Prevention of burnout syndrome among emergency service workers

The thesis is focused on the prevention of burnout syndrome in the IRS workers. This thesis was prepared in the context of the SARS-CoV-2 pandemic, which hit the Czech Republic in 2020, and due to the pandemic situation, including extraordinary governmental measures, integrated rescue system workers were exposed to a great psychological, but also physical load. In the theoretical part of this thesis, the integrated rescue system was briefly described, but also the stress that contributes to the development of burnout syndrome in the long term. Subsequently, the theoretical part described the burnout syndrome and its phases. Furthermore, the symptoms of burnout syndrome, risk factors or treatment were presented. An indispensable part,without which this thesis could not be complete, is the section devoted to the prevention of burnout syndrome. The main objective of this thesis was to list the factors that contribute to the development of burnout syndrome including preventive measures that reduce the likelihood of burnout syndrome. The topic of prevention of burnout syndrome was focused only on the personnel of basic units of the IZS. To achieve the stated aim of this thesis, a custom questionnaire was constructed and used in the survey. The practical part of this thesis deals with the prevention of burnout syndrome in the workers of basic units of the IZS. The questionnaire that was constructed contains twenty-four questions, fourteen of which are related to the SARS-Cov-2 pandemic and also intertwined with the prevention of this syndrome. From the analysis of the questionnaire survey, it was found that the prevention of burnout syndrome is neglected among the integrated rescue system workers in the Czech Republic. On the basis of subjective answers of the personnel of basic units of the IZS it was found that they may be more at risk of developing burnout syndrome due to the SARS-CoV-2 pandemic. Since no official diagnostic questionnaire was used, I would have welcomed the involvement of some erudite social psychology personnel on this issue. From the resulting findings included in this thesis, a recommendation for practice has been developed, which includes five practices that, if applied, could improve the current state of the art in the prevention of the syndrome in future integrated rescue workers.

- 4 - ABSTRAKT Barbora Danielová In vitro-cultures of medicinal plants-III. Elicitation is one of the methods that can be used for increasing secondary metabolites production in in vitro cultures. The present study investigates the effect of methylviologen as the abiotic elicitor on the flavonoids production in the callus and suspension cultures of Ononis arvensis L. after 6, 12, 24, 48, 72 and 168-hours methylviologen administration. The cultures were cultivated on Murashige-Skoog medium with addition of 10 mg/l α-naphtylacetic acid (α-NAA). The flavonoids content was determined using spectrophotometry methods. The maximal content of flavonoids was detected in callus culture after 168-hours methylviologen elicitation in concentration c1 (2,1929 . 10-3 mol/l), the production was increased about 444%. The maximal increase of flavonoids production in suspension culture was after 12-hours methylviologen elicitation in concentration c3 (2,1929 . 10-5 mol/l), the production was increased about 42%. In the opposite, the decreased flavonoids production was detected in callus culture after methylviologem treatment in the concentration c1 (2,1929 . 10-3 mol/l) after 6 and 12 hours about 100% and in suspension culture in the concentration c2 (2,1929 . 10-4 mol/l) after 12 hours about 88%.

Genista Tinctoria in vitro - the affecting of Secondary Metabolites Production

Genista tinctoria in vitro - the affecting of secondary metabolites production Elicitation is one of the methods that can be used for increasing secondary metabolites production in in vitro cultures. The present study investigates the effect of 6, 12, 24, 48, 72 and 168-hours lasting elicitation by two elicitors - substance A (N-(3-iodo-4-methylphenyl)pyrazine-2-carboxamide) and substance B (5-tert-butyl-6- chloro-N-(3-iodo-4-methylphenyl)pyrazine-2-carboxamide) on the isoflavonoids production in the callus cultures of Genista tinctoria. The cultures were cultivated on Schenk-Hildebrandt medium with addition of 0,5mg/l 2,4-dichlorophenoxyacetic acid a kinetine in concentration 0,1 mg/l. The isoflavonoids content was determined using HPLC. The maximal content of genistin was detected after 6-hours elicitation with substace A at concentration cA 2= 2,95 . 10-4 mol/l, the production was increased about 3200%. Within using substance B the maximal increase of genistin was detected after 12-hours lasting impact of concentration cB 1=2,33 . 10-3 mol/l (by 5640%). Generally higher production of isoflavonoids was proved when substance A was used.
