Air conditioning area of the car showroom
Svobodová, Leona ; Blasinski, Petr (referee) ; Rubina, Aleš (advisor)
The diploma thesis deals with shielding elements in the theoretical, project and experimental level. The theoretical part describes the variants of building shading and compares individual shading elements. It explains several concepts from the field of environmental technology, which are related to the topic. The design part deals with the air conditioning of the showroom space with a specific indoor environment. The space has a flush facade, so it has a high heat load. This is a showroom for veterans, where it is necessary to maintain optimal constant hu-midity throughout the year. An air conditioning unit was designed for these needs. The experimental part describes the measurement of the values of the shielding factor of external blinds for individual slopes of slats. The measurement results are applied to the reference room and the dependence between the inclination of the slats and the energy demand for space cooling is evaluated by the software.
Air conditioning in multifunctional center
Svobodová, Leona ; Blasinski, Petr (referee) ; Rubina, Aleš (advisor)
The object of this bachelor thesis is the design of air-conditioning equipment in a multifunctional centre. The devices serve a multifunctional hall and a room with over-head, the lighting and the sound. The units are designed in a way to meet all the hygienical, operational and functional requirements and to create optimal in-door microclimatic conditions for people to stay. There were used all the available budget calculations, applicable regulations and standards for this design. The air-conditioning provides a supply of fresh air and conducting of depreciated air, cov-erage of heat losses, removal of heat load and humidification of air. The theoreti-cal part deals with the system of return gaining of heat and describes advantages and disadvantages of every device enabling.
Studying the regulation of expression of genes involved in barley malting quality
Hoffmannová, Viktorie ; Svobodová, Leona (advisor) ; Lipavská, Helena (referee)
Barley (Hordeum vulgare L.) is a grain cultivated as an agricultural crop across many varying climatic areas of the world. It is mainly used for its feed and brewing industry properties. This work will focus on studying the malting process on a molecular level, which mainly includes the genes for degradative enzymes of storage proteins, starch and compounds of endosperm cell walls. We shall observe how all these components interact during malting and other processes of brewing beer. It is known that malting quality parameters are tied to several genes, whose expression is regulated. These regulatory pathways will also be included in this work.

Detection of five garlic viruses using real- time PCR analysis: methodology for practice
Mitrová, Katarína ; Svobodová, Leona ; Ovesná, Jaroslava
The objective of the assay is detection of five viruses in garlic using real- time PCR analysis. . This assay allows to identify five of the viruses in the varieties of garlic - Onion yellow dwarf virus (OYDV), Leek yellow stripe virus (LYSV), Garlic common latent virus (GCLV), Shallot latent virus (SLV) and Garlic mite-borne filamentous virus (GMbFV) based on the detection of gene sequences for the coat protein by SYBR Green real-time PCR. This method was shown in our work to be a suitable tool for the detection of highly variable pathogens, such as garlic viruses. The method brings a new procedure for detection of these viruses.

The use of microsatellite analysis for the characterisation of garlic
Mitrová, Katarína ; Ovesná, Jaroslava ; Svobodová, Leona
The object of the methodology is the process characterizing the autenticity of varieties of garlic put through microsatellite analysis (SSR- Single Sequence Repeats) for the characterization of varieties of garlic into the practice. This method allows to identify varieties of garlic and characterize genetic resources using DNA markers and determine their genetic similarity based on the length variability of short repeating sequences (microsatellites) in the characterization of varieties of garlic, Allium sativum L. The principle of the method is amplification of the genome containing the microsatelite locus by polymerase chain reaction (PCR) using specific primers and subsequent analysis of the lengtht of the products and the identification of the DNA profile to the declared variety.
