Development and Validation of HPLC Method for Determination of 3-Hydroxykynurenine in Biological Material

Charles University Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical Chemistry and Pharmaceutical Analysis Candidate: Dominika Knoblochová Supervisor: PharmDr. Petr Kastner, Ph.D. Title of thesis: Development and Validation of HPLC Method for Determination of 3-Hydroxykynurenine in Biological Material 3-Hydroxykynurenine is one of the metabolites of tryptophan and is studied especially for its oxidative modulatory activity, which provides the ability to protect cells from oxidative stress. It is formed from kynurenine by catalysis of the enzyme kynurenine monooxygenase, whose activity is monitored at the Department of Pharmacology and Toxicology. It is necessary to quantify 3-hydroxykynurenine in a biological matrix for its study. The aim of this diploma thesis was to develop an HPLC method for the determination of 3-hydroxykynurenine in biological material using high performance liquid chromatography and subsequent method validation. During development, several mobile phases with different pH values were tested. The resulting mobile phase consisted of 100 mM formic acid (pH = 2.25) and methanol (95:5). Different types of stationary phases were also tested, from which a YMC TRIART PFP column with dimensions of 15 × 3 mm and a particle size of 5 µm was finally chosen. A UV-VIS...

Engaged Art in Prague's Public Space after 2000 from the Perspective of Participants

This diploma thesis is focused on engaged art in the Prague public space after 2000. The theoretical part deals with the characteristics of engaged art and artivism and places them in the context of civil society. It describes the participants dealing with engaged art in Prague after 2000, the topics of the participant's interventions and the tactics they use for their implementation. The empirical part offers, based on biographical research and ethnography, an perspective of the participants. It describes their motivations leading to the creation of interventions, their relationship to public space and to the audience. Work deals with the goals of their activities and focuses on their own concept of success. Last but not least, the thesis is focused on the political orientation of the participants. The thesis thus provides a comprehensive view of contemporary engaged art and pursues on its important role in our society.
The influence of the League of Community Schools for inclusion of socially disadvantaged students

The thesis deals with the education of socially disadvantaged pupils, specifically pupils studying at three selected schools that are members of the "Liga komunitních škol"(The League of Community Schools), which focuses on community-based and inclusive education. The theoretical part analyzes the social deprivation and its impact on the education of pupils who come from families with low socioeconomic status. It also discusses disparities in education and inclusive education. The practical part is critical to assessing whether pupils studying at three selected schools receives a quality inclusive education, which they should help to overcome their handicaps to full-fledged integration into society.
