Kohoutová, K. Implementation of age management in a selected sector. Master’s the- sis. Brno: Mendel University in Brno, 2023. The thesis deals with age management in the health and social services sector. The aim of the thesis is to develop proposals and recommendations for improving the current level of age management. Quantitative and qualitative research was con- ducted to develop recommendations. The qualitative research was done in the form of semi-structured interviews with managers and the human resources department staff members. The quantitative research involved the processing of data obtained from the project: 'Proposal for measures to prevent risks of damage to employee’s health as a result of occupational accident or an occupational disease financed from the contribution under Section 320a(b) of Act No. 262/2006 Coll., the Labour Code, as amended in 2022'. Based on this research, recommendations are made for insti- tutions in the health and social services sector, which should contribute to improv- ing the age-related management of employees, to supporting and improving the working capacity of employees in this sector, and therefore increasing employee sat- isfaction in the workplace
Characterization of a recombinant exonuclease nsp14 of the SARS-CoV-2

Coronaviruses are enveloped viruses with a genome composed of a single-stranded positive-sense RNA molecule. With genome sizes, in some species, exceeding 30 kb, they represent the largest known RNA viruses. They infect birds and mammals, including humans, causing a wide spectrum of diseases. Most human coronaviruses predominantly cause mild respiratory infections. However, in the last two decades, there have been two extensive epidemics and a global pandemic of severe respiratory illnesses, often ending in fatalities, caused by coronaviruses. The most recent and extensive of these was caused by the coronavirus SARS-CoV-2 (severe acute respiratory syndrome-related coronavirus). Within the coronavirus genome, there is a gene encoding a non-structural protein 14 (nsp14), which possesses 3'-5' exonuclease and methyltransferase enzymatic activities. The exonuclease nsp14 participates in the repair of misincorporated nucleotides during viral genome replication, and its presence is exceptionally rare among RNA viruses. The exonuclease activity of nsp14 is significantly enhanced by the binding of another viral protein, nsp10, which lacks enzymatic activity but acts as a critical cofactor for several enzymatically active coronavirus nsps. Eastern Slovaks' reflections of the year 1989. Biographical narratives

This diploma work deals with the reflections of Slovak citizens for 1989's, focusing at the region of Košice city and the surrounding area, especially eastern Slovakia. The fall of the communist regime influenced the further development of the whole of Czechoslovakia. Diploma work focuses at the Slovak nation, which is an integral part of Czechoslovak history, but Czech knowledge of Slovak region is severely restricted. Memories and stories of Eastern Slovak habitants are building blocks of this thesis used to factually illustrate that time. In addition, thesis is based both on the available literature sources and archival materials from the collections of the State Regional Archives in Košice city. The theoretical part of work is based on historical sources and results of various researches on November '89. The research part of the diploma work deals with views, opinions and attitudes of Eastern Slovak's participants of November 1989. Interviews with narrators are analyzed and interpreted using oral history methods. This diploma work aims to reflect the everyday life of people, their work and living conditions before and after November '89. Structure and interactions of selected forkhead transcription factors

This diploma thesis is a part of a project aiming to develop and study specific inhibitors of FOXO3 transcriptional activity. FOXO3 belongs together with FOXO1, FOXO4 and FOXO6 to FOXO subfamily of forkhead family transcription factors. FOXO transcription factors are evolutionary conserved proteins playing important roles in numerous cellular processes, such as apoptosis, cell cycle regulation and metabolism. Due to their ability to induce apoptosis and to block the cell cycle they are considered tumor suppresors. However, it has been shown that increased activity of FOXO proteins is connected with many kinds of cancer. In such cases FOXO proteins function to maintain cell homeostasis. They promote tumor resistance against chemotherapy as well as they speed up its growth. The aim of this project is to develop specific inhibitors able to bind to FOXO3 DNA-binding domain (DBD, residues 156-269) and to block its interaction with target DNA. Development of specific inhibitors of FOXO3 transcriptional programme requires knowledge of solution structure of all FOXO DBDs and detailed insight into their interaction with target DNA. Study of interactions between low-molecular mass compounds and the DNA-binding domain of forkhead transcription factor FOXO3

This bachelor thesis is part of a project focused on studying low-molecular mass compounds able to inhibit the interaction between DNA-binding domain of human forkhead transcription factor FOXO3 and the target DNA. FOXO3 is one of four members of FOXO class transcription factors which belong to forkhead family of transcription factors. Forkhead transcription factors are evolutionary conserved proteins playing important roles in numerous cellular processes. These include cell-cycle regulation, oxidative stress response, control of cellular metabolism and apoptosis. FOXO3 plays an important role in cancer cells where it acts not only as a tumor suppressor but also can enhance their resistance to chemotherapy. Considering its biological functions, the study of small-molecule inhibitors of FOXO3 transcription factor is of particular importance. This bachelor thesis was focused on compound S9 oxalate as a potential inhibitor of FOXO3-DNA interaction. The classification of accommodation facilities in the Czech Republic and foreign countries

The aim of my thesis is to aprise the reader with the meaning and purpose of classification of accommodation in the Czech Republic. I am trying to compare the classification of 2009 with the present classification valid since 2010 till 2012 but also with classifications of other countries. For this comparison I chose the classification of the accommodation in Great Britain which is according my opinion very detailed but also with the classification of other, mainly European countries (with the American classification I compare the classification only in general terms).
