Hodnocení odrůdových sortimentů aksamitníků (Tagetes L.)

This diploma thesis deals with description and evaluation of Tagetes L. varieties, according to predetermined descriptors. It also deals with its historical, morphological, cultivating and introductory historical aspects. A total of 28 varieties of taxaTagetes patula L., 7 varieties of taxa Tagetes erecta L. and 6 varieties of taxa Tagetes tenuifolia Cav., were evaluated. Their seeds were obtained from the gene resources of VÚRV Prague - Ruzyně (Gene Bank), from breeding companies Volmary (Germany), SEMO a.s. (Czech Republic) and Petunia Černý (Czech Republic). The observation results are summarized in the commented tables, which are recording the monitored characters of the varieties in the descriptor point scale as well as in the specific found values. The comparison of the individual varieties can be the basis for the recommendations of the best varieties for both breeders or gardeners as well as for use in public greenery.
Apple dormant bud cryopreservation: Methodology

In the Czech Republic, ex situ and in vitro conservation is used for apple tree germplasm conservation. In relation to the strategy of maintenance of broad genetic potential of apple tree, there is a necessity to preserve genotypes by storage at ultralow temperatures – cryoconservation. Based on broad spectrum of apple cultivars, an optimised dormant bud cryopreservation protocol was developed. The developed cryoprotocol enables to establish a safe backup storage system for ex situ and in vitro collections of apple tree germplasm. The Ministry of Agriculture of the Czech Republic is the user of this methodology and it will utilize it in the framework of “National Programme on Conservation and Utilization of Plant, Animal and Microbial Genetic Resources for Food and Agriculture”.

The main purpose of the bachelor thesis was to collect information about breeding of Czech Hen and the importance of the genetic resources. The history of the breed was described. Another part of the bachelor thesis was the description of the colour strains of the Czech Hen, other similar breeds, the current state, the utility and the principles of breeding. Possible perspectives in breeding of the Czech Hen, its use in creation of laying type of hybrids and possibility of use in organic breeding are suggested at the end of the thesis.
Old Czech breeds of hens, their conservation and utilization in organic farming.

This thesis deals mainly with the only two typical Czech breeds of hens - Šumavanka and Bohemian Fowl. It gathers information about their history and assesses their characteristics, such as stand-alone behavior, demands of breeding, laying of eggs, food searching, resistance in rough climatic conditions. For these breeds, suitability for organic farming is considered. Furthermore, this study focuses on the significance of ecological farming and genetic resources, and their importance for conservation of natural resources for future generations. It also includes examples and practical experiences and results of breeding of Czech hens.

Resume The bachelor work deals with the difficulty of gene sources of decorative wood species in Czech Republic and abroad. We tried to gain information about the current state of decorative wood species in Czech Republic and concerning research institutions that are involved in the process of gene pool preservation, especially using the forms ex situ, in situ, on farm, in vitro. The Crop Research Institute in Prague, Ruzyně is the main research institution for plant production and is also covering all other research centres, organizations and institutions in Czech Republic. We focused on National Program for genetic sources of agricultural plants and its coordination. It was vital to introduce information connected to genetic sources of decorative wood species. The main aim of our bachelor work was oriented to history and present of genetic sources of decorative wood species and summing-up of complex information about the collection state in Czech Republic an abroad.
The use of microsatellite analysis for the characterisation of onion

The object of the methodology is the process characterizing the autenticity of varieties of onion put through microsatellite analysis (SSR- Single Sequence Repeats) for the characterization of varieties of garlic into the practice. This method allows to identify varieties of onion and characterize genetic resources using DNA markers and determine their genetic similarity based on the length variability of short repeating sequences (microsatellites) in the characterization of varieties of onion, Allium cepa L. The principle of the method is amplification of the genome containing the microsatelite locus by polymerase chain reaction (PCR) using specific primers and subsequent analysis of the lengtht of the products and the identification of the DNA profile to the declared variety.

This thesis was tasked in the location to choose and describe apricot trees older than 40 years. Assess the health of trees, observe phenophases flowering and ripening fruits inclucing pomological description. For most promising trees in terms of age and vitality remove grafts for further propagation and conservation. Part of it was on to determine their coordinates GPS and introduce the map. The literary section was described species Prunus armeniaca L., were characterized traditional varieties. It also describes the history of cultivation of apricots in this area, apricot breeding and conservation of genetic resources. Practical work describe climatic conditions of the place, includes a description of 61 trees, a description of the phenological stages of flowering and ripening, description pomological and growing characteristics and evaluation of vitality trees. Based on the characteristics and outcomes were selected trees for sampling grafts, and a further propagation and were planted 106 apricot trees from 28 clones for preservation conservation on farm. All trees were localized GPS coordinates and place on a map.
The Meat Utilization of Rabbit Breed Czech Solver

This master's thesis focuses on the utilization of the Czech Solver, a Czech national rabbit breed, which is registered in The National Programme on Conservation of genetic resources of plants, animals and micro-organisms for Food and Agriculture. The Czech Solver was originally invented to be able to confirm or disprove the purebred origin of the tested animal. This feature is useful for breeders so that compensates for low meat performance breed. Fertility monitored breeding is less than the average fertility medium breeds, among that is the Czech Solver sorted, and significantly lower than the fertility of broiler rabbits. Surveyed individuals have achieved monthly weight gain sampler breeds of rabbits, but fattening period to 2.70 kg live weight ranged from an average of 105 days for semi-intensive rearing system in cages to 135 days at a traditional pens housing system farming, which is at least 27 days longer than the period of fattening broiler rabbits. Average growth rates were observed in the amount of 19.7 g in traditional breeding and 20.6 g in semi-intensive farming. Because the rabbits were slaughtered when reaching 2.70 kg live weight, were at abattoir analysis revealed no statistically significant differences between males and females or between traditional and semi-intensive farming. Slaughter analysis confirmed the breed Czech Solver average percentages of the intermediate part, fore and hind parts of the carcass and a higher proportion of kidney fat compared to other breeds of rabbits included in the genetic resources and compared with broiler breeds.
