Retardant components of the tableting materials are sodium alginate in concentrations of 30, 40 and 50 % or its combinations with hypromellose 15M in a ratio of 1:1 in the same concentrations. Magnesium stearate in a concentration of 1 % is used as a lubricant, indomethacin in a concentration of 20 % is used as a model drug. Tablets without the lubricant and the drug are compressed by compression forces of 3, 3,5 and 4 kN, tablets with the lubricant and the drug are compressed by compression force of 4 kN. The tablets were compressed from the granules prepared by wet-granulation method. 11 batches of tablets which contained two different fillers: either lactose or microcrystalline cellulose, respectively; and extragranularly added disintegrant: either croscarmellose or crospovidone, respectively, in three concentration levels of 2 %, 3,7 % or 5,4 % were prepared. Tablets were packed into aluminium/PVC blisters. The paddle dissolution test was used to determine the release of the active substance into phospate buffer pH 7,2 at the time of preparation (time 0) and at the time points 1.5, 3 and 6 months of stability assay at 40 řC and 75 % relative air humidity. The results show that the drug release from tablets containing microcrystalline celulose was generally faster than from those containing lactose. The retardant compound consists of Sodium Alginate and Hypromellose in concentrations of 30 %, 40 % and 50 %. Theoretical part describes Chitosan, Sodium Alginate and Hypromellose. As next is division of matrix tablets. Theoretically is also described evaluation of rotational, oscillational and adhesive testing which was used in practical part. From viscosity curves can be seen, that viscosity is decreasing with increasing stress. Viscosity increases with increasing concentration of Sodium Alginate and Hypromellose. Oscillational testing is better for gel characterization, because it provides lower stress on test samples. Samples of given compound were prepared by hydrating with phosphate buffer of pH 6,8. Juraj Martiška The aim of the diploma thesis is the study of rheological and adhesive properties of tableting materials for matrix tablets based on chitosan. The theoretical part describes in detail the materials used to prepare the matrix tablets, which were chitosan, sodium alginate and hypromellose. The experimental section was focused on the flow and adhesive properties of the chitosan-based mixtures after exposing them to a media of different pH. The flow tests were done on an absolute rotational rheometer. Obtained viscosity curves were analysed by Power law model. Viscosity at 10 s-1 and Power Law model coefficients were used to evaluate and compare the flow behaviour of dispersions with different concentrations of retarding component in different media. The test of adhesion was carried out using a pull away test on a rotational rheometer and evaluated as maximal detachment force. The remarkably high viscosity of 5% dispersions at pH 1.2 is due to the dissolution of chitosan, as sodium alginate is not soluble in acidic media. Influence of fillers and disintegrants type on properties of tablets with high dose drug.

PharmDr. Jitka Mužíková, Ph.D. Student: Thao Tranová Title of Thesis: Influence of fillers' and disintegrants' type on properties of tablets with high dose drug The work engages in the study of fillers' and extragranular superdisintegrants' influence on properties of tablets with high dose drug. Two types of granulate with lactose or microcrystalline cellulose were produced in high shear granulator. Eleven tablets groups with different type and amount of superdisintegrants were prepared from the granulates. Tablets contained 2 %, 3.7 %, 5.4 % of sodium croscarmellose or 2 % and 5.4 % of crospovidone. The tested parameters of tablets were friability, hardness, disintegration time and density. Accelerated stability studies lasting 6 months at temperature of 40 řC and relative humidity 75 % were done. The tablets were evaluated after 1.5, 3 and 6 months. All tablets had adequate friability, hardness, disintegration and density at T0. Increasing concentration of superdisintegrant decreased disintegration time. This thesis deals with influence of rice extract, as a new kind of glidant on compressibility of microcrystalline celulose as a model filler. Furthermore it evaluates the impact of rice extract on tablet properties. Effects of rice extract were compared with two standard glidants - magnesium stearate and colloidal silicon dioxide. As first the influence of diferent glidants concetrations on flow properties of mixtures were assesed. Tablets were compresed using three different compression forces. A force- displacement record was used to evaluate the compressibility of microcrystalline cellulose and its mixtures with glidants. Subsequently, the properties of tablets were evaluated. The radial strenght, abrasion, and tablet disintegration time were measured. The results revealed that the rice extract has the least effect on mixtures flow poperties.
