The thesis describes successful transient transfections of suspension-adapted HEK 293 cells with linear polyethyleneimines (PEIs) in serum-free medium. The cells were transfected with pcDNA5/SEAP plasmid expressing the secreted form of human placental alkaline phosphatase (SEAP). SEAP concentrations in cell culture supernatants were determined in order to compare efficiencies of individual transfections. The goal of the thesis was to optimize various factors affecting transfection efficiencies, primarily the DNA:PEI ratio.
