This bachelor thesis describes common information about cyanobacteria and cyanotoxins, environmental drivers of cyanobacterial blooms and summaries present knowledges of the occurrence of their secondary metabolite neurotoxin \recke{beta}-N-methylamin-L-alanin. The thesis includes a project proposal regarding monitoring of neurotoxin \recke{beta}-N-methylamin-L-alanin in fishponds and water reservoirs.
The role of caspase-3 in apoptosis

Caspases provide anti-inflammatory, apoptotic and developmental processes in organisms. They are enzymes with a wide range of activities in all cells, and various pathogeneses can occur if their proper function is disturbed. Since the 1990s, caspases have been a topic of interest for scientists, as their direct link to the triggering of apoptotic processes is a promising possibility for the therapy of diseases related to apoptosis, such as cancer, neurodegenerative diseases, but also cardiac ischemia and diabetes. The cascade of apoptotic processes is controlled by the aforementioned caspases, which are located in the caspase cascade. When the cascade is triggered in a cell, it is due to the presence of a "danger" signal, which can be very different. The most well-known triggers of the apoptotic cascade include activated Fas receptor and FasL ligand, cytochrome c present in the cytoplasm, an imbalance of IAPs in the cell, damaged DNA, and many others. Upon receipt of a signal, initiator caspase-2, caspase-8, and caspase-9 are activated, which in turn activate effector caspases-3, caspase-6, and caspase- 7, cleaving many substrates to promote apoptosis. Natural compounds potentially influencing neurodegenerative diseases

Diploma thesis, Charles University in Prag, Faculty of Pharmacy in Hradec Králové, Department os Pharmacognosy and Pharmaceutical botany, Hradec Králové 2022, 88 s. This research diploma thesis draws mainly from foreign professional literature. It provides an overview of important natural substances that potentially influence neurodegenerative diseases. The first part describes the moct common neurodegenerative diseases, their characteristics, epidemiology, pathogenesis and clinical picture. These diseases include Alzheimer's disease, Parkinson's disease, Lewy body dementia, multisystem atrophy, Pick's disease and other tauopathies, Huntington's disease, spinocerebellar ataxia and amyotrophic latelar sclerosis. The second part of diploma thesis presents natural substances that have been studied in preclinical and clinical studies in the last two decades for their potential to positively affect neurodegenerative diseases. The last part discusses the contribution of these natural substances fot therapy and prevention of neurodegenerative diseases. Case Study of Physiotherapy Treatment of a Patient with Diagnosis morbus Parkinson

Title: A case report of physiotherapeutic care for a patient diagnosed with Parkinson's disease. Objective: The aim of this bachelor's thesis in the theoretical part is to provide information on the diagnosis of Parkinson's disease, especially information on etiopathogenesis, diagnosis and treatment. The aim of the practical part is to process a case report of a patient suffering from this diagnosis and evaluate the effect of the performed physiotherapeutic intervention. Method: The general part includes theoretical information about Parkinson's disease in terms of pathophysiology, etiopathogenesis, clinical picture and other areas necessary for a comprehensive understanding of the diagnosis. There is also information on the treatment options for Parkinson's disease, including rehabilitation. The special part contains the case report of a patient diagnosed with Parkinson's disease, which arose within the bachelor's internships at Vršovická Zdravotní. Results: The goals of the theoretical and practical part were met. Following the evaluation of the effect of therapy, it can be concluded that the physiotherapeutic intervention described in the special section had a partial positive effect on the patient's condition. Identification of sleep disorders based on actigraphy data and sleep diaries

This master’s thesis deals with prediction of Parkinson's disease using sleep parameters from actigraphy and sleep diaries. The goal is to design a machine learning approach, which will be able to recognize pacients suffering from Parkinson's disease. For training dataset supplied by St. Anne's University Hospital Brno was used, which was variously modified for achieving best possible results. These adjustments were evaluated according to the results of the trained models and based on these results, two models (achieving test accuracies of 85 and 82%) were selected.

Brain metabolic substrates must be strictly regulated both spatially and temporarily to ensure the energy homeostasis of the brain. Prions in yeast

The thesis describes yeast prions and their biological effects on yeast in general. It defines the basic characteristics of yeast prions, that distinguish prions from other proteins. The thesis introduces various possibilities of prion formation, and propagation as well as specific types of yeast prions, including various functions of most studied types of prions. The thesis also focuses on chaperones that affect the state of yeast prions in cells. Lastly, the thesis indicates similarities between yeast prions and mammalian prions that are related to neurodegenerative diseases. The use of induced pluripotent stem cells in the treatment of spinal cord injury and ALS.

Induced pluripotent stem cells (iPSCs) have become a new phenomenon of regenerative medicine. It is obvious that they share some common characteristics with embryonic stem cells (ESCs) such as stemness potential, self-renewal p., differentiation p. iPSCs retain their epigenetic memory, allowing becoming patient-specific and so it is not necessary to apply immunosuppressants. The use of ESCs is controversial, because their acquisition is associated with embryo destruction. As a cell source for iPSCs derivation we can use any somatic cells, however, fibroblasts are preferably used due to their easy availability. With transcriptional reprogramming cocktail (OCT4, SOX2, KLF4, c- MYC / OCT4, SOX2, NANOG, LIN28) we can obtain required iPSCs line, which is then further differentiated into neural precursors (NPCs). These cells can be grafted into lesion site, where they can facilitate regeneration by several mechanisms (cell replacement, protective effect, facilitation the expression of trophic factors). Nevertheless, here we are still dealing with the risk of tumorogenesis or low cell derivation efficiency that limits the use of iPSCs in clinical practice. The m6A pathway at the molecular level and its role in neurological diseases

N6-methyladenosine is the most abundant modification in eukaryotic messenger RNA. This modification is reversible, thanks to a complex of methyltransferases and demethylases. The biological effects of m6 A are mediated through reader proteins. This complex mechanism of proteins contributes to many molecular processes such splicing, translation and transport. It also plays a role in many serious neurological diseases, such as Alzeheimer's disease, Parkinson's disease, major depressive disorder and attention deficit hyperactivity disorder. The purpose of this thesis is to describe the m6 A pathway, its regulation at the molecular level and to put it into context with neurological diseases of today.
