Protein domains utilizable for development of binding molecules

Small protein domains represent basic building blocks of naturally occurring proteins. Many of them exhibit excellent stability, lack disulfide bonds and their structures, therefore, represent attractive tools for generation of artificial binding molecules. First step in the production of novel binding proteins is the definition of a basic domain structure, called "scaffold", which is identified using in silico approaches, resulting in discovery of mutable amino acid residues. Then, randomization of such residues leads to design of a highly complex combinatorial library as a key tool for targeted selection of protein variants. Based on chosen selection approach, the particular protein variants can be tested for their ability to recognize the target molecule with high specificity and binding affinity. Small binding proteins lack post-translation modifications, exhibit thermal stability, are resistant to many organic solvents and can be produced on a mass scale in bacterial host cells. In addition, they can be easily modified and used in vivo with excellent tissue penetration. Due to these beneficial properties, small artificial binding proteins are extraordinary useful biotechnological tools and represent a promising alternative to monoclonal antibodies. This thesis is focused on the Development of the Urban Rail Transport in Prague. This thesis aims to specify the development of the Urban Rail Transport in view of the current state. In this work, a comparison is made with the suburban railway transport in Munich, the evaluation of the establishment of new stations and stops, and the forecast of the development of railway transport in Prague in the long run.
Femtosecond laser spectroscopy of diamond

Due to its extraordinary features and wide bandwidth (5.47 eV), diamond is a very promising material in the field of optoelectronics. By absorbing ultraviolet light, excited charge carriers - electrons and holes - are created in the diamond, which can create excitons due to mutual Coulomb interaction. For low temperatures and high concentrations of photoexcitated carriers, carriers can condense into electron-hole droplets and form an electron-hole liquid. The aim of this diploma thesis is to follow up with previous research at the department and to examine the dynamics of electron-hole liquid in bulk diamond at low temperatures. Using femtosecond laser spectroscopy, we investigate the influence of excitation wavelengths on the dynamics of electron-hole liquid condensation.
Alternative ECG measurement using flexible electrodes for scanning of electrical potentials on the human body

The objective of this bachelor thesis is to measure ECG signal using experimental textile electrodes. The theoretical part deals with heart anatomy, the placement of the heart within the rib cage, heart's electrical activity and ways of measuring it using electrocardiography as well as alternative ECG systems. The practical part of this thesis deals with the use of an experimental ECG T-shirt and wearable electronic equipment. This part also includes the description of the examination method as well as of the equipment which processes and records the signal. The next part this thesis deals with laboratory testing of the experimental ECG T-shirt on a selected sample of people. It also includes an assessment of the measurement results. D. thesis is focused on physical phenomena located at the interface of hybrid nanostructure composed of polycrystalline diamond and polymer polypyrrole. The main method used in our experimental study was ultrafast laser spectroscopy that allowed us to gain new findings about electron recombination processes in polycrystalline diamond layers, polypyrrole and in their hybrid structures. The research was focused on mutual influence of both components, especially through energy and charge transfer. In the first step of our research we carried out optical characterisation of different kinds of polypyrrole and complex study of recombination processes dynamics of photoexcited charge carriers in polycrystalline diamond. The measurements were realized by the methods of time-resolved photoluminescence and transmission spectroscopy in the time scale from picoseconds to milliseconds. Isometric strategy game Citadel

This thesis describes design and implementation of a real-time strategy game Citadel. The emphasis is on application architecture, which uses Allegro library for graphics and controls and Boost Serialization library for saving the game. It also includes description of techniques used in implementation such as influence maps and a target system used by artificial intelligence, pathfinding algorithms, plasma fractal for map generation and a variant of Dinitz's algorithm for maximum flow. Title: Dynamics of photoexcited charge carriers in diamond Author: Bc. Tomas Popelar Department: KCHFO MFF UK Supervisor: prof. RNDr. Petr Maly DrSc., KCHFO MFF UK Abstract: In this work we examine the luminescence from diamond in the strong excitation regime by one-photon or two-photon absorption of femtosecond laser beam. Measured sample was very pure type IIa diamond prepared by CVD method which was held in cryostat in order to measure temperature dependant luminescence in range from 12 K to 300 K. The signal was collected and analyzed either by spectrograph or streak camera so we gained time-integrated and time-resolved results. We analyzed only the part of the spectrum containing contributions from electron-hole liquid (for low temperatures), free excitons and probably exciton complexes. For higher temperatures where the condensation is not possible the contribution from e-h plasma was too weak compared to free excitons and was only detectable by time-resolved measurements. Other time-dependant results were obtained by pump and probe experiment with which we examined a change of life- times based on mode of excitation (one-photon or two-photon one) and also an influence of other beams on the condensation into electron-hole drops.
