Regulatory mechanisms of fetoplacental vasculature in hypoxia
Kafka, Petr ; Cvachovec, Karel (advisor) ; Zoban, Petr (referee) ; Bláha, Jan (referee)
The placenta and the lungs are the only organs that perform a function of blood oxygenation and have a common characteristic - vasoconstriction in response to acute hypoxia. Thus the reaction is in an opposite manner than smooth muscle of systemic arteries does. In this study we focused on the mechanisms that regulate fetoplacental blood vessels in conditions associated with hypoxia. An experimental model of isolated dually perfused rat placenta was used. Our findings demonstrate an essential role of Rho kinase signaling pathway in the mechanism of this hypoxic fetoplacental vasoconstriction (HFPV). Chronic exposure to hypoxia causes a sustain elevation of vascular resistance on fetoplacental blood vessels. It was demonstrated in experiment with low-viscosity salt solution perfusion. Our current results show a significant increase of the fetoplacental vascular resistance with blood perfusion which better reflects in vivo conditions. An increase in fetoplacental vascular resistance may lead to placental hypoperfusion and consequent fetal undernutrition, which is considered one of the key causes of serious fetal and neonatal problems, especially intrauterine growth restriction. Diabetes mellitus is a well-known factor affecting fetal growth. Both chronic hypoxia and diabetes act on vessels partly...

Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of Biophysics and Physical Chemistry Candidate Mgr. Veronika Biricová Supervisor Doc. Ing. Alice Lázníčková, CSs. Title of Doctoral Thesis Study of radiolabeling of PAMAM dendrimers of 1st and 4th generation with diagnostic and therapeutic radionuclides. Treatment and diagnosis of diseases using radioactive isotopes is important field on which pharmaceutical research is focused. In order to obtain drug which would specifically bind to receptors released by cells of affected organs and tissues and concurrently provide them a high dosage of radiation, one part of research projects is focused on finding carriers and also new metal chelators suitable for binding in a high number on biologically active molecule. This dissertation thesis is focused on a research of a new bifunctional chelator of trivalent metal ions bound in a high number on a branched dendrimer. Different parameters evaluating suitability of chelator for bond creation with radioactive istotopes were reviewed. From the biggest part is this thesis focused on labeling of dendrimer macromolecules with ligands bound on their surface with three different radioactive isotopes. Success of the labeling under different conditions based on value of radiochemical purity...
Using Windows Communication Foundation to creation client/server applications
KAFKA, Petr
This bachelor work deals with usage of Windows Communication Foundation technology to create application of client/server type. The main goal is to create learning materials, which will familiarize reader with creating WCF clients and services, written in Czech language. These materials contain a number of samples used to explain the problems.
