Opioid Crisis in the USA and its relevance to the Czech opioid situation
Sovík, David ; Kopsa Těšinová, Jolana (advisor) ; Nekola, Martin (referee)
The bachelor thesis focuses on the topic of opioid abuse in the United States of America and in the Czech Republic. The current situation of opioid abuse in the USA is called the "Opioid Crisis". The thesis describes the historical development of the problem, including its division into three main phases, in each of which a different opioid is the most abused. In the first phase, it is prescription opioids that are most commonly abused, while the last two stages are dominated by illicit opioid abuse of heroin and fentanyl. The identification of the primary causes of the Opioid Crisis is an important part of the thesis. Purdue Pharma is explicitely mentioned as having manufactured opioids that have directly contributed to the widespread abuse of opioids. Within the thesis, a typology is created in order to clarify which social groups in the USA are the most susceptible to becoming addicted to opioids. In the part of the thesis dedicated to the situation in the Czech Republic, a historical and current perspective of national opioid abuse is provided. Potential present and future threats that could lead to a worsening of the situation are also identified.

Estimation of illegal drug consumption in Kazakhstan
Ayapbekova, Dinara ; Vopravil, Jiří (advisor) ; Radimecký, Josef (referee)
Background: The republic of Kazakhstan is located on the "Northern Route" by which opiates are smuggled from Afghanistan to the Russian Federation and further into the European Union. For this reason, the state is under constant high risk of the drugs leaking to the local market, leading to drug abuse and related issues within the population. The drug situation in Kazakhstan is further complicated by the internal factor of the Chuy Valley which features the largest area of wild- growing cannabis in the world. The aim of this thesis is to estimate the consumption of cannabis-derived drugs and heroin in Kazakhstan based on the latest national and international reports and research. In terms of research, the thesis examines which organisations collect statistical information on users of illegal drugs and what methods are used to do so. Methods: the data analysis consisted of a secondary analysis of existing collected data through quantitative descriptive statistics and a qualitative analysis of individual international and national reports. Drug consumption was estimated based on demand. Results: According to estimates, there were 127,800 problem drug users in Kazakhstan in 2014; the total volume of consumed heroin was estimated to more than 46 tonnes. The estimated consumption of cannabis drugs in...
Narcoterrorism: Searching for the crossroads of drug trade and political violence
Kolínský, Prokop ; Makariusová, Radana (advisor) ; Záhora, Jakub (referee)
The main objective of thesis ''Narcoterrorism: Crossroads between drug trade and political violence'' is to describe and analyze the phenomenon of narcoterrorism, a concept that combines in its core the politically-motivated violence, the illicit drug trade, and the use of terrorist methods. The goal will be to establish a comprehensive theoretical framework for the various types of narcoterrorism and the different supportive types that may occur between the politically-motivated or ideological groups, and the drug-trafficking criminal groups. This framework will be later primarily tested on the case of Colombia, where various warring ideological and criminal groups will be observed and analyzed. Other cases that will be secondarily used to further prove the established assumptions, will be focusing on other countries which seen in recent history a surge of political or criminal violence, and are either manufacturing or trafficking illegal drugs. These will include Peru, Mexico, and Afghanistan. In the final section of the thesis, the theoretical framework will be assigned to real historical cases, and a question of the prerequisites of narcoterrorist emergence will be discussed.

Methadone and its usage in treatment of opioid drug addiction
Chona, Kembe ; Svoboda, Petr (advisor) ; Hejnová, Lucie (referee)
Methadone, a synthetic opioid created in the 1940s is a potent mu opioid receptor agonist. Opioid receptors form a sub-group of the GPCR super-family. Their most significant role is the inhibition of neural pathways by regulating the activity of ionic channels and effector proteins. µ-opioid receptors are the site of action of heroin, methadone and other classical opioid agonists. Due to the opioid receptors distribution in both the central nervous system and peripheral tissues, methadone affects a wide variety of functions in the organism. Methadone induces many of the effects of classical opioids including analgesia, respiratory suppression, sedation, euphoria. While originally being developed as an analgesic it had soon shown potential for other therapeutic methods. Methadone maintenance therapy was introduced in 1963, by professor Vincent P. Dole and his team. It quickly became clear that methadone substitution therapy is indeed very effective and shows the highest ability to retain patients. Thanks to its high oral bioavailability, higher intrinsic efficacy and long terminal half-life methadone is the first choice drug for opioid substitution therapy. Methadone, used in appropriate doses produces only mild adverse effects and has the ability to normalize physiological homeostasis disrupted by...
