The bachelor thesis is focused on the evaluation and comparison of selected reproductive parameters in individual breeds of Vienna rabbits. The introduction of the research part is devoted to the origin and domestication of rabbits. The following chapters deal with the anatomy and morphology of the reproductive systems of males and females as well as reproduction itself. Part of the thesis also discusses the genetics of rabbits, and a separate chapter is devoted to the Vienna rabbit. The practical part inc ludes the methodology of the work, which introduces the individual breeds, as well as the comparison and evaluation of selected reproductive parameters between the diffe rent breeds of Vienna rabbits.

Production of lipolytic enzymes by yeasts

This diploma thesis is focused on controlled production of lipolytic enzymes, bioactive substances and lipids by carotenogenic yeasts. Theoretical part deals with characterization of lipolytic enzymes, carotenoids, lipids and their properties, possibility of production and application. In experimental part the enzymes, carotenoids and lipids were produced by red yeasts Rhodotorula mucilaginosa, Cystofilobasidium macerans and Sporidiobolus salmonicolor by submerged cultivation in mineral medium with different additions: glucose, glycerol, fat, fat with glucose, fat with polysorbate 80, fat with glycerol, fat with polyethylene glycol, fat with higher and lower addition of palmitic acid, enzymatic fat hydrolysate, acidic hydrolysate a basic hydrolysate. The activity of extracellular lipase was monitored in medium after 96-hour cultivation. Concentration of -carotene, total carotenoids, ergosterol and ubiquinone was determined by HPLC, concentration of fatty acids and amount of fat by GC. Production had differed depending on used yeasts and substrate. As the best producer of carotenoids Cystofilobasidium macerans was found, ergosterol was highly produced by Sporidiobolus salmonicolor. The production of ubiquinone was almost equivalent in all yeasts and lipolytic activity was the highest in Sporidiobolus salmonicolor. The patricular medium sample with high lipolytic activity was further separated and analysed by ultrafiltration and PAGE-SDS electrophoresis. This diploma thesis was done within the international project ,,LipoFungi“.

Characetrization of selected microbial enzymes

This bachelor´s thesis is focused on controlled production and identification of extracellular microbial hydrolytic enzymes by fungi. Theoretical part deals with characterization of selected hydrolytic enzymes, their properties, possibility of production and application. In experimental part the production of enzymes by fungal strains Phanerochaete chrysosporium, Aureobasidium pullulans and Aspergillus oryzae was performed. Cultivation was conducted in submersed mode in mineral medium and in media with waste co-substrates such as wheat bran, sawdust, rapeseed cake (lipids content 2,55 %) and rapeseed cake rich in lipids (9 %). The activity of cellulases, xylanases, amylases, ligninperoxidase, manganese-dependent peroxidase and laccase was monitored during cultivation process and regularly on 3rd, 7th, 10th and 15th day of cultivation. Production of enzymes depended on time and the subsrate type. Cellulases and xylanases were produced mainly on 3rd and 7th day of cultivation, amylases on 3rd and 15th day and lignolytic enzymes on 7th and 15th day. Samples were further separated and analyzed by ultrafiltration, gel filtration and PAGE-SDS electroforesis.

The Basic Issues in Expertising

This diploma thesis deals with basic questions of expert activities and a more detailed focus on issues coming up in relation to practical expert activities working out expert opinions, especially obtaining materials from executive bodies, authorities, partaking parties or the ordering party. This thesis consists of a questionnaire construction containing expert opinion by professional witnesses, of an analysis of the current legislation and consequent suggested options how to deal with the given issues.

Development of Singing Skills

The Bachelor thesis deals with improvement of singing skills. It is divided into two parts. First part describes parts of respiratory tract, techniques for correct breathing during singing, articulation and working with voice. The conclusion of the first part includes guidelines for styling of the singing tone. The second part follows up with the first part. It describes exercises for practicing of singing skills in a fun way.
