Function of Pragmatic Markers in Political Interviews Based on Pragmatic and Sociolinguistic Research

The thesis focuses on the occurrence and communicative function of pragmatic markers as interactive reinforcement between the interviewer and the interviewee in three political interviews found on the Internet. The analysis is based on pragmatic and sociolinguistic research. At first, terminology which is necessary for understanding the following analysis is introduced. Then, corpora as well as speakers are described. Attention is paid to background data, which can influence the occurrence, frequency and function of the markers. The thesis also observes several interpretations of the term ``pragmatic marker{\crqq}. A detailed examination of individual pragmatic markers adopting the quantitative as well as qualitative approach follows. The occurrence, frequency and function of pragmatic markers in this genre is summarised at the end of the work.
Cariovascular Profile, Level of Its Control in Relation to Complications in Type 2 Diabetic Patients

1. A sample of 415 type 2 diabetic (DM2) subjects was examined. The control of cardiovascular risk factors, e.g. blood pressure, lipids and glucose metabolism, is unsatisfactory in this sample. There is a lack in nonpharmacological treatment and regimen - physical activity, smoking, low energy diet. The choice of antihypertensive treatment is satisfactory. Many patients have only the monotherapy. The antihypertensives are underdosed. We are not sure about the compliance. The treatment of dyslipidemia is not sufficient enough, there is a lack of the statin + fibrate combination therapy. The nonpharmacological regimen in needed for better glucose metabolism compensation. The pharmacotherapy should be indicated earlier (esp. metformin). 2. The prevalence of microvascular complications (mvc) was high in this sample (54%), especially the diabetic nefropathy (95%). The macrovascular compliactions (MVC) were present in 27% subjects, more in men, 55% of them had coronary heart disease. Subjects with MVC had more often mvc. The risk profile was worse in subjects with any vascular complications. The common risk factors in association with both MVC and mvc were the age over 60 years and hsCRP plasma level over 1 mg/L. Thesis is the benefit of cholesterol lowering in prevention KVCH, because I think that this very issue is one of the most dynamic part of preventive medicine and preventive cardiology at all. The work also tries to explain the risk factors, treatment, importance of lowering cholesterol and also explain the basic mechanisms of lipid metabolism and their individual components.

Gut microbiota is important for our health and well-being, but when its composition is disrupted, it can induce or perpetuate several chronic inflammatory disorders, including inflammatory bowel diseases (IBD). The mechanisms which distinguish protective microbes from the deleterious or indifferent ones are largely unknown. The aim of this thesis was to study the interaction of the immune system with microbes that have different relationships to IBD pathogenesis. Escherichia coli is a predominant aerobic microorganism of the gastrointestinal tract. This species includes microbes implicated in induction of IBD as well as in its therapy. Four E. coli strains with different relations to IBD were selected for our experiments: E. coli Nissle 1917 (EcN), which has been successfully used in IBD therapy, E. coli strains LF82 and p19A, which have been implicated in the pathogenesis of IBD, and E. coli strain K6, which has neither been implicated in pathogenesis nor in protection from this disease. The experiments were performed both with living bacteria and inactivated ones. Part I. The aldosterone synthase gene (CYP11B2) polymorphism T-344C in blood pressure and left ventricular hypertrophy. BACKGROUND: Aldosterone is a key cardovascular hormone, it significantly influences volume, pressure and electrolyte balance. Aldosterone plays an important role in development of left ventricular (LV) hypertrophy and myocardial fibrosis. The aldosterone synthase gene (CYP11B2) is an important candidate gene region in essential hypertension. DESIGN AND METHODS: We assessed the influence of the T-344C polymorphism of aldosterone synthase - the rate-limiting enzyme in aldosterone biosynthesis - on the structure of the left ventricle in young normotensive men. The population included 113 normotensive mid-European Caucasian men aged 18-40 years (mean 27 +/- 5 years). We also studied the association of -344T/C polymorphism of the CYP11B2 gene with the presence and severity of hypertension in 369 individuals, of whom 213 were hypertensive patients (139 controlled hypertensive, 74 resistant hypertensive) and 156 were healthy normotensive subjects. The main aim of thesis is the econometric model suggestion of consumption function corresponding with contemporary economic conditions in the Czech Republic and Slovakia. The thesis is also focused on testing selected models of consumption function and assessing the suitability these models for contemporary conditions of real economic situation in the Czech Republic and Slovakia. The close attention is concentrated on testing three major economic models of consumption function--the Theory of consumption (Keynes, 1936), the Life Cycle Hypothesis (Modigliani, 1986) and the Permanent Income Hypothesis (Friedman, 1957), where is made by econometric modeling of time series a comparison of real data with macroeconomic theories of consumption function.
