Meiotic sex chromosome inactivation within mouse spermatogenesis
Homolka, David ; Jansa, Petr (advisor) ; Kubelka, Michal (referee) ; Pěknicová, Jana (referee)
Meiotic sex chromosome inactivation (MSCI) is an essential epigenetic process, which transcriptionally silences X and Y chromosomes during spermatogenesis. It is accompanied by substantial chromatin remodeling resulting in a formation of so called sex or XY body, which is a characteristic of male pachytene spermatocytes. In spite of MSCI indispensability for male fertility, its biological role and molecular nature still remain rather unclear. However, the described link between chromosomal asynapsis and transcriptional silencing demonstrated that MSCI is tightly associated with the asynapsis of largely non-homologous sex chromosomes and is a specific form of more general mechanism called meiotic silencing of unsynapsed chromatin (MSUC). The essential role of MSCI was demonstrated using mouse models, such as carriers of X- autosome translocations, where anomalous synapsis of sex chromosomes leads to impairment of MSCI and male sterility. Intriguingly, the exclusive spermatogenic arrest is a hallmark of not only X-autosome translocations but even various autosomal rearrangements, including autosomal translocations, inversions, or other structural mutations. Because the rearranged autosomes often intimately associate with the sex body, it...

Studies on glycosphingolipids as immune targets in bioprosthetic heart valves
Jansa, Petr ; Doležal, Martin (advisor) ; Jirkovská, Anna (referee)
Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical Chemistry and Drug Control Candidate: Petr Jansa Supervisor: prof. PharmDr. Martin Doležal, Ph.D. Supervisor - specialist: prof. Susann Teneberg, M.D., Ph.D. Title: Studies on glycosphingolipids as immune targets in bioprosthetic heart valves The treatment of valvular heart disease represents approximately 20% of all cardiac surgery. One alternative of this treatment is replacement of diseased valve with bioprosthetic heart valves (BHV). These BHV are manufactured from divergent human or animal tissues e.g. porcine pericardium. Although this treatment is beneficial for certain groups of patients, it is susceptible to some complications such as rejection of the xenograft or early BHV deterioration. In both of them the immune system is involved and both might result in BHV failure. This study aims to isolate and characterize important targets of this immune response - glycosphingolipids (GSL). Therefore, 8 non-acidic and 7 acidic GSL from porcine pericardium have been isolated and characterized by mass spectrometry and carbohydrate binding assays. The acidic GSL from goat erythrocytes have been isolated and characterized. Notable is characterization of a new NeuGc-containing GSL - NeuGc-GT1b ganglioside....
Internal motivation of adolscents in sport
Špidlenová, Kristýna ; Jansa, Petr (advisor) ; Křiček, Jan (referee)
Title: Internal motivation of adolescents in sport Objectives: Check to compare emotionally and motivational processes in adolescents using questionnaires. Focus on boys and girl agend 16-18 years, specifically for secondary school students with sports and non-sports focus. Statistically processed and then Compaq the measured data, between the sexes and between schools. Then sorts the results in clear graphs and images. Methods: Students'responses analyzed using questionnaires. School choice was made in secondary schools in Liberec and Prague. Results: Internal motivation does not affect the propensity to exercise. Athletes do not show better results. Keywords: Motivation, motive, emotion, achievement motivation, adolescent.
Physical education in school curriculum at primary schools
Hájevská, Marcela ; Jansa, Petr (advisor) ; Křiček, Jan (referee)
Physical education in educational programme at primary schools The main objective of this work is to find out a conception of physical education in school educational programmes on the second level at primary school. Our research was made with the help of a questionnaire, which is the mostly used research method in social disciplines. We have done with the help of the questionnaire a collection of informations about the interviewee and his opinions and facts to the interwieved themes. The questionnaire is constructed as a battery of questions, that the interviewee answers in form of clear, closed answers yes, no, I do not know or with a choice of alternative possibilities or free open answers. We have found, that the most interviewees had participated on the creation of school educational programmes and their contents are coming from the circumstances of the school. Keywords: children and youth, curiculuum, movement activity, life style, reform
Activity life style of adolescent youths
Bárek, Michal ; Jansa, Petr (advisor) ; Kovář, Karel (referee)
The main aim of this dissertation is to find out to which extent do the today's teenagers lead an active lifestyle. Specifically, it's focused on uncovering importance of sport and athletic activities for gymnasium students aged 15- 18. The survey took part on gymnasium schools on the Vysocina and South Bohemian region via questionnaire method. The intention of the survey was to compare attitudes of both Vysocina and South-bohemian students' pool. 570 gymnasists were interview in total. Our quest was to confront and prove opinion diversity between the regions, i.e. Vysocina and South-bohemian region, as well as to carry out a gender comparison. The results in most cases affirmed our operative hypotheses. All in all, we can state that sport, as well as athletic activites in general still take an important part of today's teenage lifestyle. Keywords: Teenagers, questionnaire, survey, sport, athletic activities
Characterization of the Hstx1 and Hstx2 hybrid sterility candidate genes
Kašíková, Lenka ; Jansa, Petr (advisor) ; Rothová, Olga (referee)
Speciation, the formation of new species, is an essential evolutionary process that causes species diversity on the Earth. At the beginning of this process is the separation of two populations by a reproductive barrier that prevents gene flow between these populations. One of the mechanisms, which enable reproductive isolation, is hybrid sterility (HS). It is a mechanism of postzygotic isolation that is described in a number of eukaryotes. The first discovered gene of hybrid sterility in vertebrates is the mice gene Hst1, later identified as gene Prdm9. By genetic and molecular analysis the locus on the X chromosome was determined, whose interaction with Prdm9 causes sterility or reduced fitness in male hybrids. This locus contains two genetic factors: Hstx1, causing an abnormal morphology of spermatozoa, and Hstx2, causing an arrest in spermatogenesis in pachytene spermatocytes and sterility. In my thesis I focus on the effect of deletion of a candidate hybrid sterility gene Fmr1nb on the X chromosome. The analysis of males B6N.Fmr1nbmut with deletion variants of the Fmr1nb gene showed that Fmr1nb is one of the factors influencing spermatogenesis. An increase in morphologic abnormalities in spermatozoa occurred in males with Fmr1nb gene deletion. This phenotype is identical with Hstx1. The effect...
