Radiolabelling of hyaluronic acid

The aim of the work was studying of recent literature about biophysical and pharmacological features of hyaluronic acid, which are important for application of this biomolecule in many regions of medicine. Another part of work deals with methods of radioactive labelling of compound used for studying of metabolism of hyaluronic acid in vivo and in vitro. Radioactive - labelled material is used for identification of hyaluronan - binding proteins, which play important roles in lot of physiological and pathological phenomena in organism, for biodistribution and pharmakokinetic studies for example an antineoplastic agents based on hyaluronic acid available for treatment of liver tumor.

Labelling of DOTA-Tyr(3)-octreotate by oxidative iodination

SOUHRN Značení DOTA-Tyr(3)-oktreotátu oxidativní jodací Diplomová práce Klára Tvrdíková Univerzita Karlova v Praze, Farmaceutická fakulta v Hradci Králové Katedra biofyziky a fyzikální chemie V této diplomové práci byla vyzkoušena metoda značení somatostatinového analogu DOTA-Tyr3 -octreotátu radioaktivním jódem. Peptid byl značen přímou elektrofilní substitucí a jako oxidační činidlo byl použit chloramin-T. Ten oxiduje jodid na aktivní kationy jódu, které jsou pak začleněny do tyrosinových částí peptidu. K oddělení značeného peptidu a nenavázaného radioaktivního jódu byly použity dvě metody, extrakce na pevné fázi (Sep-Pak Vac 1 cc, C 18 Cartridges) a vysokoúčinná kapalinová chromatografie (Lichrocart Lichrospher C-18, 250 x 4 mm, 5 m). HPLC metoda je výhodnější, jelikož odstraní nenavázaný jodid a zároveň di-jodované deriváty. Dále byla zkoušena stabilita a farmakokinetické vlastnosti radioaktivně značeného DOTA-Tyr3 -octreotátu. Výsledky biodistribučních studií naznačily poměrně rychlou eliminaci jodovaného peptidu z ledvin a z orgánů s vysokou hustotou somatostatinových receptorů (pankreas, nadledviny).

Interaction of Tensides and Organic Dyes in Solutions II

Title of diploma thesis: Interaction of Tensides and Organic Dyes in Solutions II This diploma work is devoted to examination of the aqueous solutions of surfactants and organic dyes in the solution of Britton-Robinson buffer by two spectral methods: UV-VIS absorption spectrophotometry and fluorescence spectroscopy. There are two different dye-surfactant systems investigated: neutral red with sodium lauryl sulfate and phenol red with hexadecyl-trimethyl-ammonium bromide. The possible interactions were observed in the several surfactant concentration levels of the solutions with increased dye concentration. The task of my work was investigation of changes in dye spectra as a consequence of the dye-surfactant interaction and determination of the possible origin of the spectra changes. The results showed that absorption spectra of solutions containing neutral red itself differ sharply from those taken for solutions of neutral red and the tenside. Study of Fluorescence of Selected Benzimidazoles

The influence of used solvents on fluorescence of seven selected benzimidazoles (reduced flubendazole, albendazole, 2-aminobenzimidazole, carbendazim, benomyl, thiabendazole a oxibendazole) is studied in this thesis. It was found, that solvents, used in liquid chromatography as mobile phases, influence the fluorescence of individual benzimidazoles in a very diverse ways. The shape of the fluorescence spectra is very strongly dependent on the structure of a benzimidazole. The obtained results can serve as starting points during a development of chromatographic methods for a benzmidaloles determination with the fluorimetric detection. In the other part of the thesis, a derivatization of hydrolyzed mebendazole with 7- chloro-4-nitrobenzo-2-oxa-1,3-diazole is studied as a way enabling a fluorescence detection of this substance. Unfortunately, the obtained results are not promising.

Stability studies of monoclonal antibody nimotuzumab modified with chelator DTPA radiolabelled with lutetium-177 ([177Lu]Lu-hR3(p-SCN-Bn)DTPA) Diploma thesis David Korec Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of Biophysics and Physical Chemistry In this work was target to study the stability of monoclonal antibody Nimotuzumab, which was modified with chelator DTPA radiolabelled with radioactive Lutetium-177 ([177Lu] Lu-hR3 (p- SCN-Bn) DTPA). The radiochemical purity of the supplied antibody was 61%. For other uses had to purified. The chelator EDTA adds in addition for the labeled antibody. It has lower stability constant than the original chelating agent. He can catch the free ligand, which is excreted through the kidneys. For biodistribution studies is needed to ensure the highest purity that was well described by the distribution of radiopharmaceuticals in small bodies. Purification was used for gel permeation chromatography Sephadex. Purity was about 99%. HPLC analysis after one hour showed a small percentage of low molecular weight forms. It may be modified ligand with radionuclide or free radionuclide. At times long after purification the HPLC profile of low molecular weight fraction appeared in two peaks.
