Elucidation of ERK1 and ERK2 protein kinases effect on cap-independent translation initiation
Přibyl, Miroslav ; Vopálenský, Václav (advisor) ; Vomastek, Tomáš (referee)
Protein kinases ERK1 and ERK2 are one of the most studied proteins in cell signalling. Both proteins are involved in a plethora of processes, such as phosphorylation and activation of kinases as part of signalling pathways. Enzymes ERK1 and ERK2 are part of MAPK/ERK signalling cascade, connected to many cellular including cell proliferation, cell growth or differentiation. The MAPK/ERK signalling cascade is often activated in different types of tumors, making it a candidate for developing new chemical inhibitors. One of the important questions in fundamental research of ERK1 and ERK2 protein kinases is the search for difference between these proteins. Current knowledge points to redundancy of both proteins, howver several examples suggest otherwise. Recently, the work presented in Casanova et al. 2012 indirectly suggests divergent effect of ERK1 and ERK2 on cap-independent translation initiation. In the Laboratory of RNA biochemistry we focus on HCV IRES (Hepatitis C Virus Internal Ribosome Entry Site) dependent translation initiation. This diploma thesis lead to establish RNA interference method in our laboratory and to establish reporter system to study ERK1 and ERK2 effect on HCV IRES dependent translation initiation. Based on our data acquired during our research, we present in this work...

Theoretically, It was summarized the anatomy and kinesiology of the shoulder joint, rotator cuff tears, impingement syndrome and treatment of these diagnoses. Much of the theoretical part also focuses on a summary of physiotherapy care for patients with this condition. The special part is the case of the patient after rotator cuff repair. This section contains the medical history of the patient, input and output kinesiology analysis, therapeutic units and their results. Results: The objectives of physiotherapy care were met. Data for case study I have gained during the special practice in hospital Bulovka in January 2016. Keywords: shoulder joint, rotator cuff, case study, physiotherapy
ERK1/2 MAP kinase - Structure and Interaction Partners
Přibyl, Miroslav ; Vopálenský, Václav (advisor) ; Žíla, Vojtěch (referee)
Extracellular signal molecules are recognized by membrane receptors on the surface of eukaryotic cells. Receptors transmit the signal into the intracellular space where activation of the concrete enzymes occurs. Activated enzymes may be protein kinases that phosphorylate the substrate proteins corresponding to the requirements for specific recognition by a protein kinase. Substrate proteins may be structural proteins and enzymes, which in turn transmit the signal or directly affect the physiological processes of the cell. The protein kinase family accounts for ERK1 and ERK2 (Extracellular signal-regulated kinase 1 and 2), enzymes which affect cell growth, cell cycle and numerous other physiological processes of the cell. Protein kinases are dynamic molecules, which undergo a series of conformational changes during their catalytic cycle and whose stability and function are affected by conformational changes. Conserved amino acid residues carry out the function of protein kinases. These factors are also involved in interactions with protein substrates and regulatory proteins, and are responsible for specific function of protein kinase.

Boarding of students living in the dormitory Hvězda
Přibyl, Marek ; Tuček, Milan (advisor) ; Balážová, Eva (referee)
Dining together, feasting and other rituals associated with food form are significant part of the history of human society. However, nowadays we can see the decline of their importance. The aim of my thesis is to show how students perceive their dining at the dormitory and how much the dining is important for them. This thesis could be divided into three parts. The first one presents the social and cultural meaning of the joint dining in the human society and there are also presented research questions. The second part is explaining the research and the methods of data collection and analysis used in this work. The results of the research are then summarized in the last part. After moving to the dormitory the boarding of the students is very individual and they perceive it as something ordinary and therefore not so important. The change occurs in two possible ways. First, the individual change of personal values associated with meal (e.g. by confrontation with long-term impact of dining). Second, a meal shared with partner when food is again seen as important. During a visit home these changes in boarding habits can cause conflicts with parents. My thesis is also focused on borrowing and sharing equipment resulting in closer relationship between students.
Návrh metody výběru nejvhodnější lokality pro logistický objekt
Přibyl, Martin
The thesis deals with the method layout of choosing the most suitable locality for a logistical entity. Regarding the difficulties of multicriterion decison process, when quantitative and qualitative criterions enter to the decision-making, and regarding the fact, that for optimalization of quantitative criterions exist many well accessible instruments, the aim of this thesis is the layout of a new model, which will evaluate only qualitative criterions in chosen localities. Submitted model is based on the expert system's principle and it compile through statistical enquiry empirical knowledge of more than 110 experts -- logistical managers, who evaluated localities for logistical entities during their praxis. Integral part of an expert system, knowledge base, contains 57 characteristics divided into six groups, which enable evaluation of the locality appropriateness for the logistical entity. Consequently, the expert system is capable to compare particular variants and to vizualize the classification. The proposed method represents a subsidiary deciding instrument while solving strategic problems in company development and it facilitates objective decision-making about suitable location of logistical entity. In the thesis a case study is processed. This case study assesses 3 localities, where centres of University forest enterprise Křtiny are situated.
