Neutral genetic variability and structure in chamois populations in Slovakia

The Tatra chamois (Rupicapra rupicapra tatrica), an endemic mountain ungulate, occurs in the High Tatra Mts. (northern Slovakia and southern Poland). In the second part of the 20th century several chamois introductions occurred in Slovakia: Tatra chamois (from the High Tatra Mts.) were introduced into the Low Tatra Mts., while Alpine chamois (R. r. rupicapra) into the Veľká Fatra and Slovenský raj Mts. The High Tatra Mts. population underwent several population declines (bottlenecks) and all other Slovak populations were founded from only a few individuals (founder effect). Moreover, because the Low Tatra, Veľká Fatra and Slovenský raj are neighbouring mountain ranges, there is a potential risk of migration and hybridisation between the subspecies. Using 18 microsatellite loci, we studied neutral genetic variability, structure and potential hybridisation in chamois populations in Slovakia. The study is based on 193 samples: 95 tissue and 5 blood samples, and 88 faecal and 5 hair samples. Low genetic variability was found in all populations, the lowest one in High Tatra Mts. population. Hybrid origin of beavers in Central Europe

The aim of the master diploma work is to investigate origin of Czech populations and genetic variability of almost all Eurasian populations of the European beaver (Castor fiber). For our purposes we collected specimens from Czech populations, reintroduced population in Kirov district in Russia and all European and Asian refugia. At the end of the 19th century the species was on the verge of extinction due to overhunting, surviving only in eight isolated refugial areas. The population size at the end of the 19th century was estimated as 1 200 animals. The number of individuals has increased thanks to the legal protection and reintroductions. The current population size is estimated at 640 000. It is obvious that the populations of beaver have recently undergone severe bottleneck. Beaver was completely extirpated in the Czech Republic in the 18th century. Since the 80th of the 20th century beavers re-established in the Czech Republic due to migration from neighbouring countries and reintroduction programmes, however we knew only little about the origin of beavers in the Czech Republic. Analysis of primary photosynthetic processes in conifers: A comparison of selected methods and their possible utilisation for the study of genetic variability

Conifers are important both ecologically and socioeconomically, however, same parts of their biology are not that well researched. This includes genetics and breeding and partly even physiology. Because quantitative genetic analyzes applied in breeding necessitate an analysis of a large number of samples, and conventional methods of analysis are quite time-consuming, certain parameters describing e.g. the activity of photosynthetic electron-transport chain (ETC) are considered for such use. Several methods of the measurement of the activity of photosynthetic ETC exist, but there are some problems with their usage in conifers. I studied this issue from different points of view in three parts of this thesis. 1) I compared the photosynthetic ETC activity in 8 species of conifers using chlorophyll (Chl) fluorescence measurements on intact needles and polarographic measurements in isolated chloroplasts. Each method brought different information. 2) I measured Chl fluorescence parameters, reflectance spectra and pigment content in 536 genetically defined trees of Pinus sylvestris L. Genetic variability of \kur{Isaria} genus in Czech Republic

My diploma thesis deals with genetic variability of entomopathogenic fungi of the Isaria genus encountered in the Czech Republic. Individual representative of the genus can be found in soil where they attack all developmental stages of insects, giving preference to larvae and pupae. The Isaria fungi find application first and foremost where plants have to be provided biological protection. In case of mitosporic fungi is the precise identification very difficult, taxonomy is often unclear in many genera, including the genus Paecilomyces/Isaria to demonstrate their polyphyletic nature. The fungi are classified primarily with reliance on morphological studies. The most common markers used to identify fungi are the shapes and sizes of their conidia and the biological properties (germination of spores, tests of biological efficiency). Identification made in consideration of the morphological markers is inaccurate and very variable. To overcome those accuracies, there are very useful molecular DNA markers, which can be relevant in ecology, biology and in fungi genetics. This paper relies on applying the ITS region (Internal Transcribed Spacer) as a molecular marker. ITS regions are partial constituent rDNA carrying no code - that is why the regions are likely to accumulate evolutionary changes in the DNA sequence, which makes them suitable for extensive use in taxonomic analyses of many organisms. The study results in a phylogenetic trees constructed by comparing different sequences of ITS regions obtained from the samples of entomopathogenic fungi of the Isaria genus gathered in the Czech Republic during the monitoring stage 2013 to 2014. Genetic variability and phylogeography of Russian wheat aphid, \kur{Diuraphis noxia} (Aphididae)

Genetic analysis of 433 samples of serious crop pest aphid Diuraphis noxia was conducted with the use of 8 microsatellites loci. Statistical analysis revealed sexual reproduction of D. noxia in temperate regions. The linkage disequilibrium was detected because of the excess of heterozygotes. These results support the theory of RNDr. Starý about the invasion of D. noxia to American continent via states of North Africa, Spain and France.
Genetic variation in populations of reed canarygrass, \kur{Phalaris arundinacea} L.

The spread of invasive plant species in natural habitats has become a worldwide problem with negative environmental and economic impacts. An increasing number of invasive organisms are responsible for adverse environmental and economic impacts worldwide, including species extinction, crop failures, reduced water supply, and damage to industrial infrastructures (KERCHER et al., 2007). Phalaris arundinacea L. is widespread throughout the world, except Antarctica and Greenland. Center of diversity of this genus is in the Mediterranean. Members of the genus Phalaris occurs in moist habitats from lower to alpine altitudes (ANDERSON, 1997). Phalaris has a plethora of uses. Its most frequent use is as the root wastewater treatment plants. Phalaris grown as feed for livestock and is also used as an ornamental grass. Phalaris have recently received a lot of attention as a new biomass source for the production of renewable energy in USA. In recent years there has been a massive spread of P. arundinacea across North America (currently occurs in 43 states) and Canada (ZEDLER & KERCHER, 2004). Phalaris represents a significant threat to its original wetland vegetation and is classified as a harmful agens in nine state of U.S. states (LAVERGNE & MOLOFSKY, 2004). It is believed that these aggressive population have European origin.
Morphologic-genetic study of populations of salt-water gastropod \kur{Hexaplex trunculus} from Mediterranean sea

Variability in genes and shell shape of salt water gastropod Hexaplex trunculus (Linné, 1758) was compared among the samples from three Croatian coastline localities, four localities on Greek island Lefkada and two localities from south France coastline in total number of 464 specimens. Genetical criteria were not found for separating individual populations. No variability were found in 683 bp long fragment of gene for michondrial cytochrome c oxidase subunit one (COI). Differences among populations were not significant, even in conection of my data with the data of recent portuguese study (sequences from GenBank). Differences among populations were found by the geometric morphometric method. Based on these results, morphotypes for certain habitats were determined. During the study, high level of tributyltin (TBT) pollution in the Mediterranean sea causing sexual malformation called imposex phenomenon, was proved in this species of gastropod.
Genetic variability of dung beetles on pastures.

The aim of this thesis was a research of the genetic variability of 8 populations of the dung beetle Aphodius prodromus using allozyme markers. Following population-genetic analysis were used to determine genetic structure of these populations: estimates of heterozygosity, Wright´s F- statistics, estimates of gene flow, Mantel test, PCA, Nei´s genetic distance, testing of Hardy-Weinberg equilibrium and linkage disequilibrium. The results revealed low gene flow between populations, low number of heterozygotes and disequilibrium within and between populations. Cluster analyses showed, that populations from Tichá and Rychnov nad Malší-hřbitov are genetically most different from other populations.
Genetic variability of natural populations of the \kur{Diuraphis noxia} (Aphididae)

Genetic variability of natural populations of the species Diuraphis noxia (Aphididae) was studied with RAPD method. First goal was to establish the chosen method in the laboratory and to optimize conditions of RAPD-PCR for D. noxia. This was followed by the pilot study, in which 12 populations were analyzed. Expected variability among individual populations and within population was acknowledged. The results of this investigation came up to the noted progress of diffusion of this species around the World. Results will be used in further more complex study of D. noxia.
