Myosin I and actin binding proteins in cell

Many studies have established the presence and essenciality of actin in the nucleus. Recently, actin has been associated with processes in the nucleus ranging from chromatin remodeling to transcription, splicing or nuclear transport. To ensure the dynamics of the nuclear processes, actin is coupled with one of the main motor protein such as myosin. This study demonstrates a structural role of actin and the nuclear myosin I (NMI) take in the transcription of ribonuclear genes by RNA polymerase I (Pol l). We suppressed the transcription Pol I in vitro by microinjections of antibodies anti actin and anti MNI. The series of in vitro experiments confirm transcript Pol I inhibition after applying antibodies anti actin and MNI on pure DNA as well as on pre-assembled chromatine template. The co-immunoprecipitation experiments reveal direct bound between actin, NMI and rRNA genes and transcription complex Pol I. As actin binds to the primer and elonged Pol I molecule, NMI interacts with subunit of Pol I and is capable of assembling into productive initiation complex by binding up to TIF-IA, transcriptional factor responsible for regulation rRNA synthesis. Effect of Type 2 diabetes on the mouse reproductive parametres

Infertility is defined as an inability to conceive a child within one year of regular sexual intercourse. It affects up to 15 % of couples worldwide (WHO, 2010). The male factor contributes to the total infertility with more than 50 %. Fertility of a man is influenced by several factors such as genetic background, environment and various diseases such as diabetes mellitus (DM). Diabetes mellitus is a serious health problem that affects 451 million people worldwide (18-99 years) and the number of people with this disease still increases (Cho a spol., 2018). In addition, parenthood is postponed to middle age when the fertility decreases and metabolic diseases such as type 2 diabetes mellitus (DM2) appear. The aim of this thesis was to investigate the effect of type 2 diabetes on reproductive parameters of mouse inbred line C57BL/6J compared to the control group and the possible effect of paternal diabetes on the first filial generation. In the evaluation of the effect of DM2 on reproductive parameters, we used innovative methods to study internal state of sperm and testes. CoNct usloN The aim of the thesis was to establish, in the national conditions, technology of preparation of antibody reconrbinant fragrnents and to verífo the cornplete procedure using several model monoclonal antibodies with a potential diagnostic and therapeutic use. For tfuee antibodies' mAb TU-20, M75 and MEM97, recombinant |Ťagments in various formats (scFv fragments monovalent and bivalent, i.e. diabody, and intrabody for intracellular expression) were constructed and for their heterologous expression, vectors allowing expression in E. coli (as cytoplasmic inclusions, periplasmic inclusions and in soluble form) and in Drosophila 32 cells (expression of glycosylated forms of scFv fragments into the medium) were used. In case of proteins expressed in irrsoluble form, especially scFv F|1.2.32, renaturation procedures to obtain active scFv fragments were developed and optimized. The efnect of the length of the linker -(GlyrSer).- (where x is I to 4) connecting the variable domains of the light and heavy chain on the formation of different multimeric forms of scFv rvas studied.
