over 10 million new cases per year and one death every three minutes. TB can usually be treated with a 6- to 9-month course of combined therapy. The necessity of using a cocktail of anti-TB drugs and the long-term treatment schedules required for conventional therapy, however, result in poor patient compliance; therefore, the risk of treatment failure and relapses is higher. Multicompartment nanoparticles based on ABC triblock terpolymers as potential drug delivery vehicels

This thesis deals with the self-assembly behavior of newly synthetized well-defined amphiphilic poly(acrylic acid)-b-poly(4-hydroxystyrene)-b-poly(styrene-carborane) triblock terpolymer in aqueous solution. A reproducible method was developed for the preparation of diverse multicompartment boron-containing nanostructures (spherical micelles, wormlike micelles) depending on the initial solvent composition and solvophobic-solvophilic interactions. Self-assembled nanostructures were characterized by various techniques (static and dynamic light scattering, cryogenic transmission-electron microscopy and fluorescence spectroscopy). The solubilization of two model drugs (2-methylnaphtalene and 7- methoxycoumarine-carborane fluorophores) in polymer nanoparticles was studied by fluorescence spectroscopy to investigate its potential in drugs solubilization and delivery.

DOCTORAL THESIS ABSTRACT 2-Oxazoline triblock copolymers with hydrophilic, lipophilic and fluorophilic blocks: from synthesis to hierarchical self-assembly MSc. Leonid Kaberov The focus of this research was on the study of di- and triblock poly(2-oxazoline) copolymers with fluorinated blocks. The synthesis and solution properties of novel copolymers combining hydrophilic, hydrophobic (lipophilic) and fluorophilic moieties into one segmented molecule were reported. The simple synthetic approach which provides an easy way to attach a CnF2n+1 terminal chain to a poly(2-methyl-2-oxazoline)-block-poly(2-n-octyl-2-oxazoline) copolymer was described. Small-angle neutron and x-ray scattering experiments unambiguously proved the existence of polymersomes, worm-like micelles and their aggregates in aqueous solution. It was shown that increasing content of fluorine in the poly(2-oxazoline) copolymers results in a morphological transition from bilayered or multi-layered vesicles to worm-like micelles. The synthesis of poly(2-perfluoroalkyl-2-oxazoline)s is complicated by their extremely low activity in cationic ring-opening polymerization reaction (CROP), both in the initiation and in the propagation due to strong electron-withdrawing effect of perfluoroalkyl substituent. Numerical simulation of the laser light scattering from rough surfaces

A Matlab numerical model for scattering simulation was proposed based on the solution derived from the Beckmann-Kirchhoff scalar theory of scattering of electromagnetic waves from rough surfaces. A series of various synthetic surface samples were obtained using the open source software Gwyddion on which numerical simulations of scattering were carried out. The validity of this numerical model was tested and compared with experimental results.
Preparation and Characterisation of Polyelectrolyte Complexes in the System Dextran-Surfactant

This bachelor thesis deals with the study on preparation and characterization of the polyelectrolyte complex formation in the dextran-surfactant system. The aim of the thesis was the investigation of the influence of the ionic strenght on the formation of the interactions in the studied system. For these purposes, components having opposite charges (DEAE-dextran hydrochloride and anion-active sodium dodecyl sulphate) were selected. For individual measurements mainly the methods of light scattering, dynamic and electrophoretic light scattering in particular, have been used. In the thesis have been studied interactions and aggregation concentrations of individual components having the potential to form polyelectrolyte complexes. It was verified in all the studied samples, by measuring the intensity of scattered light and the size of Z-diameter of particles, that the increase in sizes of particles is caused by the formation of these complexes. There were also measured additional characteristicts of the system such as pH or conductivity. Their dependences provide comprehensive view on the formation of complexes in studied system and can be utilized for other measurements and possible future applications, especially for purposes in medical applications.
Self-assembly of polymers with amphiphilic compounds (surfactants) in aqueous solutions

Title: Self-assembly of polymers with amphiphilic compounds (surfactants) in aqueous solutions Abstract: This PhD Thesis is devoted to the co-assembly in systems containing electrically charged polymers (polyelectrolytes and block copolymers containing polyelectrolyte sequences). I studied the interactions between block copolymers and oppositely charged surfactants in aqueous solutions, and the structure and properties of co-assembled nanoparticles by a combination of several experimental methods. I found that the spontaneous formation, solubility and stability of complex nanoparticles depend not only on the electrostatic attractive forces but also on the hydrophobic effects. In a major part of my Thesis, I studied the interaction of polyelectrolytes with oppositely charged gemini surfactants (containing two charged head-groups interconnected by a short linker and two hydrophobic tails) which is a relatively new topic - much less studied than the co-assembly with conventional single tail surfactants. Better understanding of the formation and properties of complexes containing gemini surfactants and polymers provides knowledge that should lead to novel tailor-made nanoparticles with desired properties for applications in medicine and new technologies (including nano-technologies).
