Complex urban study of the public space Sportovní, Drobného, Tř. gen. Píky, Porgesova

The subject of the masters thesis is an urban solution of the area circumscribed by the streets Sportovní, Drobného, tř. Gen. Píky, Porgesova. The emphasis is put primarily on restoring and enhancing the central point, which is the football stadium, building more effective conection to nearby traffic hubs and revitalising the surrounding parks. Estimated result is to bring back the sport function to the area as well as overall increase of the interest from public.
Factors important for Cajal body formation

This research describes the structure and function of nuclear domains called Cajal bodies (CB). CB contain proteins and factors involved in assembly and modification of snRNPs. These bodies are found in vertebrates and invertebrates and even plants. Not all cell types contain CB. Their number and size depends on the transcription activity of cell and cell cycle phase. This paper discusses the factors that affect the CB formation. One of the most important factors is the level of snRNPs and transcription activity. Recently shows that an important role in CB formation has coilin and other components phosphorylation. Other works show the influence of the environment. The relationship between chromatin organization and RNA splicing

It is well known that RNA splicing and other pre-mRNA processing reactions happen cotranscriptionally. Surprisingly, there were recently discovered some chromatin features that had uneven distribution between exons and introns, which directly links chromatin organisation to splicing. This work summarizes all the studies that detected these chromatin patterns on exons and discuss their inconsistencies. In these studies nucleosomes were found to be preferentially positioned on exons, specific histone modifications and DNA methylation were also enriched on exons. These local patterns of chromatin organisation were evolutionarily conserved from mammals (human and mouse) to worm C. elegans and fly D. melanogaster. Their findings indicate that the role for chromatin structure in pre-mRNA splicing is to promote exon recognition. There are two mechanisms proposed for this role of chromatin in splicing. The first one is influence on RNA polymerase II elongation speed, and the second is specific recruitment of splicing machinery. In the near future we can expect studies searching for concrete examples of these two mechanisms and assessing their significance. Indeed it was reported very recently that H3K36me3 regulates alternative splicing via the second mechanism.
Genetic interactions of PRP45 mutantion in S. cerevisiae

Human SKIP/NcoA-62/SNW1 ortholog proteins are nuclear factors which have been found in connection with the regulation of gene expression and subsequent modifications of primary transcripts. Human representative SKIP acts primarily as a transcription factor during the processes involving differentiation, carcinogenesis and other cellular signaling. It operates in the signaling pathways, eg TGFß/Smad, E2F/pRb, Notch/CBF1 and nuclear hormone receptors. SKIP protein is also essential for pre-mRNA splicing and recently its contribution during the elongation phase of RNA polymerase II transcription and contribution to some HIV genes alternative transcripts formation has been identified. The ortholog of S. cerevisiae FUN2O/Prp45 has been shown to participate in pre-mRNA splicing, others processes mentioned have not yet been associated with the function of this factor. Pre-mRNA splicing is a complicated process leading to the formation of native mRNA lacking non-coding regions, introns. In terms of composition of the spliceosome subunits and biochemical principle of its function is this process very well described but despite this knowledge the specific roles of most splicing factors are not known.
