Peptide profilig of tryptic digests of water soluble proteins from Bacillus thuringiensis - transgenic and non-transgenic maize species by capillary zone electrophoresis
Sázelová, Petra ; Kašička, Václav ; Ibanez, E. ; Cifuentes, A.
The tryptic digests of water-soluble proteins from transgenic Aristis-Bt and non-transgenic Aristis and Coventry maize varieties were analyzed by capillary zone electrophoresis (CZE) with UV-absorption detection at 206 nm in several acidic background electrolytes. Resolved tryptic peptides were characterized by effective electrophoretic mobilities and corrected peak areas. Some significant qualitative and quantitative differences in CZE-UV profiling of tryptic peptides were found between Aristis Bt and Aristis or between Aristis and Coventry varieties.
Recent applications of capillary electrophoresis to analysis and physicochemical characterization of peptides
Kašička, Václav ; Koval, Dušan ; Ehala, Sille ; Sázelová, Petra ; Šolínová, Veronika ; Hlaváček, Jan
Recent developments in the methodology of capillary electrophoresis are presented with respect to their application to analysis, separation and physicochemical characterization of biologically active peptides. New strongly acidic/basic classical and isoelectric background electrolytes were applied to high-sensitive analysis and high-efficient separations of structurally related peptides, such as insect oostatic and antimicrobial peptides, phosphinic pseudopeptides and tryptic peptide fragments of proteins from native and genetically modified maize species.
Enantiomer separation and determination of helix inversion barrier of helical dications by capillary electrophoresis
Koval, Dušan ; Adriaenssens, Louis ; Severa, Lukáš ; Teplý, Filip ; Sázelová, Petra ; Kašička, Václav
Enantiomeric helical forms of dicationic helquats, a novel family of charged helicene-like molecules, have been separated by capillary electrophoresis (CE) using sodium-phosphate buffer, pH 2.5, as background electrolyte and sulfated beta- and gamma-cyclodextrins as chiral selectors. Additionally, CE was used to determine barrier to helix inversion of helquats.
Extraction of water-soluble proteins from Bt-transgenic and non-transgenic maize species and tryptic digests peptide profiling by capillary electrophoresis
Sázelová, Petra ; Kašička, Václav ; Ibanez, E. ; Cifuentes, A.
Water-soluble proteins extracted from Bt-transgenic and non-transgenic maize varieties were digested by trypsin and analyzed by capillary zone electrophoresis (CZE) in several acidic background electrolytes. The found qualitative and quantitative differences among CZE profiles of tryptic peptide maps can be potentially used to differentiate transgenic and non-transgenic maize species.
Capillary electrophoresis employed for quantitative characterization of peptide interactions with small ions and cyclodextrins
Kašička, Václav ; Ehala, Sille ; Šolínová, Veronika ; Schimperková, Tereza ; Sázelová, Petra ; Koval, Dušan ; Makrlík, E.
Capillary affinity electrophoresis has been applied to investigate quantitatively two types of non-covalent peptide interactions: i) complexes of pseudopeptide valinomycin with ammonium ion and alkali metal ions, Li+, Na+, K+, Rb+ and Cs+, in methanol, and ii) complexes of enantiomers of dipeptide beta alanyl-D,L-tyrosine and its derivatives with chiral selector, 2-hydroxypropyl-beta-cyclodextrin, in water.
Analysis of water soluble proteins and peptides from Bt transgenic and non-transgenic maize varieties by capillary electrophoresis
Sázelová, Petra ; Kašička, Václav ; Ibanez, E. ; Cifuentes, A.
Water-soluble proteins and peptides extracted from the transgenic Aristis-Bt and non-transgenic Aristis and Coventry maize varieties were analyzed by capillary zone electrophoresis (CZE). The found qualitative and quantitative differences among CZE profiles of these protein/peptide extracts from transgenic and non-transgenic maize species can be potentially used to detect transgenic maize in foods.
Investigation of peptide complexes with small cations and cyclodextrins by capillary affinity electrophoresis
Kašička, Václav ; Ehala, Sille ; Šolínová, Veronika ; Schimperková, Tereza ; Sázelová, Petra ; Koval, Dušan
Capillary affinity electrophoresis was applied to quantitative evaluation of the strength of the complexes of i) valinomycin, macrocyclic dodecadepsipeptide ionophore, with small cations (ammonium and alkali metal ions, Li+, Na+, K+, Rb+ and Cs+) in methanol, and ii) enantiomers of dipeptide beta-alanyl-D,L-tyrosine and its derivatives with a chiral selector, 2 hydroxypropyl-beta-cyclodextrin (2-HP-beta-CD), in water.
Predikce tvaru molekul a separace strukturně příbuzných hormonů uvolňujících gonadotropin (GnRH) kapilární elektroforézou
Šolínová, Veronika ; Kašička, Václav ; Sázelová, Petra ; Barth, Tomislav
Semiempirické modely závislosti efektivní mobility peptidů na poměru náboje a relativní molekulové hmotnosti byly použity k popisu elektromigračního chování peptidů, zejména pak k předpovědi tvaru jejich molekuly.
Užití kapilární zónové elektroforézy a izotachoforézy k vyhodnocení stupně čistoty a obsahu protiiontů v preparátech lecirelinu
Sázelová, Petra ; Kašička, Václav ; Šolínová, Veronika ; Flegel, Martin ; Barth, Tomislav
Různé šarže lecirelinu, analogu LH-RH, byly studovány co do čistoty. Hmotnostní procento nečistot bylo stanoveno metodou CITP.
Separace a charakterizace peptidů kapilární elektroforézou v silně kyselých/bazických základních elektrolytech
