Determination of Cystine, Cysteine and Homocysteine in Biological Matrix
Bašová, Pavlína ; Navrátil, Tomáš ; Šestáková, Ivana ; Novotný, Ladislav ; Šenholdová-Dlasková, Z.
In order to determine cystine, cysteine and homocysteine contents separately in a mixture of these compounds in real urine samples from patients suffering from cystinuremia and homocistinuremia, three voltammetric methods were tested: Brdička reaction, reduction of their complexes with copper ions and cathodic reduction of cystine. In all cases the hanging mercury drop electrode was used.
Metabolic Causes of Thiodiglycolic Acid Formation and Importance of its Determination
Šenholdová-Dlasková, Z. ; Navrátil, Tomáš ; Přistoupil, T. ; Přistoupilová, K.
Thiodiglycolic acid (TDGA), is one of the normal products of human metabolism, and occurs at low concentrations in urine. An increased level of TDGA in urine is a sensitive indicator of human organism exposure to some physiologically active or toxic substances, gases and vapors (vinylchloride monomer, etc.). The attention was paid to suggestion and explanation of the metabolic pathways related to thiodiglycolic acid in non-exposed organism.
Determination of thiodiglycolic acid concentration in case remethylation disorder
Navrátil, Tomáš ; Šenholdová, Z. ; Heyrovský, Michael ; Přistoupilová, K. ; Přistoupil, T. I. ; Surovcová, H.
The new simple voltammetric method we described recently was useful in determining thiodiglycolic acid (TDGA) levels in urine samples. The article deals with revealing of time dependencies of the concentration levels of this metabolic product in healthy persons as well as persons suffering from various diseases. The corresponding metabolic processes are described.
Voltammetric Determination Phenylglyoxylic Acid in Urine Using Graphite Composite Electrode
Navrátil, Tomáš ; Šenholdová, Z. ; Shanmugam, K. ; Barek, J. ; Surovcová, H.
Phenylglyoxylic acid is metabolized in human body from vinylbenzene (styrene) (from which polystyrene is produced). Presented work deals with determination of this metabolite in human urine using carbon composite electrode. In the article are compared the results of analysis of the certified material (urine), achieved on the mercury and on the carbon composite electrodes.
Recyclation and Transformation of Homocysteine as a Part of Detoxication Processes in the Cell
Přistoupilová, K. ; Přistoupil, T. ; Šenholdová, Z. ; Navrátil, Tomáš ; Heyrovský, Michael
The presented metabolic pathways explain how the transformation of homocysteine affects the detoxication processes in the human cell. On the one hand homocysteine is recycled to methionine; on the other hand it is transformed to cysteine. Both pathways are connected by B12 vitamin. Their relation to the thiodiglycolic acid (TDGA), which represents a part of the detoxication processes, is demonstrated on the scheme. The changes in TDGA concentration in urine were found to depend on the changes in redox equilibria (of vitamin B12, folic acid, homocysteine). Key Words: Thiodiglycolic acid, TDGA, vitamin B12, folic acid, metabolic pathways, homocysteine, detoxication.
Změny koncentrace TDGA v moči jako možný marker metabolismu thiosloučenin
Přistoupilová, K. ; Přistoupil, T. I. ; Šenholdová, Z. ; Navrátil, Tomáš ; Heyrovský, Michael ; Fenclová, Z.
The new simple voltammetric method we described recently, proved already to be useful in determining thiodiglycolic acid (TDGA) levels in urine samples of people influenced by some chemicals, victuals and pharmaceuticals. The present pilot study is a continuation of our previous work devoted to test general feasibility of the method. We concentrated upon time dependent quantitative changes of TDGA excretion into urine of selected volunteers. The TDGA level in all morning samples was the highest in comparison with samples collected during the day. The TDGA level was dependent on the health condition of each person involved. However, a further marked increase of TDGA in the morning urine was observed individually, in general when vitamin B12 had been administered in the evening the day before. Vitamin B12 is known to play an important role in the metabolism of one- and two-carbon units, of homocysteine and of sulfhydryls in general. Certain differences in excretion course of TDGA.
The Use of Voltammetry by Interesting Poisons by Lead
Šenholdová, Z. ; Navrátil, Tomáš ; Vlček, K. ; Žák, J. ; Mádlová, P. ; Surovcová, H.
Two interesting nonprofessional intoxications by lead and their treatments are described. The first one was the case of a man, who by mistake swallowed 20 peaces of lead slugs. The other one was the case of the woman, who used a lead containing liniment prescribed by an astrologist to treat diabetes.
Does the Mercury Represent a Danger in Polarographic Laboratory?
Navrátil, Tomáš ; Rakovcová, H. ; Šenholdová, Z.
The danger represented by the use of mercury in polarographic (voltammetric) laboratory is discussed. All possible forms of mercury were taken into account (metallic, vapors, amalgams, inorganic salts). The history of professional and nonprofessional intoxications by mercury was discussed.
Daily Rhythm of Excretion of Thiodiglykolic Acid in Urine
Navrátil, Tomáš ; Šenholdová, Z. ; Heyrovský, Michael ; Přistoupilová, K. ; Přistoupil, T. I.
Thiodiglycolic acid (TDGA) represents one of metabolites of S-karboxymethylcysteine and it is excreted in urine. This acid can represent one of many detoxication ways of human organism.
