Monitoring system for monitoring of laboratory processes

The presented work deals with the development of a monitoring system for monitoring the operation of the laboratory. The theoretical part of the work includes information on the safety of operation and work in biological laboratories, measured physical quantities and the principle of their measurement. The monitoring system is built on a compact ESP32-CAM camera and a WeMos D1 development board. The practical part consists of the design and implementation of a given system and the creation of a full-stack application for displaying live input from the laboratory for monitoring measured quantities in real time.
Segmentation of cells in images from high-speed digital camera

The first part of this thesis is dealing with topic of microscopic techniques focusing on monitoring of cardiomyocytes. Techniques used for image segmentation are described in the second part. The practical part is about image segmentation of six records. The results are presented and discussed. The contraction functions are described in the final part.
Preparation and Characterization of Drug Carrier Systems on the Basis of Magnetic Nanoparticles

This bachelor thesis deals with the study of magnetic nanoparticles, optimizing their preparation and stabilization. The nanoparticles with different ratios of Fe2+ and Fe3+ ions were prepared by coprecipitation by variously concentrated sodium hydroxide. Three samples of particles were selected and were further observed. Their stability in Milli-Q water, buffer, polyvinylalcohol, hyaluronic acid and chitosan was studied in function of time using methods of dynamic and electrophoretic light scattering. By UV-VIS spectrometry it was confirmed that the magnetic nanoparticles were actually prepared. Nanoparticles coated with chitosan and hyaluronic acid were then further characterized using FT-IR, XRD and SEM.
Design of a Complex Modular UHV Aparature for Preparing, Observing and Modifying of Nanostructures in Situ

The thesis deals with the development of complex modular ultra-high vacuum system based on ultra-high vacuum scanning electron microscope UHV SEM. This discourse focuses primarily on engineering design of the system, which is underlying demands of ultra-high vacuum, limitation of mechanical vibrations and future R&D applications. The device is designed for preparing, observing and modifying of nanostructures in situ.
Design of the electron microscope

The theme of this diploma thesis is the design of a scanning electron microscope with a focused ion beam. Specifically, the thesis is focused on the design of the microscope covers and the adjacent workplace of the operator for Tescan Orsay Holding a.s.. Design is solved as the first proposal aimed at future innovation of the design of the entire product line.
Accuracy of measurement on workshop microscope

This thesis deals with the performance of the measurement accuracy in the optical measuring systems. The first part describes basic principles of metrology and the guidelines of the expression of uncertainty and the factors which influence measurement result. The second part classifies optical measuring systems and describes some measuring principles of them. The final part compares two optical gauges and the evaluation of some of their characteristics through the use of very simple experiments.
Contrast in image acquired by ionization detector in VP SEM

This work deal with problems of investigation materials electron beam. This project is focuses on investigation materials by the help of method environmental scan electron microscopy (ESEM) and describes her patterns. The perfection of ESEM excel above all at studies non - conducting or water containing, often biological samples. These samples not to be in no way prepared or cut – and – dries, in contradistinction to classical scanning electron microscope and thanks is under investigation their native surface structure without desiccation breaking. The general aim is determination of contrast in image acquired by ionization detector.
Three-Dimensional Reconstruction of Image in Digital Holographic Microscopy

This thesis deals with the topic of 3D image processing for digital holographic microscopy - numerical refocusing. This method allows to perform mathematically accurate defocus correction on image of a sample captured away from the sample plane and it was applicable only for images that were made made using coherent illumination source. It has been generalized to a form in which it is also applicable to devices that use incoherent (non-monochromatic or extended) illumination sources. Another presented achievement concerns hologram processing. The advanced hologram processing method enables obtaining more data mainly concerning precision of quantities from one hologram — normally, one would have to capture multiple holograms to get those. Both methods have been verified experimentally.
Analysis of the Structure and Properties of Al-Si Alloy during Holding Time of the Melt

This thesis is dedicated to the production of aluminum casting alloys. It maps the course of changes in the morphology of eutectic and its influence on mechanical properties during the long-term meltdown. In particular, the use of image analysis methods contributes to describing these processes. The presented results show that the eutectic shape does not change even during the meltdown for 110 minutes. There is only a roughing and an increase in grain size. There was also no influence on the mechanical properties that fit the practical application of the alloy examined
The Influence of Fines on Pore System of Concrete

Literature does not provide a satisfactory answer to maximum and minimum particle size or the particle size of the mortar phase especially for optimal porosity of concrete. To overcome the shortcomings of the design methods were thought to design a new method for design of concrete mix. This idea is based on a complex analysis of powder materials (determination of granular properties, shape factor of fillers, porosity, packing of powder materials, surface area, etc.) and subsequently to assess the influence to pore system of concrete and physical and mechanical properties in hardened state of concrete. In this work are presented and analyzed two types of fillers – fly ash and limestone dust.
