Analysis of pre-mRNA alternative splicing products and their importance in breast cancer oncogenesis.

Breast cancer is the most common tumor disease diagnosed in women worldwide. The hereditary character of this disease is observed in 5-10 % of all cases, and it is usually caused by a pathogenic mutation in one of the predisposition genes. Although a variety of pathogenic mutations in the coding sequences of these genes was described, the cause of the disease is still unknown in many familial cases (> 50%). A great number of identified pathogenic mutations were localized in the consensus splicing sites, which results in the formation of aberrant mRNA splicing variants and their damaged protein isoforms. However, little is known about mutations affecting regulatory splicing sites, which can result in the translation of similarly affected mRNAs. In this work, we proposed a method for indirect detection of mutations affecting the natural splicing pattern of any gene of our interest based on multiplex PCR and NGS with high sensitivity. Verification of this method on the BRCA1 model gene revealed the presence of the total of 94 splicing variants in peripheral leucocytes and healthy breast and adjacent fat tissues. Processing of gothic wall painting as compensational haptic aids for the visually impaired

The theoretical and practical part is dealing with the detail of a czech wall painting with the motif of the Virgin Mary from the peak era of the Middle Ages. The theoretical part introduces the marian theme. It describes its iconography, meaning in art and closely specifies the division of images according to devotional themes. The next part is focused on the description and analysis of the motif of the Virgin Mary the Protector, which is depicted on wall painting in a cloister of the Church of Presentation of the Blessed Virgin Mary in České Budějovice. In the practical part is fundamental creation of the relief as a haptic aid for the visually impaired. Created reliefs are polychrome reconstruction of the wall paintings of the above-mentioned motif. The work is supplemented by other materials, implementation procedure, sketches and photo documentation.
Processing of architectural elements as haptic aids for the visually impaired

Theoretical and practical work is concerned with an architectural detail of the Czech early Middle Ages architecture. The theoretical part introduces the history of Gothic art and describes stone carvings structures developing from the third quarter of the 13th century until its end on Czech territory. The work focuses in more detail on the history of the city České Budějovice and the main attention is paid to the description of the Church of the Presentation of the Blessed Virgin Mary. The main part of a practical work is a creation of three consoles as haptic aids for the visually impaired. Created objects are shaped and polychromed reconstructions of real consoles of the north wall of the mentioned monastery. The work is complemented of other materials, implementation process, sketches and photodocumentation.
Economics of Discount Portals

This thesis examines the effect of various factors on the sale of vouchers of group buying deals. The data for the analysis were collected from the website Slevomat for four weeks. The observation was performed daily at 8 a.m., 4 p.m., 12 a.m. Based on the existing literature and theoretical considerations, six hypotheses were formulated, three of which turned out to be consistent with the data. It was confirmed, that the decrease in time remaining till the end of the offer induces bigger amount of vouchers sold per time period. Positive reviews and the fact that the offer is displayed on the main page of Slevomat also positively influence the number of sold vouchers. The influence of the amount of earlier sold vouchers on the present purchases ("bandwagon effect") and of the size of the relative or absolute discount was not found. The "Monday effect", according to which sales should be lower at the beginning of the week, was also disproved.
Human's behaviour during voluntary payment in restaurant

The aim of this work is to find out, wheter in a restaurant (which is running "pay what you want" system) people behave selfishly paying as little as possible, and what factors will influence their behavior. I carried out an experiment to find out the result. The experiment took one whole day in a small town restaurant and was not announces in advance. The price for food was upon customers to decide. Drinks were charged the usual fixed price. On the basis of the experiment I have found that acquaintance between customers and staff has positive influence on the payment. Close acquaintance between customers themselves has positive effect too. Gender and age of customers appears insignificant for this experiment. The final result of the experiment shows non selfish acting.
