Biochar effect on distribution of organic matter in soil

Biochar is a carbon-rich material that can be used in agronomy as a soil conditioner. The selection of the source biomass and the conditions used during pyrolysis (for example residence time and temperature) can have different effect on the physicochemical properties of this material. In this bachelor thesis biochar was produced from oat bran, woodchips, corn and compost by pyrolysis at 600 °C. The woodchips were also pyrolysed at temperatures in the range 400–700 °C. These biochar samples were subsequently leached with model rainwater. The main aim of this thesis was to assess how the conditions of biochar production influence the quantity and nature of substances, which can be released from this material. In the second part of this thesis, the commercial biochar Agrouhel (produced from sewage sludge at temperature 450–470 °C) was used (“Káňa”). This biochar was leached with model rainwater and dilute solutions of citric acid (which simulates root exudates) and hydrogen peroxide (which simulates enzymatic biochar degradation). Physicochemical characterization (pH and conductivity) of the extracts was realized after each leaching cycle. Additional analyses (UV-VIS and ICP-OES) of the solution were measured after the 1st, 5th and 10th cycle. The original solid biochar and biochar samples after the 10th cycle of leaching were used to determine the inorganic and organic fractions and to determine the elemental composition of the organic fraction (TGA and EA). The laboratory cultivation experiment with biochar Káňa was performed on the model plant (Lactuca sativa). Biochar obtained after termination of the cultivation was analysed (pH, conductivity, ICP-OES, TGA and EA) and the results were compared with data obtained for biochar Káňa leached by different model solutions. Fulvic and humic acids were extracted from the original biochar Káňa as well as from the biochar obtained after the termination of cultivation experiment. These samples were analysed on their inorganic and organic content and the abundance of organic elements (TGA and EA). Afterwards, the results were compared to each other and the correlations between the laboratory exposure to each soil factor and the cumulative exposure during the soil culture experiment were assessed.
Study on potential applications of glutamic acid polymer

The subject of the thesis is study of possible applications of isoform of glutamic acid polymer (-PGA). The theoretical part is focused on the properties of this biopolymer and potential applications in various areas. Producers and mechanisms of biosynthesis are also mentioned. In the experimental part, the polymer was firstly characterised by following methods: FT-IR spectroscopy, TGA, DSC and SEC-MALS. Its isoelectric point, antimicrobial activity and solubility in various solvents were also determined. The biopolymer was also precipitated by divalent cations and its interaction with oppositely charged CTAB surfactant was studied. The main experimental study was researching the effect of -PGA on viability of Saccharomyces cerevisiae and Lactobacillus rhamnosus under stress conditions by flow cytometry. The performed stresses included ethanol exposure, high temperature and freezing stress, in which its effects were compared to conventional cryoprotectants. The cells of the mentioned microorganisms were also stressed osmotically and exposed to model gastrointestinal juices - gastric, pancreatic and bile. The protective effects of -PGA on the cells were recorded in ethanol stress on Lactobacillus rhamnosus. Its excellent cryoprotection properties were confirmed and its protective effect of gastric juice exposure on Saccharomyces cerevisiae cells was also observed. At the end of the experimental part, -PGA/alginate beads suitable for encapsulation of probiotic bacteria and -PGA/chitosan nanoparticles for encapsulation of biologically active substances.
Raman spectormetry in Art

This thesis deals with the use of Raman spectroscopy in the arts, specifically in the identification of precious stones and semi-precious stones in various art works. This work describes the basics of this method, options and practical use based on selected scientific publications. The existing results of research in recent years shows that this spectroscopic analytical method on the rise. It is thanks to the many advantages that this method offers skilled in the art. The main advantages of Raman spectroscopy include speed and affordability analysis, minimal or no sample preparation, and most importantly - non-destructive to sample and explore the possibility of sample on the spot, without having to often very difficult transport to the laboratory. Characterisation of \kur{Drosophila melanogaster} mutated for all genes of the Sirtuin family

The aim of my study was to create a Drosophila line lacking the expression of all Sirtuin genes, check its developmental phenotype and characterise its response in stress conditions. The flies had bigger weight than controls, they had decreased fertility and fecundity and they developed more slowly. They showed a trend towards increased resistance to chill coma, but they did not show a significant difference in starvation or oxidative stress assay. Its effect on lifespan is being investigated.

These materials are quite popular last few years, after they were predicted and then char- acterized by [4] as topological crystaline insulators (TCI). TCI is a quantum state of crys- tals, in which symetry causes existance of the metal states on the surface. Samples had been prepared from molten flux and by using Bridgman method. Homogenity and pu- rity of single crystals had been determined. Compounds PbxSn1−xTe and PbxSn1−xSe crystalized in cubical structure and were defined as Fm-3m space group, existance of PbxSn1−xSe in Pnma space group were confirmed. 1
