Charles University, Faculty of Pharmacy in Hradec Králové Department of Analytical Chemistry Candidate: Zdeňka Němcová Supervisor: Assoc. Prof. PharmDr. Lucie Nováková, Ph.D. Title of Diploma Thesis: Optimization and validation of UHPSFC-UV methods for quality control of agomelatin and atorvastatin in drug substance and in tablets The aim of this master thesis was to develop two ultra-high performance supercritical fluid chromatography (UHPSFC) methods with PDA detection for determination of agomelatine and atorvastatin and its potential impurities. UHPSFC system Acquity UPC2 with PDA detector was used for measurement. The separation of agomelatine was performed by Torus Diol column (100 × 3.0 mm, 1.7 µm). Gradient elution was performed using CO2 with the mixture of methanol/acetonitrile (1:1) and 31.5 mM (0,1 %) ammonium hydroxide as a mobile phase. Column temperature was set at 40 řC and BPR (back pressure regulator) pressure at 2000 psi. The detection wavelength was set at 225 nm. The method was developed for measurment of API (active pharmaceutical ingredient) and subsequently for tablets. The separation of atorvastatin was performed using Acquity UPLC HSS C18 SB column (100 × 3.0 mm, 1.8 µm). Gradient elution was performed using CO2 with the mixture of methanol/ acetonitrile (2:1), 15 mM...

ACCESSABILITY OF PUBLIC INSTITUTIONS AND SERVICES IN THE TOWN OF SOKOLOV FOR PEOPLE WITH A LOWERED MOVEABILITY
NĚMCOVÁ, Zdeňka
Bachelor thesis is divided into two parts, theoretical and practical. In the theoretical part with the use of literature I deal with the problems of people with disabilities and barriers in human life. It is the real fact that among us there is certain number of people living with disabilities. For people afflicted with such kind of disability is difficult to access buildings which are not adapted. These people have the same right to a full life, to equality, dignity, independent living and full participation in society as well as humans for which architectural barriers are not an obstacle. These barriers make it difficult or completely impossible to move people with disabilities in such environment. The aim of this work is to map the availability of public institutions and services in the town of Sokolov for people with reduced mobility and find out the information about facilities of selected public buildings if they are equipped with toilets for invalid people. Part of the research related to movement in a wheelchair. In this undergraduate work were answered two research questions: Are the facilities providing health services available, in terms of physical access to persons with reduced mobility? Do the monitored public buildings have barrier-free access to toilets? For monitoring of selected public buildings was chosen qualitative research, so called , participant observation technique. During this research some people participated the life condition together with people for whom the use of wheelchair is a common part of their lives. This research was conducted from 18 1st 2011 until 30 3rd 2011 Research has shown that facilities that provide health services are accessible to people with reduced mobility but on the other hand also showed that not all observed public buildings are equipped with wheelchair access and toilets for people with reduced mobility. Currently the situation is gradually improving and, as the time goes people realize the considerable responsibility for removing barriers.
