Utilization of pulse labelling techniques for studying the dynamics of proteins and protein complexes

(In English) Mass spectrometry (MS) techniques are routinely used to probe the structure and dynamics of proteins and protein complexes. Although MS techniques lack the high resolution of data provided by X-ray crystallography, NMR, or cryo-EM, they excel in providing insights into analyte dynamics, structure, and interactions with other components, such as ligands. This doctoral thesis presents a contribution to the field of structural biology employing and extending covalent labelling approaches, namely Fast Photochemical oxidation of Proteins (FPOP) and oxidation by singlet oxygen (1 O2). These approaches were followed to study the structure, dynamics, and interaction of proteins, nucleic acids, and protein-DNA complexes in solution. Initially, FPOP was used to investigate the interaction interface of FOXO4 and DAF16-DNA response element and to show the possibilities of analyzing such a complex using both 'bottom- up' and 'top-down' approaches. Furthermore, an isotope depletion strategy combined with multiCASI-ECD proved effective in delivering structural information with the highest possible resolution for mapping protein-DNA interfaces. Subject of master thesis “Comparison of the possibilities of financial security of real estate” is the issue of alternative funding of housing in Czech republic. This thesis deals with the major housing finances products. It surveys building society accounts and mortgage loans withing the selected financial institutions. In the practical part these products are compared in an effort to determine the best form of securing of chosen property whose value was determined by estimation of cost of the property for credit management.

Táto práca sa zaoberá problematikou identifikácie sieťových zariadení na základe ich chovania v sieti. S neustále sa zvyšujúcim počtom zariadení na sieti je neustále dôležitejšia schopnosť identifikovať zariadenia z bezpečnostných dôvodov. Táto práca ďalej pojednáva o základoch počítačových sietí a metódach, ktoré boli využívané v minulosti na identifikáciu sieťových zariadení. Následne sú popísané algoritmy využívané v strojovom učení a taktiež sú popísané ich výhody i nevýhody. Nakoniec, táto práca otestuje dva tradičné algorithmy strojového učenia a navrhuje dva nové prístupy na identifikáciu sieťových zariadení. Výsledný navrhovaný algoritmus v tejto práci dosahuje 89% presnosť identifikácii sieťových zariadení na reálnej dátovej sade s viac ako 10000 zariadeniami.

This bachelor's thesis deals with optimization of payment obligations of self-employed person by change of legal form. The theoretical part describes the constituents of payment liabilities, the methods of determination their amount and optimization opportunities. In the practical part there is an analysis of the entity and subsequently there is a finding of the best solution of determination of payment liabilities with regard to the costs which are connected with the change of the legal form of the entity.

At the conclusion, the bachelor thesis evaluates which model is optimal for the company proprietor and the company.
