Sport and physical culture. Sociological perspective

This work is devoted to the issue of sport and physical culture from sociological perspective. It deals with social aspects of contemporary sport and physical culture and how these activities were constituted in the past and how they developed to the present specific form. The text is divided into seven main chapters. The content of the first chapter is a terminological definition of the basic concepts of this work. A large space is devoted to the definition of the term sport, which is difficult to specify. Different authors vary significantly in defining this term. The second chapter describes the history of physical culture and sport. Emphasis is placed on the capture of social context affecting the form of sporting activities of the period. This chapter is divided into seven parts. The first describes the sports activities in ancient cultures - ancient China, India, Mesopotamia, Egypt and Crete. The second part deals with sport activities of ancient Greece, where the care of the body acquired non- ordinary meaning. The third part of the historical chapter is devoted to physical culture of the Roman Empire, whose main features were formed with regard to permanent military expansion. Transformation of \kur{Chlamydomonas reinhardtii} ptDNA

The aim of this master thesis was to test and compare two available methods of genetic transformation (biolistics, electroporation) of the plastid genome of green algae Chlamydomonas reinhardtii. For biolistic transformation a wide range of experimental parameters which generally influence ptDNA transformation efficiency was optimized: physiological condition of acceptor cells, type and size of microparticles, pressure of propulsion gas (helium), length of projectile trajectory, transformation of cells directly on selective medium (containing spectinomycin 150 ?g/ml) or on nonselective medium (without spectinomycin) and form of transforming DNA. In contrast to nuclear transformation ptDNA transformation of the experimental object C. reinhardtii by means of electroporation hadn´t been described. Hence a wide range of values of different physical parameters which can significantly influence the transformation efficiency was tested.

Effect of recombinant toxin Cry3Aa on development of \kur{Spodoptera littoralis}

This bachelor thesis compares the effect of recombinant and natural Bacillus thuringiensis Cry3Aa toxin upon the development of non-target insect Spodoptera littoralis (Lepidoptera, Noctuidae). Caterpillars of Spodoptera littoralis were fed with Premix artificial diet containing both toxins in 8 ppm concentration. Following characteristics were monitored: weight of larval increments, pupal weight, duration of the 5th, the 6th, prepupal and pupal instar, number of laid eggs and number of hatched larvae. The efficiency of conversion of ingested food (ECI) was calculated and growth curves were constructed. No significant effect of both toxins was recorded.
