Application of bioscanners and videotechnique for studies of fish migration
Porš, Jaroslav ; Kalous, Lukáš (advisor) ; Lucie, Lucie (referee)
In recent years there has been a significant decrease in abundance and species diversity of migrating fish. A significant decrease was recorded especially in diadromous fish species such as Atlantic salmon (Salmo salar) and eel (Anquilla anquilla). A significant population decline was also observed in potamodromous abundant fish species. Transverse barrier which prevents fish migration in rivers and impaired water quality are considered as the main causes of this situation. At present there is being improved a surface water quality, habitat rehabilitation and restoration of the possibility of free migration of aquatic animals. Design and construction of fish ladders are the most important factors to improve conditions of the fish migration. Closely related to this is a migration permeability testing of these fish ladders while the biological function and the activity of a single technical solution is evaluated. A non-invasive automatic monitoring method using a bioscanner Riverwatcher was selected to assess the functionality of fish passage on the river Ohře in Kynšperk nad Ohří. Bio scanner was installed at the end of the track of fish passage. Riverwatcher camera system, a product of Vaki Aquaculture Systems Ltd., was used in the Czech Republic for the very first time. Direct catches of fish samples in the river using an electric unit (section under fishway) were also used for the data collection. Size and species selectivity was tested at the fish ladder. The influence of external factors affecting the course of migration, such as flow rate, water temperature and light, was monitored at the same time.

Landscape development and landscape values of a selected area in a specified relief type
Učňová, Klára ; Halamová, Jana (advisor) ; Lucie, Lucie (referee)
The submitted thesis deals with landscape analyzing and permanent landscape structures evolution in chosen area with respect to land relief. Also with documentation and evaluation of landscape values (such as medieval field patterns, scattered vegetation, viewpoints, relief, etc.) and last but not least with suggestion of recommendations for future development, including proposal of new development, specific for chosen area. The theoretical part consists of summary of current status. Mainly the elements characterizing landscape of the Czech Republic are defined here. The thesis also describes history of Czech landscape since the Neolithic period and also evolution of medieval field patterns. The landscape aesthetics and its influence to humans from emotional and content point of view is mentioned in the theoretical part as well. In the next chapter there is an evaluation of maps and description of possibilities of their usability for evaluation of landscape evolution. The practical part is about project itself. At first the suitable area based on maps from 1 952 was chosen and their comparison with present state. By maps vectorization (both historical and present one) supported by personal exploration, two map underlays were created. Based on them, the statistical data for evaluation and comparison of land use categories evolution were created. Also the analysis of appearance of given land use categories was performed. Comparison of changes in evolution of balks is the part of the project as well.. One of this thesis purposes was also evaluation of landscape directly on the spot. Based on this evaluation and personal feelings from the area, design for better land usability was created. This design was also economically considered. In discussion at the end of the thesis was determined that epoch around year 1952 was a breakthrough for landscape evolution. After this year, there was a huge decrease of ploughland area. This decrease was replaced by permanent grassland and forests. In the end there is an estimation about medieval field patterns important for landscape in chosen area and also anywhere else.
Level of sales and maintenance of cut flowers in CR
Žížalová, Dagmar ; Augustinová, Ludmila (advisor) ; Lucie, Lucie (referee)
In my dissertation work about the topic The level of sales and general care of cut flowers in the Czech republic I have tried to describe all the aspects which influence the conservation, longevity and quality of cut flowers. Currently, where this market segment is seeing dynamic growth, there is also an increased demand for growing cut flowers, their after harvesting technology, keeping and prolonging their longevity. The choice of cut flowers in the Czech Republic is very versatile which is highly appreciated by the customer who prefers to have flowers with higher longevity in his vase. My dissertation work is divided into three parts. The first part is mainly dealing with all the aspects influencing the picking (temperature, humidity, light, fertilization, age of the flowers), the time of picking, cutting of the stem, temperature by picking and the quality of the cut flowers. There are also mentioned other factors influencing the storage of the plants (temperature, water, ethylene), the packaging description and transport of the flowers. Further attention is paid to the keeping and maintaining of the flowers in shops, by the customer and also to the types of agents prolonging the longevity of the flowers in the vase. The second part is dealing with the assortment of imported flowers and greenery into the Czech Republic. The categorization of plants is divided into different categories -- the imported cut flowers, exotic flowers and leaves used in floristry. The world producers of cut flowers also get a mention and the favorite types of flowers are -- Alstroemeria, Anthurium, Cymbidium, Dendrobium, Dianthus, Freesia, Gerbera, Gladiolus, Chrysanthemum, Lilium, Rosa, Tulipa, Zantedeschia. For each type there is also a description of the sensitivity, picking season, type of cut, treatment after picking, transport and longevity in the vase. The third part is dedicated to a customer knowledge survey about the care of cut flowers. As part of my work florists were approached from four Czech regions with a request to participate in a survey and to complete a questionnaire (Ústecký region, Středočeský region, Plzeňský region and Jihočeský region). This resulted in 62 completed questionnaires out of 100 flower shops, and the answers were put together in tables and graphs. The main focus of the questionnaire was the analysis on the various possibilities of purchasing cut flowers, the most frequently sold flowers, the florists providing their customers with information as to how to care for the flowers, which agents to use to keep the longevity of the flowers and whether these agents are also offered to the customers. The survey was also aimed at the cleanliness of the vases and the care for the flowers in the shops. The results of the survey have reassured me, that the florists have to face the economic situation of their customers, the competitiveness of other retail chains selling flowers (where the care for flowers is non-existent), that they have to be mindful of the selection of the flowers as they are subject to fast deterioration and they are always trying to please their customers, so that they return.

Floricultural utilization of Dionaea muscipula and its in vitro culture
Danko, Jiří ; Augustinová, Ludmila (advisor) ; Lucie, Lucie (referee)
This work focuses on the proper foundation of the Venus Flytrap (Dionaea muscipula Ell.) culture under in vitro conditions. The hypothesis of this paper is that the solution of Savo containing sodium hypochlorite will be able to sterilize effectively seeds of Flytrap without damaging them at certain concentrations. This experiment was founded in three repetitions in two locations - in the Prague Botanical Garden, where one repetition was founded, and at the Department of Horticulture at the Czech University of Life Sciences Prague, where two more repetitions were founded. The seeds were treated in five solutions of Savo Original namely concentrations of 5, 10, 15, 20 and 25 % with the addition of soaking agent Trend 90 and planted on 1/2 MS medium where plants were grown for 4 months and then transplanted on the same medium for another 4 months. Cultivation was carried out under light conditions 16 hours of light, 8 hours of dark. During the cultivation the formation of contamination was monitored after the foundation of the culture, and also the number of germinated seeds and the number of plants with developed phyllomae. Also the qualitative appearance and the mass of plants were evaluated. The results show that all solutions of Savo are effective enough for sterilizing seeds for in vitro culture. Influence of individual solutions on the development of germination is statistically significant at two solutions - 15 and 20% solutions of Savo. In these solutions, the development of germination is much faster than the other. On the final number of germinated seeds nor the mass of plants, these 2 solutions didn't have a statistically significant effect. During cultivation, there were occurred some interesting phenomena - spontaneous multiplication of plants, production of polyphenols, flowering of explants, chlorosis and vitrification of tissues. After the previous cultivation, the plants were transferred to non-sterile substrates. Variants of substrates were perlite, peat and perlite in the ratio 2:1 and pure peat. Before cultivation, substrates were treated with a fungicide Previcur Energy. High humidity was ensured during the cultivation. For these conditions the mixture of peat and perlit seems to be the most suitable because of minimum losses.
The nutrients balance on the farm when fertilizing by organic fertilizers
Pejsar, Petr ; Černý, Jindřich (advisor) ; Lucie, Lucie (referee)
The aim of this thesis is the evaluation and balance calculation of the basic nutrients in soil. The content of this thesis is focused on the evaluation of nutrients input, particularly nitrogen, phosphorus, potassium, magnesium and sulphur in organic fertilizers, while taking into account the direct influence of the nutrients after their application as well as long-term effects during delayed and recurring application of the organic fertilizers. During the observed period from 2008 to 2014 the following individual fields were assessed Březinka (68,51 ha), Jiříkovo (16,93 ha), Kocourovské (58,16 ha) and U Akátu (35,84 ha). The balance of nitrogen, phosphorus, potassium, magnesium and sulphur in these certain areas was assessed within the scope of balance on individual fields for one crop rotation period. The balance calculation inputs include the amount of nutrients that was provided in form of organic and mineral fertilizers, which in case of nitrogen was furthermore extended by the amount acquired by its symbiotic fixation, overall deposition and sowing inputs (i.e. soil surface balance of nitrogen). The outputs of the balance calculation consist of amounts of major nutrients that were drawn away by crop yield including the calculated losses in case of surface balance of nitrogen. In the thesis an elementary balance of organic compounds was performed together with evaluation of change of the supply state of P, K and Mg based on the available data from agrochemical soil properties conducted in the period of 2009 to 2015 and balance results for individual nutrients. The results of surface balance of nitrogen in individual fields exhibited notably positive values, which according to the final balance might be the cause of heightened or excessive nitrogen loss. It is however difficult to determine the direct and residual effect of nitrogen applied trough the organic fertilizers and its utilization by plants. Without the mineral fertilizer inputs, the resulting values of other nutrients in specific years were dependant on the amount of input trough organic fertilizers. Within the scope of simple balance of organic compounds for individual fields, the overall sum of organic compound requirements were achieved over the observed years.
