Quality of sports services in selected public daycare centers

Title: Quality of sports services in selected public daycare centers Objectives: The main objective of this bachelor thesis is to determine the quality of sports services in selected public daycare centers in the territory of Mladá Boleslav and based on the results to propose recommendations that will reflect the shortcomings in the services provided. Methods: The principles of the SERVQUAL and SERVIMPERF methods were used to investigate service quality. The data were collected using an electronic questionnaire addressed to the parents of the children. The questionnaire was sent to the directors of two selected daycare centers with a total capacity of 274 children and subsequently published on their websites. Results: The survey shows that in none of the selected public daycare centers did the average value of the perceived quality of services exceed the value of importance. For daycare centre A the average value of importance is 6.38 and the value of perceived quality is 6.07, in the case daycare centre B it is 6.06 for importance and 5.84 for perceived quality. None of the average values of the examined dimensions was lower than 5. The thermodynamics of DNA-hairpin formation

Serum response factor (SRF) is a transcription factor which binds to a highly conserved DNA sequence called the CArG box. According to the nucleotide sequence of CArG box it could form a hairpin structure or a cruciform. In this master thesis, the structure of the CArG box in a human gene c-fos was studied by nuclear magnetic resonance. 1 H spectra at temperatures 274{356 K, two-dimensional 1 H{1 H NOESY spectra, and two-dimensional 1 H{13 C HMBC spectra for DNA sequences with lengths of 12, 14 and 16 nucleoti- des were acquired. The thermodynamic parameters of formation of the secondary structure in the samples were determined from the measured temperature depen- dencies. The hairpin formation in the samples was con rmed based on the NOESY spectra and the lack of dependency of the melting temperature on concentration. The observed difference of the secondary structure from B-DNA could serve as a possible explanation of the high a nity of SRF to CArG box. 1
Structural properties and stability of DNA hairpins characterized by NMR spectroscopy

CArG box is a highly conservative DNA motif found in Serum Response Element (SRE), which regulates expression of c-fos gene. In this thesis, short oligodeoxynucleotides containing CArG box were measured using nuclear magnetic resonance spectroscopy to evaluate their ability to preferentially form hairpin over duplex. 1H spectra were measured at temperature range (274-360) K. We acquired thermodynamic parameters of the transition between hairpin and single strand by fitting the temperature- dependent chemical shifts. The hairpin structure of our oligonucleotide samples was confirmed by non- B-DNA patterns in NOESY spectra, absence of concentration dependence of melting, and other pieces of evidence. Thus, occurrence of unusual DNA conformation of CArG box in native DNA, potentially even cruciform is highly possible. This could explain the high affinity between SRE and its transcription factor. Analysis of Efficiency of Perforated Steel Strips in Timber-Concrete Structures

Perforated steel strips as an alternative coupling method of steel-concrete structures have found application mainly in bridge construction, but they are also increasingly used in building structures, where it is necessary to overcome large spans. Due to the absence of complex knowledge, the usage of perforated steel strips in timber-concrete structures does have not reached the level of other types of coupling methods. Numerical simulations can be used to analyse the behaviour of such structures, as well as monitor the stress parameters of individual elements. Based on the results of these simulations, it is possible to evaluate the potential of such structures, as well as other directions of research of timber-concrete structures with perforated steel strips.
Identification Of Sleep/Wake Stages In Actigraphy Data Utilising Gradient Boosting Algorithm

Sleep disorders are early markers of various serious diseases that can be treated moreeffectively when diagnosed in their prodromal stage. Actigraphy is a noninvasive sleep monitoringmethod for the detection of sleep patterns and determination of sleep parameters that could support thediagnosis of these disorders. This study aims to compare a newly proposed actigraphy-based methodof sleep/wake detection with a conventional one in terms of consistency with a polysomnography(PSG) reference. 55 recordings (acquired in 28 subjects) of actigraphy and PSG were modelled by aheuristics-based method and by a new approach utilising a gradient boosting algorithm. In addition,another database (22 subjects, 150 recordings) was used to compare scores of the new method withdata reported in sleep diaries. The proposed method achieves 89% accuracy and Mathews correlationcoefficient equal to 0.75 when compared to the polysomnography reference. Such results outperformthe ones provided by the heuristic technique. The newly proposed method has good consistency withthe PSG reference, thus being a good alternative to the golden standard in sleep disorders assessment,especially in decentralised clinical trials.
Simulation Of Electroporation Process In Stent Oclussion Therapy

The main focus of this work is to compare the simulations of electroporation in the biliarytract with and without the stent. The aim of this paper is to determine the ablated areas using differentcombinations of electrodes and different amplitudes of applied voltage. The resuluts of thesimulations shows distribution of intnesity of electric field and joule heating. Presented simulationsare done using finite element method (FEM) software COMSOL MULTYPHISIC 5.4.
Design And Optimization Of Special Low-Level Amplifier For Measurement Of Air Ions

This paper deals with low-current measurement in order of picoamps up to a few dozens of femtoamps. Such low currents measuring is necessary for determination of air ions concentration. Disturbances, which are otherwise negligible for ordinary measurements must be considered. For example, leakage currents and noise generated in measuring device circuit. The choice of a precise operation amplifier with low input bias current is as important as the selection of low noise passive components.

The issue of legal regulation of drug related crimes in Czech criminal code Abstract The thesis deals with the alcohol and drug-related substantive crimes and aims to provide the reader with the detailed analysis of the basic substituent elements of a crime with the analysis of the basic terms of the problematic and application of the existing case law. The analysis of some vague terms which are the subject of unending discussion of both the legal community and the general public is also provided in the thesis. I processed the thesis with the focus on the substantive law with emphasis on the drug- related crimes regulated in legislative provisions § 284 to § 287 of the Criminal Code and further on the chosen crimes from the area of secondary drug-related criminal activity, especially on the crime of endangerment under the influence of drugs or alcohol according to provision § 274 of the Criminal Code and the crime of drunkenness according to provision § 360 of the Criminal Code. Zhodnocení zdravotního stavu dřevin z hlediska výskytu dřevních hub v parku Anthropos (Brno-střed)

Bachelor thesis dealing with assessment of health status of trees with the occurence of wood-decay fungus in the Anthropos park in the Brno-střed district. The health survey was made between December 2015 and May 2017. Among the 274 trees were wound 8 wood-decay fungus, mostly with white rot. The most frequently attacked species are Aesculus hippocastanum and Fraxinus excelsior. From a health status point of view the most important wood-decay fungus are Fomes fomentarius and Kretzschmaria deusta. On the basis of a health survey was proposed technical measures and treatments according to the arboristics standards of Nature Protection Agency of the Czech republic.
Beech and spruce forest stands conditions in the area of the Moravian-Silesian Beskids and soil zoocenosis

Soil epigeal fauna of beech and spruce forest stands had been monitored for 8 years (2007-2014) in 37 selected research area in the Moravian-Silesian Beskids. Collection of material carried out twice per year in spring and autumn aspects. Site characteristics of research localities were characterized by high variability while 4 vegetation altitudinal zones, 8 edaphic categories and 12 forest site types had been distinguished. Soil fauna was extracted by Tullgren devices. Within the monitoring, 11 target animal groups were engaged in: mites, springtails, larvae stages of ground beetles, symphylans, diplurans, proturans, centipedes and millipedes, larvae stages of rove beetles, adults of rove beetles, earthworms, larvae stages of click beetles (wireworms). Earthworm´s and wireworm´s cenosis were deterimined into species and genera, respectively. Remaing groups of soil fauna had been studied within larger systematic categories. Subsequently, abundance and dominance of the particular groups of soil epigeal fauna. The outputs had been compared each other and potential relations between particular animal groups to the specific site properties, vegetation altitudinal zones, edaphic categories and forest site types had been assessed thereby. Overall, 274 015 individuals were captured within the monitored soil animal groups. Several groups of soil cenosis were disvored specific relations to the site conditions. Additionally, 12 species of earthwomrs were distinguished while Dendrobaena octaedra seemed to be significantly dominant species representing 68,33 % of total earthworm´s species spectrum. Larvae stages of wireworms were determined into 5 genera. Representatives of Athous genera formed almost 86 % of the population of wireworms.
