Využití spektrofotometrických metod ke kontrole falšování potravin

The theme od this dissertation is " Application of Spectrophotometric methods for inspection adulteration of food". In the theoretical part, there are stated chapters regarding adulteration of food (in general), adulteration of honey, quality requirements on honey, spectroscopy and its categories. The dissertaion is focused on spectrophotometric measurements of honey and sugar syrup samples where colour and consistence of honey was monitored. The techniques used for the analysis are NIR spectrophotometry and UV/VIS spectrophotometry. The results gained were statistically and graphically processed. It follows from the outcome that using FT-NIR for adulteration of honey detection is an appropriate method for honey inspection. The least effective method for proving adulteration of honey is the spectrophotometry specification of the colour of honey. Using this method, it is not possible to prove the links between colour of honey and colour of sugar syrup.
Activity of extracelular acid phosphomonoesterase of the soil and the roots in differently managed Norway spruce stands

The presented study was aimed to measurement of extracellular acid phosphomonoesterase activity in Norway spruce (Picea abies /L./ Karst.) pure stands. All study plots were chosen with respect to silvicultural treatment. Norway spruce mature stand 108-year-old (108); 36-year-old Norway spruce stand thinned from above (C3); 36-year-old Norway spruce stand thinned from below for (D4). Research was situated in Ecosystem station Rajec-Nemcice, the Czech Republic. The nature forest region of Drahanska Vrchovina upland (Drahansky bioregion) is ground area for Ecosystem Station Rajec-Nemcice, soil type was recognized as modal oligotrophic Cambisol (KAmd'). Typical is fir-beech forest vegetation zone. There are two major goals. As for soil activity of extracellular acid phosphomonoesterase, it was expressed from the view point of basal activity of the enzyme studied jointly with the response following the adding of the salicyl acid. As for roots, only the basal activity was measured by p-nitropohenol concentration. Presented data indicate negative effect of the salicylic acid adding into the enzyme activity.
The effect of stress on basic urban environment ecophysiological characteristics on tree

The subject of this bachelor thesis is the effect of stress on Platanus acerifolia which grows in different areas of the city. The first location is in the centrum of the city (Lannova třída) and the second location is in the city´s park in České Budějovice (Stromovka). There was a search of quantity of photosynthetic pigments and maximum of photochemical efficiency PS (photosystem) II. This search was conducted by a metod of measuring chlorophyll fluorescence and spectrophotometry. Samples of leaves from Platanus acerifolia were taken during growing season May to September. There were three trees chosen from each location and there were five leaves from each tree examined by metod of measuring chlorophyll fluorescence. The leaves were examined approximately at the same height and the examination was done at noon time. There were 150 leaves examined in total. There were four leaves of each tree used for a method of spectrophotometry. Those leaves were immediately taken to laboratory where the examination took place. There were 120 leaves used for this metod in total. The search showed that the more stressful environment Platanus acerifolia has a smaller content of photosynthetic pigments and photochemical efficiency of PS II decreases. Furthermore, the study showed that Platanus acerifolia belongs among relatively resistant trees. Values maximum photochemical efficiency of PS II and chlorophyll measured at both sites is almost statistically different. The statistical difference between the two sites were usually recorded at the beginning and end of the growing season.
The efficiency of the anionactive surfactants degradation in a constructed wetland vegetation bed.

This bachelor thesis is focused on the validation of the anionactive surfactants spectrophotometric determination method. Data from the seasonal monitoring of the anionactive surfactants in a constructed wetland for wastewater treatment with a horizontal subsurface flow were used for the anionactive surfactants degradation efficiency determination and for the determination of the relationship of the wastewater inflow, the constructed wetland´s water temperature, and the constructed wetland´s water redox potential to the degradation efficiency. The spectrophotometric method of the anionactive surfactants determination with methylene blue was used during the study.
