Jandová, Karolína ; Mikešová,, Martina (referee) ; Trachtová, Štěpánka (advisor)
The theoretical part describes soft smeared cheeses, then deals with the technology of manufacturing said cheese with importance on the development of microbiome and describes the most commonly wanted microorganisms and contaminants. Shortly also deals with legislation requirements on the microbial quality of these chesses. The experimental part studies the development of microbiome and contamination by fungi of soft smeared cheese. Firstly, samples were collected in manufacture from each step of manufacturing. In the first part, microbial and microscopical assessment was done. In the second part, DNA was isolated from samples by a commercial kit and phenol-chloroform extraction. The concentration and purity of isolates were determined by spectrophotometry. By qPCR of the DNA isolates the main microbiome and contamination by fungi were examined. As part of the microbial and microscopical assessment, the expected bacteria were determined, and several species of yeast were found, instead of the expected one. Mold contamination was also found. Using molecular diagnostic techniques, the presence of bacteria, yeasts, fungi and the genera Brevibacterium and Candida was proved.

Changes in the composition of the company's fleet in relation to the EU's "green deal" objectives
Tobiášová, Eliška ; Jandová, Kristýna (referee) ; Bača, Petr (advisor)
The purpose of this master thesis is to examine the so-called Green Deal and the related directives and regulations that are intended to help achieve the objectives set out in the Green Deal, as well as to analyse the fleets of the Member States of the European Union and the company with which cooperation has been established as part of this work. The thesis describes the Green Deal and its objectives. The European Union directives and plans with the aim of reducing CO2 that are currently approved or are yet to come into force are also mentioned, while at the same time they are compared with the directives and targets in force in the EU Member States, including the Czech Republic. In the following chapters, a brief description of the cars by type of fuel is given in order to follow a detailed analysis of the fleets of Germany, Sweden, Spain, Slovakia, France, Poland and the Czech Republic. This analysis is carried out using graphs and official statistics available from government institutions. Furthermore, an analysis of the company's fleet is made, summarising in tables the parameters of the vehicles available to the company. The results of the fleet analysis are also summarised in graphs based on the year of vehicle manufacture, emissions, and fuel type. In addition, an analysis of hybrids and electric vehicles currently in the company's fleet is also provided. This analysis works with data provided by the company and examines the cost of driving these vehicles, consumption, and battery usage ratio. Finally, a suggestion is made for the replacement of the current vehicles with the aim of saving emissions.
Study of the ecological impact of production, operation and disposal of various types of cars
Kutnar, Václav ; Jandová, Kristýna (referee) ; Jaššo, Kamil (advisor)
To reduce greenhouse gas emissions, countries have focused on (among other things) automobile transport. However, there are different car powertrains, and each produces a different amount of emissions from different parts of their lifecycles, which also vary from state to state. Knowing exact, comparable values is necessary for deciding how to reduce these emissions most efficiently and economically possible. Existing studies often focus on only one part of the life cycle and/or are not localized to a particular state. This study analyses the entire life cycle of a vehicle and its fuel or energy for a vehicle manufactured, operated, and recycled in the Czech Republic. It draws on existing studies, supplemented by data from state agencies, vehicle manufacturers, and our calculations. It was found that in the Czech Republic, compared to a vehicle with a petrol internal combustion engine (ICEVg), an electric vehicle (BEV) has 33% lower total GHG emissions, and a vehicle with hydrogen fuel cells (FCEV) 19% lower. However, an internal combustion engine vehicle has lower emissions from its production and as a result, its cumulative emissions are lower than an electric vehicle until 70,000 km driven and a hydrogen fuel cell vehicle until 111,000 km
Measurement and creation of irradiance maps of irradiated surfaces

This diploma thesis deals with the measurement and creation of irradiance maps of irradiated surfaces. In the first part of the thesis, there are defined light and light radiation and its types and there are explained the basic concepts closely related to light radiation. Furthermore, this thesis also describes different types of radiation sources and their effect on irradiated surfaces. The theoretical part concludes with a chapter on radiation sensors, specifically pyranometers. In the second part of the thesis, the intensity of the radiation incident on the selected surface was measured by the selected radiation source, and then the radiation distribution was simulated using the Agilent VEE Pro 9.2 software environment. Subsequently, the measurement and its simulation were processed and evaluated.
Critically endangered Large Heath butterfly in Šumava: situation, trends and threats
JANDOVÁ, Kateřina
The main goal of this work was to map the occurrence of the large heath (Coenonympha tullia) from the Nymphalidae family in the Bohemian Forest National Park. This species currently occurs in the Czech Republic only in the Bohemian Forest peatlands up to an altitude of 900 m above sea level and very rarely in the Třeboň region (Hanč et al., 2019). Monitoring took place in June 2023 in the Bohemian Forest peatlands in the Moldau floodplain. I visited in total 51 sites with a potential distribution of the species. The large heath (Coenonympha tullia) was found in 11 of them. I monitored also several potential predictors of the butterfly occurrence, including an information about the distribution of its host plants, cottonsedge (Eriophorum vaginatum) and common cottongrass (Eriophorum angustifolium). Subsequent statistical comparison of various indicators at the sites from 2023 with monitoring from 2021 (Fric and Walter, 2022) showed that indeed the Bohemian Forest peatlands are drying out. After comparing the number of large heath (Coenonympha tullia) individuals, it was found that the number of individuals in 2023 was higher than in 2021.
