Obtaining and complex charaterization of Aronia spp. extracts

Aronia (Aronia melanocarpa) is a berry fruit with distinctive sensory characteristics and health-promoting properties. Polyphenols are the main bioactive compounds found in aronia including natural pigments – anthocyanins. Bioactive compounds are usually obtained from natural materials by extraction, in this work, two methods of extraction were compared – maceration and PHWE. Based on the total phenolic content, maceration was chosen as a more suitable method and then was optimized with statistical model – Design of Experiment. Optimal conditions were set to temperature of 30 °C, extraction agent 50 % ethanol, solid-solvent ratio 10 g per 50 ml and time of extraction for 30 minutes. Extract obtained under these conditions was characterised by total phenolic content – 1441 ± 90 mg/100 g DW, total anthocyanin content 943 ± 8 mg/100 g DW and antioxidation activity by ABTS 24,78 ± 0,09 molTE/g. In total of 25 volatile compounds were indentified in the optimal extract with content > 0,5 %. Major part created aldehydes and esters, with the main compounds being benzaldehyde (32,25 %) and methyl hexanoate (21,37 %).
Impact of temperature and drought on gliadins content in two varieties of wheat

This bachelor’s thesis focuses on the effect of high temperature and drought on protein content of gliadin fraction on two varieties of wheat. Chosen varieties were Hyfi and Julie, cultivated at 26, 29, 32, 35 and 38 °C during flowering in watering controlled conditions. The condition for ‘wet’ samples was at least 70 % soil moisture and for ‘dry’ samples less than 30 % soil moisture. After harvesting, the seeds were milled into flour from which the gliadins were extracted with 2-chlorethanol. A-PAGE method was used for gliadin separation, quantification was carried out through computer densitometry. A significant genotype effect was discovered. Whilst temperature ranging from 26-38 °C with simultaneous drought stress had no significant effect on gliadin content of Hyfi variation, gliadin content of Julie variation shown obvious maximum at 32 °C. Therefore, Hyfi variation shown better resistance to heat stress than Julie variation. Both variations had higher gliadin content under drought stress than under good watering conditions.

The Symbolic of Colours in the Apocalypse of Saint John

Anglická anotace Symbolic Colours in the Apocalypse of Saint John This thesis attempts to study the use of colours in the depiction of certain themes in the Apocalypse of st. John. The colours used by the author of Revelation are taken both from apocalyptic literature of the Old Testament era, but mainly from the books of Daniel, Ezechiel, Zacharias and Joel. The most often used colour is white: the white stone (Rev 2,17); the white cloud (Rev 14,14); the white horse (Rev 19,11); the white throne (Rev 20,11). Most likely the reason is that the author wanted to encourage his contemporaries in time of persecution. An exception is the text Rev 6,2, where white is used in a concrete historical event. Red or scarlet is used in opposition to white: the red horse (Rev 9,3-4); the red body-armour of locusts (Rev 9,1-21); purple (or scarlet) in Rev 17,4. The author also names colours of such minerals as emerald, ruby and jasper (Rev 4,3). However most attention is paid to Revelation 6, where four riders appear on four horses of different colours. My endeavour therefore was to find as many possible interpretations of colours in the Revelations of st. John as proposed in various biblical lexicons, biblical periodicals and books dealing with this unexplored fascinating topic. Key words New Testament Symbolic Colours...
