Voltammetric Determination of 5-Nitroimidazole - The Structural Unit of Nitroimidazole Drugs

Presented Diploma Thesis deals with the study of electrochemical behavior of 5-nitroimidazole (5-NI), with the search for optimal conditions for its determination using techniques of DC voltammetry (DCV) and differential pulse voltammetry (DPV) at a mercury meniscus modified silver solid amalgam electrode (m-AgSAE) and at a boron-doped diamond film electrode (BDDFE), and with the comparison of reached limits of quantification (LQs). The optimal conditions found for measuring calibration dependences of 5-NI at the m-AgSAE in deionized water were as follows: Britton-Robinson buffer of pH 7.0 and 10.0 for DCV and DPV, respectively. At both thechniques, it is advisable to use an electrochemical regeneration of the electrode surface by the application of a couple of regeneration potentials Ereg,1 = 0 mV and Ereg,2 = −800 mV. The calibration dependences were measured in the concentration range from 1·10−6 to 1·10−4 mol·l−1 , with the LQs of 1.0·10−6 mol·l−1 for DCV at the m-AgSAE and 1.5·10−6 mol·l−1 for DPV at the m-AgSAE. The concentration dependences of 5-NI at the BDDFE were measured using the DCV technique in the BR buffer of pH 3.0. 4. Conclusion This Thesis represents a contribution to the search for new analytical methods applicable on environmental and biological samples. The current state- of-art concerning the formation, occurrence, and biological activity of fluorene and its nitro and oxo derivatives was summarized in Chapter 1. Further experimental work was focused on assessment of new approaches in the detection of the studied compounds. Attention was paid mainly to nitrofluorenes, fluorenone and nitrofluorenones, namely 2-nitrofluorene, 2,7-dinitrofluorene, 9-fluorenone, 2-nitro-9-fluorenone, and 2,7-dinitro-9- fluorenone and the possibilities of their determination using polarographic and voltammetric methods. Interactions of calf thymus dsDNA with 2-nitrofluorene and 2,7-dinitrofluorene at the DNA/SPCPE were investigated and subtle DNA damage under conditions of direct DNA-analyte interaction at room temperature and damage to DNA bases under condition of electrogeneration of short-lived radicals of nitrofluorenes at human body temperature were found. Electrochemical determination of carboxine using carbon paste electrode

Pesticides are substances widely used for preventing damage by weeds, insects and fungi on plants, feedstuffs, foodstuffs, etc. Those compounds were often overused and this resulted in widespread contamination of the environment. Water ecosystem is the most endangered one - pesticides possess a huge risk for living organisms and primary sources of drinking water. Therefore, development of new analytical methods utilizable for reliable monitoring of these substances in environment is of necessity. Main objective of this diploma thesis was finding the optimal conditions for determination of carboxin using differential pulse voltammetry (DPV), high-performance liquid chromatography with spectrophotometric (HPLC-UV) and electrochemical (HPLC-ED) detection, latter using carbon paste electrode (CPE) as working electrode, reaching the lowest possible limits of detection (LD). Next objective was the application of the developed methods for the determination of carboxin in the sample matrix of river and drinking water. A system for monitoring the quality of connectivity

The aim of the theses is to design a methodology for centralized monitoring of data traffic in an ISP network that would detect individual connection with connectivity problems, and evaluate these problems. The methodology should at least evaluate packet loss and latency between a centrally located monitoring point and endpoints at user premises. A further goal is to implement this methodology, test it and evaluate the results.

Osteoporosis belongs between the most common civilization diseases. Insufficient vitamin D supplementation attributes to pathogenesis of the illness. Aim of the study was vitamin D levels monitorning in both healthy population and vitamin D treated patients. I have proved deficit of vitamin D at these patiens, over recommended dosage of vitamin D. Vitamin D deficiency starts to be probably more serious problem than ever before. Large-Surface Carbon Film Electrode - A Novel Sensor for Voltammetric Determination of Electrochemically Oxidizable Organic Compounds

of the Diploma Thesis In this Diploma Thesis, the electrochemical behavior of 4-nitrophenol (4-NP) was studied at a newly prepared large-surface carbon film electrode (ls-CFE) using techniques of DC voltammetry (DCV) and differential pulse voltammetry (DPV) with the aim to develop sensitive analytical methods for its determination. Voltammetric behavior of 4-NP was investigated in the region of anodic potentials, in dependence on the pH of the medium used (realized using Britton-Robinson buffer). The optimum pH values for the determination of 4-NP were chosen as follows: pH 3.0 (for DCV) and pH 7.0 (for DPV). During anodic oxidation of 4-NP on the ls-CFE at the concentration of the analyte of 1·10-4 mol/l, the passivation of the electrode surface occurred. Thus, it was decided to record series of measurements always at a new carbon film. Using the sample of 4-NP (at the concentration of 1·10-4 mol/l), the repeatability of the application of individual carbon films was tested, with obtained RSD values of 3.7% and 3.6% for DCV and DPV, respectively.
