Tato diplomová práce se zabývá problematikou finančního plánování podniku. V první části práce jsou popsána teoretická východiska, jejichž znalost je pro sestavení finančního plánu nezbytná. Následně je práce zaměřena na představení vybrané společnosti. Poté budou provedeny potřebné analýzy, strategická a finanční, pro zhodnocení její současné situace, které slouží jako podklad pro samotný finanční plán. Ten bude záhy vypracován pro tři následující hospodářské roky v optimistickém . Závěrečná část práce obsahuje zhodnocení a kontrolu tohoto finančního plánu.
Hexahistidine labeling of oligosaccharides

We have presented another approach for labeling of oligosaccharides suitable for\ncapillary zone electrophoresis coupled with mass spectrometric detection.\nOligosaccharides labeled by peptides with hexahistidine sequence have faster\nseparation resulting in two or three times shorter time in comparison to oligosaccharides\nwith singly charged labels.

A novel nanospray liquid junction interface for versatile CE-MS

In this work we present a new polyimide-based liquid junction nanospray interface, combining the advantages of sheath flow and sheathless designs. The liquid junction interface was prepared as a hybrid microfabricated plastic liquid junction sprayer allowing to perform analysis in standard (30-100 micro m ID) separation capillaries. The interface incorporates a self-aligning port for coupling of the separation capillary as well as ports for automated flushing of both the liquid junction and the separation capillary. The interface permits performing CE both with and without electroosmotic flow. The use of large bore capillaries allows injection and preconcentration of larger sample volumes, favoring detection sensitivity. Different compositions of the spray liquid and BGE can be used when needed.

In this contribution, we report the separation of AETMA-labeled glycans on a wide bore (320-μm i. d.) silica-based monolithic capillary column modified by [2-(methacryloyloxy)ethyl]-dimethyl-(3-sulfopropyl)-ammonium hydroxide.
Simple route of caspase-3 FRET sensor synthesis using “click chemistry”

Programmed cell death or apoptosis is regulated process of cell suicide. The central role in apoptosis play cysteine proteases called caspases. Caspases recognize tetra-peptide sequences Asp-Glu-Val-Asp (DEVD) on their substrates and hydrolyze peptide bonds after aspartic acid residues. Various techniques for the determination of caspase-3 are commercially available e.g. Enzyme Linked Immuno-Sorbent Assay (ELISA), Western blotting or flow cytometric analysis. The products of the cleavage can be detected by spectrophotometry, fluorimetry, chemiluminescence (CL) or ELISA. In this work, we suggested fluorescent sensor based on easily prepared Förster Resonance Energy Transfer (FRET). We use very simple chemistry called “click”. This type of chemistry takes advantages of quickness, simplicity and cheapness. “Click chemistry” is based on usage of various functional group and cross-linkers to combine individual molecules together.
