Produkce IL-1? a IFN? po stimulaci mléčné žlázy lipopolysacharidem

This thesis was focused on the pro-inflammatory cytokines IL-1beta and IFN-gamma. Absolute and differential leukocyte count was also monitored. The experiment was conducted at 8 clinically healthy heifers, hybrids of Holstein and Czech Pied that have been housed by tethering in stalls and fed with a standard diet. The inflammatory reaction was induced by lipopolysaccharide (LPS; 5 ug in 20 ml PBS), as a control a phosphate buff-ered saline (PBS) was used. Results were measured at 1, 2, 3 and 7 days after stimulation of mammary gland by above-mentioned factors. Concentration of each cytokine was detected by a sandwich ELISA using commercially available kits. At 1 day after stimulation of mammary gland by LPS and PBS an average number of leukocytes, which was statistically significantly higher in the case of stimulation by LPS (P <0.01), was detected. After 7 days there was a significant decrease in the total number of leukocytes. There has also been a shift in the differential leukocyte count. Most abundant cell type were neutrophils, whose number was higher in the case of stimulation by LPS. Between day 1 and day 7 after challenge, there was a gradual reduction in the proportion of neutrophils. In the same period an increase in the proportion of macrophages and lymphocytes was detected. Concentration of IL-1beta also increased, 1 day after the activation a striking increase has been detected. In following days there was gradual decline of IL-1beta concentration almost to the level prior to treatment of the mammary gland. In the case of IFN-gamma similar pattern in the form of strong growth and a subsequent gradual decline in concentration to the original values was detected. There was found positive correlation between the increase in IL-1beta and IFN-gamma concentration and a shift in the differential leukocyte count in favor of neutrophils, which confirmed the important role of these pro-inflammatory cytokines in the establishment of inflammatory response and the mobilization of the components of natural and specific immunity.
Electrical Installation Project for Kindergarten

The work has three main parts. The first part of the thesis introduces the problems of power and data installations to the reader. This includes related legislations, LPS, the methods of feeding the building by electrical power and the electrical wiring in the building, including their execution and protection. In the second part of the thesis are defined the principles of creation of electro design documentation. Mainly the requirements for their content and editing. An important point is the process of creating a design. The last part is practical. It is the elaboration of a complete wiring design of the kindergarten building, which consists of drawings and technical reports.
Electrical installation of family house - project documentation

This bachelor thesis is called Family house electro installation – project documentation. The main aim of this thesis is to introduce readers into the design of the installation family house. In the first part was explained themes of engineering, legislative rules and external influences. The next topics were instalation and description of electric way from distribution site to a plug. The next aim was to get familiar implementation of house distribution system. End of thesis discussed about project documentation – technical report and drawings.

The mammary gland is the main production body in dairy cattle breeding. These days are current problems of mastitis disease of the mammary gland, which have an appreciable economic impact. Bachelor thesis briefly describes the anatomical structure of virgin mammary gland of non-pregnant heifers and discusses the use of model virgin mammary gland of heifers to study the immunological properties of the cell. Describes its first use, because the methodology used is the basis for subsequent work performed on virgin mammary gland. The work also aims to summarize the use of the model in research practice. Research reviewed the ultrastructure and functional characteristics of the cells before and during induced inflammation, its intensity and time of resolution by used agents. Furthermore, evaluating the pros and cons of using virgin mammary gland, compared to the adult mammary gland during involution, or lactation.
The effect of tick cystatins on TLR - induced maturation of myeloid dendritic cells

Tick saliva contains a lot of molecules with antihemostatic and immunosupressive effects.The goal of this thesis is to test the effects of tick salivary cystatins from I.ricinus and I.scapularis on TLR - induced maturation of bone-marrow derived dendritic cells and production of chosen cytokines. Over all, the supressive effect of tick cystatins was observed in relation to TLR-induced maturation of DC. In addition, cystatins enhanced production of IL-10 and attenuated induction of IL-12 cytokines.
Optimization of melanoma immunotherapy based on the anchoring of laminarin on the surface of tumor cells

The aim of this thesis was to find optimal anticancer regimen of therapy, based on a naturally occuring polysaccharide laminarin. It was studied on the Mus musculus skin melanoma B16-F10. There were explored an ajuvant effect of LTA. Mixture of laminarin with LPS was applied with various anchorings and was tryed out the possible replacement of LPS. Using the flow cytometry there were determined progress of immune cells during application. This therapy resulted in reduction of tumor volume and the prolongation of overall survival.
Stratefis in mathematical education

The aim of this thesis was an analyse of individual didactic situations and their valuation based on selected strategies. There was used video material from 20 lessons on linear equation collected from one basic and one grammar school. The conclusion of the analysis is supported by theories from available sources. This thesis might be useful for students of pedagogical faculties to deepen their knowledge and to help them avoid possible mistakes. It might be used also as a self-study material for teachers.
