Biocompatible protein cages for encapsulation and internatization of small interfering RNA

This thesis is focused on creation of apoferritin nanocarrier with encapsulated small interfering RNA marked with fluorescent dye. Main objectives are optimization of pH and amount of siRNA encapsulated into apoferritin cavity and physicochemical characteristics of created nanocarrier. First part deals with theoretical knowledge necessary for understanding concept of this thesis. Second part describes used methods and evaluated results. Created apoferritin nanocarriers were optimal in size with great hemocompatibility, but long-term stability didn’t meet our expectations.

In silico design and validation of conotoxin-based peptides for neuroblastoma nanotherapy

This thesis is focused on in silico design and validation of peptides based on -MrIA conotoxin from Conus marmoreus marine snail species, and their possible use in the treatment of neuroblastoma through norepinephrine transporter targeting. Five conotoxin-based peptides were designed using molecular dynamics simulations, and their trajectories were analyzed to determine peptide properties. Two homology models of human norepinephrine transporter were prepared for further analysis of binding properties of the conotoxin-based peptides to norepinephrine transporter. Peptides were synthesized and used to coat apoferritin nanoparticles with cytotoxic agent ellipticine trapped inside apoferritin’s cavity. Created peptides and nanoparticles were analyzed to determine their physico-chemical characteristics. Furthermore, cellular uptake and cytotoxicity towards neuroblastoma and epithelial cell lines were studied. From both in silico and in vitro studies, it appears that YKL-6 peptide is the best candidate for further research.

Popis podstatných rozdílů technické přípravy ve vodním slalomu a rychlostní kanoistice v kategorii K1

Název práce: Popis podstatných rozdílů technické přípravy ve vodním slalomu a rychlostní kanoistice v kategorii K1 Cíle práce: Cílem diplomové práce je zjištění hlavních charakteristik typických pro technickou přípravu kajakářů v českých kanoistických klubech a popis případných rozdílů v přípravě rychlostních kajakářů a kajakářů ve vodním slalomu. Metody zpracování dat: Práce je založena na smíšeném výzkumu. Závodníkům a trenérům byl předložen dotazník týkající se technické přípravy. Zjištěné výsledky byly analyzovány pomocí metod kvantitativního výzkumu a analytické indukce. Výsledky: Mezi technickou přípravou rychlostních kajakářů a vodních slalomářů bylo zjištěno několik podstatných rozdílů. Závodníci ve vodním slalomu především mladších věkových kategorií častěji než rychlostní kajakáři využívají pro rozvoj základních kajakářských záběrů jiné vodní sporty, nejčastěji sjezd na divoké vodě. Z průpravných cvičení se rychlostní kajakáři omezují pouze na fázování záběru. Závodníci vodního slalomu používají pro rozvoj základních kajakářských záběrů větší množství průpravných cvičení. Pro získání zpětné vazby využívají vodní slalomáři nejen v technické přípravě několikrát do měsíce analýzu pomocí videozáznamu.
