Implementation of Antiplatelet Therapy Monitoring on the Analyzer Multiplate

This study describes the implementation of antiplatelet therapy monitoring on the analyzer Multiplate that works on the principle of impedance aggregometry. We used to determine four tests - ASPItest used for monitoring acetyl salicylic acid (ASA) therapy, ADPtest and ADPtestHS used to monitoring thienopyridines treatment and TRAPtest , which is used to monitoring treatment with GPIIb/IIIa inhibitors. The aim of this study was to verify the accuracy of this method for individual tests, to determine its own reference limits and compare them with literature data and on a group of cardiac patients receiving dual antiplatelet therapy (aspirin 100 mg per day and clopidogrel 75 mg per day) to assess any resistance to these drugs. Accuracy of the method we verified the group of 10 blood donors without any treatment. Each sample was repeatedly measured 5 times and from the measured values we calculated coefficients of variation. Our values of coefficients of variation from the values given by the manufacturer (CV 6 - 10 %) did not differ significantly (ASPItest: CV 8.7 %, ADPtest: CV 8.8 %, ADPtestHS: CV 13.3 % and TRAPtest: CV 12.1 %). Filip Vrbacký Title of diploma thesis: Monitoring of the expression of mRNA for CD105 in patients with the chronic lymphocytic leukemia Chronic lymphocytic leukemia (CLL) is heterogenous disease with very variable clinical course. Early determination of prognosis of this disorder can be helpful for finding and initiation of adequate individual therapy. Angiogenesis has prognostic potential and plays probably important role in pathogenesis of CLL. Endoglin (CD105) and angiopoietin 2 (Ang-2) also belong to angiogenic molecules. Data about expression of these molecules are still insufficient. Therefore, we quantitated CD105 and Ang-2 mRNA in B cells of 49 untreated CLL patients by using the method of real-time quantitative PCR. Subsequently, we compared the expression of these angiogenic molecules to traditional as well as modern biological prognostic factors(Rai clinical stage, disease course, IgVH mutation status, expression of ZAP70 and CD38). In the peripheral blood smear and in the bone marrow smear, we find the typical leukemic cells which are characteristic by their size, morphology of cell nucleus and cytoplasm. They are bigger than healthy lymphocytes, and they have most frequently the kidney shaped nucleus with fine net-like chromatin and the cytoplasm projecting into fine salients all over the cell perimeter. In the blood count we usually find a mild pancytopenia. To demonstrate the presence of the pathological cells in the peripheral blood smear and the bone marrow smear, we use the panoptic staining method according to Pappenheim. 1 ABSTRACT This bachelor theses deals with anaemia, one of the most common hematological diseases. The introductory part of this work defines the possible causes of this disease and its clinical picture. The following part includes the logical division of different types of anaemia accompanied with examples of real cases. Main focus of the theses was put on sideropenic and megaloblastic anaemia, as they occur considerably in the population.

The importance of the determination of activated partial thromboplastin time and partial thromboplastin sensitivity compared to the lupus anticoagulant, deficiency of coagulation factors and heparin Dagmar Drábková Abstract Activated partial thromboplastin time (aPTT) is coagulation test which detect the disturbances of hemostasis and it is the most common test for monitoring unfractioned heparin therapy. In parcial tromboplastin are present activators of contact phase and phospholipids which can significantly affect the sensitivity of reagents to coagulation factors, lupus anticoagulans and heparin. The objective of this study was firstly to compare the sensitivity of parcial tromboplastins with different origins of fosfolipids and various activators to lupus anticoagulans, to decreased and increased levels of coagulation plasmatic factor VIII and sensitivity to standard heparin. Secondly, according to measured data to determine whether any of the selected reagents is suitable for monitoring heparin while being able to detect deficit of coagulation factors and presence of inhibitor. Jana Šklíbová: An examination of trombofile statuses the territory of the Ústí region and monitoring of treatment Thrombophilia is a state of the circulatory system that is characterised by increased probability of thrombus formation. Thrombophilia is defined as either inherent or acquired defect in haemostasis which, with the highest probability, causes heightened inclination to thrombosis. However, the presence of the thrombophilic defect does not necessarily mean that the bearer will suffer from thrombosis. Thromboembolism is a serious but rather underestimated medical phenomenon that causes a lot of health trouble to patients as well as rising health- care costs. In the Czech Republic, up to 10,000 people a year die of or in relation to fatal pulmonary embolism. Thromboembolitic complications can have many causes and even external factors, such as injuries, operations, immobility, etc., are known to affect their occurrence. There exist several prothrombotic states that are hereditary and of genetic nature, e.g. Factor V Leiden and the mutation on the gene responsible for the production of Factor II Prothrombin.
