Charles University, Faculty of Pharmacy in Hradec Králové Department: Pharmaceutical Technology Student: Eliška Kučerová Consultant: Assoc. Prof. PharmDr. Jitka Mužíková, Ph.D. Title of the Diploma Thesis: Influence of mixing conditions of microcrystalline celluloses with lubricants on compression process and tablet strength. This work evaluates the influence of a type of mixing device and mixing time of three types of microcrystalline cellusoses (MCC) with lubricants on compression process and tablet strength. Microcel® MC 102, MC 200 and Prosolv® SMCC 90 are used as dry binders. Magnesium stearate and sodium stearyl fumarate at 1% concentration are used as lubricants. Two types of mixing devices are used to mix powders, a mixing cube and a 3D Turbula mixer. Mixing time is 2 or 4 minutes. All tablets are compressed by compression force of 5 kN on the Zwick / Roell T1-FRO 50 material tester. The compressibility is evaluated by the energy profile of compression process, sensitivity to the addition of lubricants is characterized by lubricant sensitivity ratio values (LSR). The total energy of compression of all MCC decreases with the addition of both types of lubricants and with longer mixing time, more in the case of mixing in the cube. The highest values of the total energy of compression and at...

Kišková, Martina ; Šnejdrová, Eva (advisor) ; Mužíková, Jitka (referee)
Charles University, Faculty of Pharmacy in Hradci Králové Department of Pharmaceutical Technology Name: Martina Kišková Title of diploma thesis: Adhesive properties of matrix tablets Supervisor: PharmDr. Eva Šnejdrová, Ph.D. The diploma thesis deals with the evaluation of rheological and adhesive properties of the mucin, aqueous dispersions of polymeric carriers and matrix tablets based on chitosan and sodium alginate or iota-carrageenan loaded with the salicylic acid using absolute rotational rheometer. The theoretical part deals with the characterization and classification of matrix tablets, polymeric carriers (sodium alginate, chitosan and carrageenan) and with the principles of evaluation of rotational, adhesive and oscillational tests performed in the experimental part. The mucin from porcine gastric used as a model substrate for adhesion tests behaves as a viscoelastic solid and its adhesive strength decreases with increasing hydration. Significantly higher adhesive strength was found for chitosan at pH 1.2 and sodium alginate at pH 6.8 compared to the adhesive strength of iota-carrageenan. In terms of viscoelastic properties, chitosan and sodium alginate are viscoelastic fluids, but iota-carrageenan is a viscoelastic solid. Iota-carrageenan forms the stiffest gel after hydration at pH 6.8...

This thesis deals with the study of compressibility of directly compressible tableting materials with ι-carrageenan.1,2 In formulations ι-carrageenan is mixed with chitosan, calcium alginate and hypromellose Methocel K15M in the ratios of 1:1, 2:1 and 3:1. The formulations with salicylic acid as a model drug at a concentration of 20 % and sodium stearylfumarate as a lubricant at a concentration of 1 % are also tested. Tablets without the drug are compressed by compression forces of 3, 4 and 5 kN. Tablets with the drug are compressed by compression force of 5 kN and 8 kN. The compressibility is evaluated by energy profile of compression process, another tested parameter is tensile strength of the tablets. The total energy of compression increases with the compression force. The parameter is increased with the addition of chitosan and hypromellose in all ratios as well. With higher proportion of carrageenan in mixtures, its values decrease. The addition of chitosan, calcium alginate, and...

Charles University, Faculty of Pharmacy in Hradec Králové Department of: Pharmaceutical Technology Supervisor: Consultants: Assoc. Prof. PharmDr. Zdeňka Šklubalová, Ph.D. Mgr. Jana Brokešová, Mgr. Daniel Pěček Student: Miroslav Ouzký Title of Thesis: Study of excipientsʼ influence on the drug dissolution from tablets The aim of this work was to study the influence of excipients on the dissolution of the high-dose active substance from tablets. The tablets were compressed from the granules prepared by wet-granulation method. 11 batches of tablets which contained two different fillers: either lactose or microcrystalline cellulose, respectively; and extragranularly added disintegrant: either croscarmellose or crospovidone, respectively, in three concentration levels of 2 %, 3,7 % or 5,4 % were prepared. Tablets were packed into aluminium/PVC blisters. The paddle dissolution test was used to determine the release of the active substance into phospate buffer pH 7,2 at the time of preparation (time 0) and at the time points 1.5, 3 and 6 months of stability assay at 40 řC and 75 % relative air humidity. The results show that the drug release from tablets containing microcrystalline celulose was generally faster than from those containing lactose. The same was true for tablets to which croscarmellose was...

Korpasová, Marie ; Šnejdrová, Eva (advisor) ; Mužíková, Jitka (referee)
CHARLES UNIVERSITY IN PRAGUE Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical technology Name and surname: Marie Korpasová Title of diploma thesis: Adhesive and rheological properties of mixtures based on chitosan Supervisor: PharmDr. Eva Šnejdrová, Ph.D. Diploma thesis deals with evaluation of rheological and adhesive properties of compounds for matrix tablets formulation, based on Chitosan with addition of retardant compound. The retardant compound consists of Sodium Alginate and Hypromellose in concentrations of 30 %, 40 % and 50 %. Theoretical part describes Chitosan, Sodium Alginate and Hypromellose. As next is division of matrix tablets. Theoretically is also described evaluation of rotational, oscillational and adhesive testing which was used in practical part. From viscosity curves can be seen, that viscosity is decreasing with increasing stress. Viscosity increases with increasing concentration of Sodium Alginate and Hypromellose. Oscillational testing is better for gel characterization, because it provides lower stress on test samples. Samples of given compound were prepared by hydrating with phosphate buffer of pH 6,8. Coefficient of consistency K and index flow rate n characterize rheological behaviour of gels. Viscoelastic compounds behave like solids with plastic...
Adhesive and rheological properties of chitosan-based mixtures in media of different pH
Omar, Alaa Abdel Gawad Ahmed Osman ; Šnejdrová, Eva (advisor) ; Mužíková, Jitka (referee)
Title of thesis: Adhesive and rheological properties of chitosan-based mixtures in media of different pH Author: Alaa Omar Supervisor: PharmDr. Eva Šnejdrová, Ph.D. Consultant: Mgr. Juraj Martiška The aim of the diploma thesis is the study of rheological and adhesive properties of tableting materials for matrix tablets based on chitosan. The theoretical part describes in detail the materials used to prepare the matrix tablets, which were chitosan, sodium alginate and hypromellose. The experimental section was focused on the flow and adhesive properties of the chitosan-based mixtures after exposing them to a media of different pH. The flow tests were done on an absolute rotational rheometer. Obtained viscosity curves were analysed by Power law model. Viscosity at 10 s-1 and Power Law model coefficients were used to evaluate and compare the flow behaviour of dispersions with different concentrations of retarding component in different media. The test of adhesion was carried out using a pull away test on a rotational rheometer and evaluated as maximal detachment force. The remarkably high viscosity of 5% dispersions at pH 1.2 is due to the dissolution of chitosan, as sodium alginate is not soluble in acidic media. The increase in viscosity with the concentration of the retarding component is insured...
