Inhibice regenerace prýtů na explantátech petunie (Petunia x hybrida) in vitro způsobená fluoranthenem

The aim of thesis was to study the effect of fluoranthene (FLT) on growth and biochemical processes of plant Petunia (Petunia hybrida cv. Lady blue) in in vitro conditions. In this experiment were used segments of the middle parts of the leaves, shoots from organogenesis in vitro and apical meristem of stem with four to five leaves from long-term in vitro culture. It was observed if fluoranthene influence organogenesis of shoots in vitro, contents of chlorophyll a and b, content of cytokinins and rating of shoots (from organogenesis in vitro) and apical meristem of stem with four to five leaves. It was used culture MS medium containing benzylaminopurin (1.5 mg/l), 1-naphtylacetic acid (0.18 mg/l) and fluoranthene (1 mg/l) or without fluoranthene (control). In the case of organogenesis in vitro, the experiment was performed at 4 different variations of media with subculturing to other media in triplicate. Variations of the media were chosen to allow period of cultivation with fluoranthene, which affects growth and biochemical processes. MS medium containing indol-3-butyric acid (0.5 mg/l) and FLT in different concentrations (0.1, 1, 3 a 5 mg/l) was used for monitoring rooting. The results showed that fluoranthen is able to affect organogenesis of shoots formed on the segments of leaves. There was lower frequency compared to the control organogenesis. It was found that fluoranthene more affects beginning of cultivation compared to later exposure of fluoranthene during cultivation. It was found the highest uptake of benzyladenine and its riboside from the culture medium. Fluoranthene affected significantly content of dihydrozeatin. Zeatin and its riboside were recorded in a minimal amount. Fluoranthene caused lower content of chlorophylls a and b in explantats. In the case of chlorophyll a were no significant differences between influence by fluoranthene at the beginning of the cultivation or during of cultivation. It was noted that fluoranthene influence the morphology of the roots subcultured shoots from organogenesis in vitro. Plant roots exhibited less branching and roots were shorter in culture medium containing higher content of fluoranthene (3 and 5 mg/l).

Antimicrobial activity of cobalt bis(dicarbollide) aminoderivatives

We carried out experiments focused on the determination of the antimicrobial acitivity of selected representatives of cobalt bis{dicarbollide) aminoderivates. lt was proved, that studied yeast strain C. parapsilosis DBM 2165 is not sensitive to the cobalt bis{dicarbollide) or its aminoderivatives. Gram-negative bacteria P. aeruginosa B-59188 is weakly susceptive to cobalt bis(dicarbollide), but no significant inhibition of growth by cobalt bis(dicarbollide) aminoderivatives was observed. The highest effect on inhibition of microbial growth was confirmed in the case of gram-positive bacteria 5. aureus. This observation demonstrates the real possibility of using these substances in the treatment of infection caused by gram-positive bacterial strains.

Characteristics of the working behavior of Czechs and Ukrainians

The aim of this work is to create a transparent and comparable characteristics of the work behavior phenomenon within the cultures of those states - what is common and what is different between them, or maybe incompatible .It is supposed to have some analysis of possible conflicts and their connection with the mentality. The primary reason of this aim choosing is the specialty I study. My work contains information that could be interesting and useful not only for me and my future job, but also for managers and other employees. Overview of examined issues contains specific points, such as the fundamentals of economic psychology and behavioral economics (symbiosis of psychological and economic opinion on motivation and rationality of individuals), theories of motivation and reaction to different occurrence, characteristic of the term "mentality", general points about the human impact of the world economy. For realizing the goals of my work the following hypotheses have been proposed - Czech and Ukrainian mentality have significant differences, though these cultures are close. - Ukrainian and Czech employees are able to work effectively in a team and find a "common language" in the workplace. The difference in mentality does not obstruct on it. - Employers could be interested in the Ukrainian employees taking into account their high concernment and motivation. All three of the hypotheses were confirmed by observed data and comparison of mentality characteristics. Czechs are often tolerant to other cultures, moreover Ukrainian culture is not much different and does not create difficulties in the working relationship. Based on the fact that the chosen company has no conflicts associated with work behavior and differences in mentality, it's hard to define my contribution to this organization work.

The development of the crime on credits in the South Bohemia before 2014

The aim of this study is to analyze the development of committing a credit fraud offence under § 211 of the Act. no. 40/2009 Coll., the Penal Code (particularly the development of this crime and the development of the amount of the damage) and consequent comparing the selected macroeconomic indicators development in the period 2005-2014 in the region of South Bohemia. In this work secondary data analysis methods were used. Statistical data obtained from the South Bohemian branch of the Czech Statistical Office and statistical data from Crime Police of the Czech Republic was used as the source. The data evaluating the amount of detected cases of this crime committed by the offenders in the Region South Bohemia, according to the region where the fraud was commited, was analyzed. The analyzed file was 5,713 offenses and 4,219 offenders. The defined set was divided into seven parts (districts), according to the local jurisdiction of the territorial departments of the Czech Police. The observed data was compared with selected macroeconomic indicators. From the results it can be concluded that the highest rate of loan fraud offence (calculated per 1,000 inhabitants) is in the district Stra-konice. Conversely, the lowest crime in the same period is in the district Jindřichův Hradec. This work also re-vealed that the development of macroeconomic indicators has no effect on the development of this crime. In conclusion, it is recommended to promote economic education at primary and secondary schools, thus improving the financial literacy of the population. Furthermore, we can recommend the consistent use of, the Central Credit Register, which is already available, as well as supporting of changes in the legislation, so that the providers have an obligation to investigate the credit applicants more and make the offenders of this crime discouraged by higher penalties imposed for credit fraud.

Dynamics of mouse sperm capacitation and acrosome reaction

Capacitation followed by the acrosome reaction (AR), is a very complex event of molecular changes, including acrosome matrix rearrangement and actin polymerization, which mammalian sperm must undergo in the female reproductive tract in order to obtain the ability to penetrate and fertilize the egg. CD46 and β1-integrin belong to specific proteins, which are predicted to interact during molecular reorganization of capacitating sperm. The IZUMO1 as the primary fusion protein of the mammalian sperm is also involved in this dynamic network. We investigated the relationship between the Izumo, CD46 and β1 integrin relocation in the sperm head during the capacitation and AR in vitro. We have already successfully monitored by immunofluorescent labelling the dynamics of proteins CD46 and β1-integrin. The changes in the localization of these proteins associated with the AR and their mutual co-localization was observed. The original β1-integrin location in the freshly released epididymal sperm is in the acrosome and it relocates during the AR further through the sperm head compartments into the equatorial segment and over the whole sperm head. Its density over the equatorial segment is decreasing with the extended time of the AR. Also its presence in the perforatorium of the mouse sperm head is very prominent. The pattern for protein CD46 is extremely similar if not identical in both aspects such as compartment localization and time progress during capacitation and AR in vitro. The molecular interaction of CD46 and β1-integrin was investigated using the Proximity Ligation Assay and Super resolution microscopy STED. The data were statistically analysed. The newly obtained results from CD46 and β1-integrin relocation are in correlation with IZUMO1 dynamics and giving a substantial knowledge on the studied protein network rearrangement during capacitation and AR in mouse spermatozoa.

Fluorescent analysis of the differential protein expression in normozoospermic and asthenozoospermic sperm samples

Asthenozoospermia is one of the main seminal pathologies underlying male infertility. Previous proteomic studies have demonstrated the significant differences in the protein profiles between normozoospermic and asthenozoospermic sperm samples. Since these studies were primarily focused on the identification of differentially expressed proteins by mass spectrometry, we aimed to evaluate the ability of our diagnostic antibodies to detect the differential expression of selected protein markers by fluorescent microscopy and flow cytometry techniques. Therefore, we analyzed sperm samples from 30 men with normal and 30 men with astheno spermiograms, average by the panel of our diagnostic anti-human sperm (Hs) antibodies. Fluorescent microscopy and flow cytometry analysis revealed quantitative differences in the protein abundances between normo and astheno sperm samples, namely, in GAPDHs, evaluated with Hs-8 MoAb, VCP, evaluated with Hs-14 MoAb, and ATP synthase, evaluated with MoAb Hs-36. From the methodological point of view, we observed very high correlation between the data obtained by fluorescent microscopy and flow cytometry techniques and therefore both methods are useful for evaluation of protein differences associated with asthenozoospermia. From the clinical point of view, we observed the strong association of the low sperm motility in the sample with the expression of proteins, playing an important role in sperm energy metabolism (expected), but also with the expression of all tested intra-acrosomal proteins. These findings further demonstrate asthenozoospermia as a complex semen disorder frequently associated with other semen pathologies, which are not diagnosed by basic semen analysis, and the possibility to use monoclonal antibodies as a tool for diagnosis of protein associated sperm pathologies in the semen with the low sperm motility.

: Time Requirement for Basic Nursing Care Procedures

A large number of nursing procedures are carried out in each ward in every hospital every day. In its theoretical part, this Bachelor thesis presents twenty basic procedures. The goal of the thesis was to find out what the most used nursing procedures are and to map the time consumption for individual nursing procedures. The quantitative research method was applied in the Bachelor thesis. Data were collected by measurement of time consumption of ten individual nursing procedures. These ten procedures were selected from a list of most used nursing procedures. Performance of the procedures was observed at the internal and surgical wards of the Hospital in České Budějovice. Both wards were observed over a 3-day span. Ten most often used nursing procedures were selected from the total number of procedures identical for both wards. Correctness of inclusion of the procedures was evaluated by the chi quadrat test in a contingency table. In addition, each of the ten most used procedures was measured ten times at each ward, from the preparation to the clean-up of instruments. The individual time values were averaged and presented along with conclusive deviations. A conformity with or difference in the time consumed was tested statistically by using the F or T test, where the applied significance level equals 5%. The research group was composed of the nurses and ancillary nursing personnel. Results are presented in graphs and tables. The following procedures differed in the individual wards: administration of medicines per os, serving of meals, venous blood sampling and blood pressure measurement. The bachelor thesis is focused on the time consumption for individual nursing procedures. The time consumption and the frequency of the individual procedures can serve as a base of explanation of the number of employees to the nursing management.

Dance as a means of integration adults with intellectual disability

The goal of the bachelor thesis called Dance as a means of integration adults with intellectual disability was to create, implement and evaluate a dance course focused on the development of social competences in adults with intellectual disability. The thesis deals with adults with intellectual disability and with a leisure activity focused on dancing. There is some space for the development of social competences in adults´ leisure time. In response to the current situation of leisure activities proposition for adults with intellectual disability. I found it beneficial to found a dance group of adults with intellectual disability, and at the same time a group of individuals intact in places unfamiliar to them. Based on an activity which may be implemented amusingly it is possible to develop an individual´s personality as well as his or her social competences, which become a presumption of a successful integration, universally. The bachelor thesis consists of four chapters.The first chapter defines the term ´intellectual disability´. Further on the attention is paid to the characteristics of the target group, i.e. adults suffering from a slight and medium mental retardation. Further on, the disability´s impact onto the human psyche as well as social area is described. In the second chapter the term ´dance´is defined and then the description of its usefulness within the leisure, artistic and therapeutic areas is given. In the third chapter the presumptions for a successful integration are described. In the conclusion of the theoretical part there is an interconnection made between the target group and the activity for developing social competences. The theoretical part of the bachelor thesis is continued with a practical part, the fourth chapter. It is a programme of a six-month course which I have set together based on the analysis of expert literature and my experience gained during the leadership of dance lessons and working with adults with intellectual disability. The programme will include work methods and an evaluation of partial activities. The six-month course was held in the school year of 2015/2016 and 6 intellectually disabled adults took part in it. Students of the University of South Bohemia joined the course. Partial goals were set for individual dance lessons and the activities were set so that they would develop social competences in participants non-violently and in the greatest extent possible. The course was implemented in the IN Civic Association facilities. To evaluate the activity and find out the progress within the area of the participants´ social competences, several development fields have been set. Several chosen social competences were divided into evaluation scales. The participants were evaluated at the beginning of the course and then again, six months later. The evaluation method chosen was my own observation. I focused on the evaluation of my own working, on group evaluation and the evaluation of each individual. Evaluation was also done on the part of each course participant, too. Dance lessons and activities connected have proven to be a suitable means for developing social competences in intellectually disabled adults. In five participants the values have risen in 5 10 out of 17 competences observed. A recourse occured in a participant who was going through a difficult life situation during the last two months of the course. Values have risen dramatically and in all participants in the area of group cooperation. Five participants made a progress in establishing and keeping eye contact and in starting a conversation. One participant has made a progress in the area of group performance, another one has made a progress in the area of independence and another one started to call the other participants by their names, as opposed to the beginning of the course. In general, the members of the group became more self-confident.

Oxygen therapy after cardiothoracic surgery from the point of view of nurses and patients

Cardiac surgeries are very serious surgical procedures with considerable specific features. This bachelor thesis focuses on oxygen therapy from the viewpoints of nurses and patients. Its theoretical part describes physiology of breathing and the oxygen therapy as such. It describes individual sources of oxygen and indications for oxygen administration. It also describes principles to be observed during oxygen administration. Specific aids are used for oxygen administration, particularly oxygen masks or oxygen goggles. Quality pulmonary rehabilitation is also necessary to make oxygen therapy more effective. Pulmonary rehabilitation is particularly important for patients after a cardiac surgery and it should be practiced even before the surgery. This approach is critical because in the post-surgery period the therapy makes it possible for the patient to breathe and to expectorate properly. Pulmonary rehabilitation is practiced with patients by general nurses at the cardio surgery ward, as well as by physiotherapists. There are various techniques available to determine efficiency of oxygen therapy and pulmonary rehabilitation. The techniques may be classified as invasive and non-invasive; the most frequently used non-invasive method is pulse oxymetry and the most frequently used invasive technique is blood collection for ABR. Two objectives were set for the thesis. The first objective was to map knowledge of nurses about how to provide care to patients treated with oxygen therapy. The second objective was to determine how the patients feel after a cardiac surgery. Two hypotheses and two research questions were formulated. The first hypotheses were: Nurses working at the resuscitation care station have more knowledge about oxygen therapy than nurses working at the intermediate care station. The second hypothesis was: Nurses working at the resuscitation care station have more knowledge about pulmonary rehabilitation than nurses working at the intermediate care station. The first research question was: How do the patients feel during oxygen therapy and pulmonary rehabilitation? The second research question was: How are the patients after a cardiac surgery informed about oxygen therapy and pulmonary rehabilitation? In the research part of the bachelor thesis I have used both quantitative and qualitative research methods. Quantitative processing of data was used for questionnaires filled out by nurses working in the hospitals Nemocnice České Budějovice a.s., Nemocnice na Homolce in Prague and in the faculty hospital Královské Vinohrady in Prague. The method was used for evaluation and assessment of knowledge of nurses working at the resuscitation and intermediate care wards. Quantitative methods were used for processing of data obtained through semi-structured interviews with patients after a cardiac surgery. The data were transcribed and coded using a pencil and paper method. The data were categorized based on the identified codes. Two categories were created: oxygen therapy and pulmonary rehabilitation. Each of the categories was further divided into three sub-categories. Two categories were created for the research oxygen therapy and pulmonary therapy. Three sub-categories were subsequently created for the oxygen therapy and the pulmonary therapy. The quantitative research has shown that the knowledge of nurses about oxygen therapy is higher among the nurses working at the resuscitation care ward. However, the situation is different in case of their knowledge about pulmonary rehabilitation. In this case the knowledge of nurses working at the intermediate care ward was higher. The processed data from the qualitative research have shown that patients do have sufficient information about post-surgery oxygen therapy, as well as about pulmonary rehabilitation. For all patients in the post-surgery period after a cardiac surgery the frequency of pulmonary rehabilitation was higher than 1 per day.

A nursing issue: peripheral venous catheter

Peripheral venous catheter is used to access the venous system. It is one of the most commonly used invasive procedures in health care facilities. This procedure falls within the competence and daily tasks of nurses. Therefore this type of nursing care is susceptible to risk of proceeding routinely which might result in conscious or unconscious errors. Peripheral venous catheter needs to be introduced into the patient's vein in the following cases: planned parenteral nutrition, infusion or transfusion therapy, application of intravascular medication or contrast media, rehydration, blood sampling (which can be performed immediately after introducing the peripheral venous catheter and before administrating medication) and also during an emergency care for critically ill (such as cardiopulmonary resuscitation). Nurses should be as knowledgeable as possible about this issue so that the peripheral venous catheter can be introduced well and fulfil its purpose. It is important to ensure that nurses obtain all the necessary information about their patients before inserting the catheter and that they are able to properly inform the patients about the procedure and to communicate with them. Next, nurses should be familiar with the correct procedure for inserting catheters, they should be informed about possible contraindications and know how to choose the correct type of catheter as well as where to place it. Furthermore, nurses should care for this vascular access correctly and provide the patient with enough information about taking care of the catheter after the procedure. Finally, nurses should be aware of their own potential errors and also of possible complications. Therefore they should employ aseptic technique so that they can eliminate nosocomial infections. The thesis is divided into a theoretical and an empirical part. The theoretical part focuses on the history of peripheral venous catheters and their types, indications and contraindications, necessary equipment, nursing care, complications, etc. The empirical part consists of a research. The aim of the thesis was to map the use of peripheral venous catheters in hospitals as well as to identify factors that may lead to problems among the patients with peripheral venous catheters. Four research questions were assigned to these aims. 1. What is the procedure prior to inserting a peripheral venous catheter? 2. What are the standards of caring for patients with an inserted peripheral venous catheter? 3. In which steps of caring for peripheral venous catheters do the nurses lack knowledge? What equipment do the nurses prepare in order to insert a peripheral venous catheter? The Bachelor's thesis was conducted using a qualitative research survey. The methods of collecting data included semi-structured interviews and participant observations. The research was carried out only in one hospital ward and it was anonymous. First, the data were collected by means of a semi-structured interview. The nurses were asked 14 questions (see Appendix 8). During this interview, several more subquestions were added. The results of the interviews were then grouped into several categories for the sake of clarity. The second method of collecting data was participant observation. The observed facts were recorded into an observation sheet (see Appendix 9). The same nurses who were interviewed were also observed. This Bachelor's thesis has been written in order to provide a basic overview of the peripheral venous catheters. It could be used as an information source for more experienced as well as novice nurses. The practical output of this Bachelor's thesis is a nursing care standard concerning peripheral venous catheters. This standard was missing in the hospital ward and could help the nurses to take care of these vascular accesses, to proceed correctly and eliminate possible errors (see Appendix10).
