Role of M4 muscarinic receptors in the central nervous system

Muscarinic receptors type 4 are one of the five subtypes of muscarinic receptors, that are members of cholinergic receptors family, together with nicotinic receptors. An acetylcholine binding on the receptor triggers the receptor activation and the signal transmission into the cell by G-proteins located on plasmatic membrane, in the case of muscarinic receptors type 4 by Gi/o proteins. The M4 receptors are, just like other subtypes on muscarinic receptors, located in central and peripheral nervous system. In the central nervous system, they are mostly located in striatum and hippocampus. The M4 receptors have a whole range of regulative functions, the most significant one is most likely the regulation of a locomotion in striatum by the dopaminergic system. The muscarinic receptors type 4 are involved in many behavioural and cognitive processes and are therefore studied as potential drug target for the treatment of mental disorders. Effect of intestinal microflora on the activity of higher brain centers

Abstract Intestinal microflora consists of large bacterial community that resides in the intestines, especially the colon, and lives in symbiosis with the host. It consists mainly representatives of the four bacterial strains (Actinobacteria, Bacteroidetes, Firmicutes and Proteobacteria). It has already been shown that the proportional representation of the bacterial strains in time does not change much, but the research of the last decade have shown irrefutable effect between the change of diet and composition of the intestinal microflora. In addition, there is evidence of changes in the representation of bacterial strains in the course of development of certain intestinal pathologies such as inflammatory bowel disease. More and more are starting to emerge papers describing the influence of altered intestinal microflora respectively its representatives, on the activity and function of the CNS. This interaction of the digestive system and CNS seems to be bidirectional and mediated by several different pathways. Finally, it shows that dysbiosis of the intestinal microflora promotes anxiety and depression that can lead to the urge to prematurely terminate life. Technology for growing Leuzey Saflor (Leuzea Carthamoids DC) and its application

Maralí kořen or parcha saflorová is very easy-growing plant from Asie, specifically from the area of Sibir. It has a specific form resembling thistle. It is very durable plant which can be grown in almost all kinds of climate conditions. Thanks to its origin it has wide range of utilization as well. Its use has been know for many years and it was mainly used in the place of origin. In the Czech Republic parcha is not so common because it came several years ago. However, it has very positive impacts on curing even serious illnesses, central nervous system and libido.
Effect of microbiota on the activity of HPA axis

Recent research shows, that gut microbiome can influence various functions of the organism and is able to communicate with the brain. The data also show that changes in the composition of gut microbiome can influence behavior and stress reactions and vice versa, psychological state of the organism can cause changes in gut microbiome. The aim of this master's thesis was to examine changes of HPA activation and local metabolism of glucocorticoids caused by stress in the presence or absence of gut microbiome. We therefore used germ-free mice and studied the effect of stress in pituitary, adrenal gland, colon and spleen. We found that, stress has different impact on gene expression in adrenal gland, colon and spleen in the presence or absence of gut microbiome. In contrast, there wasn't any significant effect of stress on pituitary in germ free mice and their conventionaly colonized counterparts.
The psychological aspects of degenerative diseases of the central nervous system.

The degenerative diseases affect not only the central nervous system, but also attacks neurological, mainly motoric, functions. These diseases undoubtedly influence mental functions as well. In the theoretical part, the thesis focuses on a possible correlation between the psychical disposition - mainly emotional, and its influence on the development or progression of this kind of disease. Furthermore, the thesis concentrates on the cognitive alternations accompanying given diseases, on the patients' behavioral changes and emotional alternation. The thesis also presents coping strategies and recommendations concerned with the issue of degenerative diseases of CNS based on the recent studies. A research proposal in the empirical part focuses on the effects of social support and coping strategies on the patient with ALS disease and it also explores the patients' motivation and potential future goals which interfere with the quality of life. Incidence and role of the purinergic P2X7 receptor in the immune system

It has been shown recently that extracellular ATP and its metabolite adenosine are key mediators in the immune system. Depending on the concentration, ATP may act as an activator or an inhibitor to the immune response, whereas adenosine is generally well known as a specific immunosupressor. Signals delivered by extracellular ATP and adenosine are detected and transduced by purinergic P2- and P1- receptors, respectively. In principle, all cells of the immune system express P2X, P2Y and adenosine P1 receptors on their plasmatic membrane. Therefore, the purinergic signaling extends to all components of the immune and inflammatory responses, including neurodegenerative diseases. This knowledge triggered a wave of interest in research, development and synthesis of new compounds having effect on P1- and P2- receptors that could help in the treatment of chronic inflammatory diseases and neurodegenerative diseases. This bachelor thesis will summarize the most recent knowledge on the role of the P2X7 receptor in these processes. This subtype of P2-receptor is unique because it is involved in the release of cytokines from immune cells and plays a role in the cell growth and apoptosis.

Changes in neuroglia in degenerative disorders of the central nervous system

Neurodegenerative diseases are a serious disorders of the central nervous system characterized by neuronal loss with a subsequent damage of the brain. This damage may have diverse consequences like a gradual loss of memory and intellect, problems with musculoskeletal system and not least death. Causes of these diseases are not yet fully understood. In addition to neurons neuroglia also plays an important role in these diseases of the central nervous system. In neurodegenerative diseases are astrocytes, oligodendrocytes, microglia and NG2 cells involved in their many aspects. They participate in the protective as well as in the detrimental aspect of these diseases. This work, therefore, presents an overview of previously acquired knowledge of neuroglia in various types of degenerative disorders of the CNS (Alzheimer`s disease, Parkinson`s disease, amyotrophic lateral sclerosis, multiple sclerosis, Wernicke encephalopathy, HIV associated dementia, frontotemporal dementia, vascular dementia). Biological changers of CNS caused by trauma

This thesis focuses on traumatic brain injury and its negative impact on both the brain as a whole and component brain structures. The first two chapters of the theoretical part summarize the information of the anatomy and functions of central nervous system and the most common causes of traumas and its characteristics. The crucial part of the thesis consists of the psychological consequences of traumas arranged according to brain lobe that plays the major role in a clinical manifestation of impaired functions. The thesis also sums up the field of neurodiagnotics and mentions some concrete methods that are being used when dealing with patients after traumas. The last chapter of the theoretical part deals with possibilities and examples of neurorehabilitation as well as factors which are involved in the prognosis of the posttraumatic condition. A part of this chapter also pays attention to the brain plasticity. The last two chapters in this segment refer to other literature from this field. The empirical part presents the research proposal of changes of the brain parts that are involved in facial emotion recognition among TBI patients.
