Controlled production of polyhydroxyalcanoates by bacterial strain Ralstonia eutropha using different substrates

This bachelor’s thesis deals with production of polyhydroxyalkanoates (PHA) by bacterial strain Ralstonia eutropha. Production of PHA on different substrates has been studied to lower the costs of feedstock. In theoretical part the review has been done about the most important types of PHA, production strains and possibilities of cultivation of Ralstonia eutropha using different substrates. In practical part there has been studied production of PHA on different vegetable oils, including waste oils from different sources (restaurants, homes, food companies). Incorporation of different precursors for copolymer production control was studied too. The highest yields of poly-3-hydroxybutyrate (P3HB) have been achieved using waste oils. For economical and ecological reasons these oils are very suitable substrates. Using concentration 20 g/l of waste rapeseed oil from university canteen there has been produced 13,32 g/l of biomass containing 58,63% of P3HB in 84th hour of cultivation. The highest yield of PHA in precursors study has been achieved applying 1% propanol in 24th hour of cultivation. The PHA increase has been 97%. This PHA was composed from 91% of 3HB and 9% of 3-hydroxyvalerate.

The main subject of this bachelor’s thesis is describing the present state of knowledge in the sphere of compression-ignition and spark-ignition engines lubrication, lubricants and future trends. First part is about internal combustion principle, engine construction. Next parts are about engine lubrication system, lubricating modes, and its locating in the engine. Last part is about oils, and insights into the future.
L-ascorbyl-6-palmitate and its utilization in dietary and cosmetic supplements.

Bachelor‘s thesis deals with the effection of antioxidants in foods and cosmetics supplements. Some antioxidants as vitamin A and beta-carotene, vitamin E and tocopherols and vitamin C as L-ascorbic acid and her derivate L-ascorbate-6-palmitate are described here. This derivate can by used as an accessory material in cosmetics and foods preparations, especially in the preparations that contain oil or fats, because as a lipophilic compound keeps them from getting rancid.

Ve své práci, jak již název napovídá, se zabývám využitím řepkového oleje jako alternativního paliva na místo nafty. Díky vyčerpatelnému zdroji: ropy, ze které se nafta vyrábí, je toto téma v dnešní době velice diskutované, prozatím jsou s ním malé zkušenosti, avšak do budoucna je jednou z mála variant náhrady fosilních paliv. Z tohoto důvodu aplikuji šetření do praxe, provádím výzkum ve skutečně existující společnosti, která denně provozuje několik mechanizačních strojů poháněných litry a litry nafty a zkoumám ekonomický dopad na náklady při nahrazení nafty alternativní pohonnou hmotou - řepkovým olejem. Pro větší zajímavost volím výrobu vlastního filtrovaného oleje z nakoupeného řepkového semene, na novém lisovacím zařízení, to vše mimo jiné za pomoci dohledu zaměstnanců na chod strojů.
