Korean buddhism in Czech Republic focusing on Zen school Kwan Um
Neuhauserová, Lucie ; Horák, Tomáš (advisor) ; Kříž, Martin (referee)
This thesis traces the history of the Korean Kwan Um School of Zen in the Czech Republic using the oral history approach. The first part of this thesis is designed as an introduction, aiming to contextualize the Kwan Um School within the broader Buddhist context. The second part of the thesis works with five interviews with current and former members of the school that were conducted for the purpose of this thesis. These interviews were subsequently processed using oral history approaches, forming a narrative that traces the history of the school in the Czech Republic. The history begins to unfold in the late 1970s when the teachings of the school started permeating socialist Czechoslovakia even before its formal establishment as the unified Kwan Um School of Zen. The thesis additionally tracks the progression of the school throughout the 1990s, as it underwent formalization in its relationship with the state, leading up to the present era.

Preparation and characterisation of mRNA/DNA transfection vectors
Horák, Tomáš ; Chmelíková, Larisa (referee) ; Skopalík, Josef (advisor)
This diploma thesis deals with genetic engineering, especially the transfection of DNA into MSCs (Mesenchymal stromal cells) and dendritic cells. Both lipoplexes and metal magnetic nanoparticles were tested to introduce the vector into cells. The research was focused on finding more efficient methods of transfection. According to analysis on MADLS and gel electrophoresis, aspects playing an important role in conjugation and subsequent transfection were found. Conjugation occurs after only 4 minutes, as evidenced by an increase in zeta potential, but to achieve full conjugation it is necessary to incubate the sample for 20 minutes. Incomplete conjugation to iron nanoparticles resulted in strong carrier-carrier interactions, which formed an unwanted conglomerates. Encapsulation into liposomes with cationic surface treatment was without complications. The success rate of GFP-labeled protein expression after transfection by these methods was calculated to be 95%, resp. 91%. This result is due to low cytotoxicity. However, commercial tested kits on dendritic cells had a success rate below 5% with high cytotoxicity.
Application of Linz's Theory of Non-Democratic Regimes to the Republic of Korea (1948-1960)
Štraicherová, Michaela ; Šamánková, Karolína (advisor) ; Horák, Tomáš (referee)
(in English): This paper aims to examine the categorization of the regime of President I Sŭng-man, who ruled the Republic of Korea in the years 1948-1960, according to Linz's typology of non-democratic regimes. Classification into a certain typology can be problematic because, as the Czech political scientist Marek Ženíšek wrote, typologies are not permanent dogmas, but serve us to understand a specific case. Commonly one case can be classifiable in several types at once and another case can be unclassifiable in a particular typology. In the first part, the author of the thesis concentrates on presenting the problem with terminology that we can encounter when studying non-democratic regimes. After that, she will give a brief overview of the political system of the Republic of Korea, where, among other things, she will specifically focus on constitutional amendments during the administration of President I Sŭng- man. In the last chapter, the author summarizes views on President I Sŭng-man. In the second part, she will then present Linz's theory of non-democratic regimes in detail, where she will focus in more detail on sultanistic regimes from the category of traditional authorities and personal rulership and two categories of authoritarian regimes - post-colonial mobilizational authoritarian...

Morphological characteristics of the Korean dialekt spoken on Cheju, comparison with standard Korean and analysis of the Middle Korean elements
Mazaná, Vladislava ; Zikmundová, Veronika (advisor) ; Horák, Tomáš (referee) ; Ferklová, Blanka (referee)
The aim of this dissertation is to describe the morphological specifics of the dialect of the South Korean island of Jeju. In 2010, the Jeju language was enlisted on the UNESCO list of critically endangered languages. This initiative led to the creation of many projects to support, protect and revitalize the Jeju dialect. Although the Jeju language is classified as a regional dialect in terms of Korean language policy, due to its incomprehensibility with the standard variety, the authorities of the Jeju Special Self-Governing province promote the designation "jejuŏ", the Jeju language. The introductory theorectical part of the dissertation introduces the reader to the issues of Korean language policy, the status of standard Korean and the regional dialects of the Korean language. In this context, the position of the Cheju dialect is described from the point of vies of language policy and from a sociolinguistic point of view. Attention is paid to the ongoing standardization of the Jeju language, as it is closely related to the current state and the development of this language. The next part presents the Jeju language, its phonological and morphological specifics, vocabulary, which is significantly marked by language contact with the languages of the surrounding great powers. The practical part of...
