Abstract The bachelor´s thesis is focused on the perception of the authority of teachers in secondary schools. The work is divided into theoretical and practical part. The aim of the theoretical part is to define the concept of authority, the division of types of authorities, the comparison of authority at various levels of schools. It tries to describe the personality of the teacher and focuses on the relationship between students and secondary school teachers. In the theoretical part, I compared and analysed theoretical knowledge from various professional sources. The aim of the practical part is to find out how students at secondary vocational schools perceive the concept of authority in the secondary school environment, what they imagine behind it. What personalities of educators as bearers of authority they prefer and how they approach to them. I also focused on the view of students, how they look at the characteristics of teachers and the differences of opinions with respect to gender. Based on the collection of data from the questionnaire survey, evaluation, and analysis of data, I tried to propose recommendations for pedagogical practice. The research was carried out by a questionnaire, whose respondents were students at secondary vocational schools.
Study of the interaction of receptor NKp46 with adhesin Epa1

One of the key components of the innate immune system are natural killer (NK) cells. The task of these cells is to induce apoptosis in target cells (e.g., cancer or virally infected cells). The target cells are identified by their interaction with surface receptors of the NK cells. On the surface of the NK cells, there are activating and inhibiting receptors. One of the activating receptors is the natural cytotoxicity receptor NKp46. Several ligands of this receptor have been identified, one of them being the epithelial adhesin Epa1 of yeast Candida glabrata. The invasive candidiasis caused by this yeast is a feared complication for patients with haematological diseases. The use of the NK cells in immunotherapy includes bispecific fusion proteins which can bind to the NK receptor with one part and to tumour antigen with the other part. This work focuses on recombinant preparation of the NKp46 protein. To facilitate a study of the effects of O-glycosylation on the binding of the ligands, a mutation of the glycosylation site NKp46 T225A was prepared. A stably transfected HEK293S GnTI- and HEK293T cells had been prepared and these proteins were then extracellularly secreted. Natural killer cells (NK cells) are cells of innate immunity that play an essential role in the immune response of an organism. In contact with infected, stressed or tumour cells, the NK cells can trigger cytotoxic mechanisms. The initiation of mechanisms depends on the presence of activating or inhibitory ligands on the surface of the cells. On the surface of the NK cells, there are activating and inhibiting receptors that upon binding their respective ligands send a signal to the NK cell. One of the activating mechanisms is the decrease of expression in MHC gp I molecules on the surface of the infected cells. This molecule is a ligand of the inhibiting receptors. One of the activating receptors of NK cells is NKp46. This receptor belongs to the natural cytotoxicity receptor (NCR) family. NKp46 has many ligands, one of them being the adhesin Epa1 of yeast Candida glabrata. This thesis aims at preparing plasmids and producing extracellular domains of the NKp46 receptor and its ligand adhesin Epa1.The interaction of NKp46 and its ligand Epa1 remains to be the subject of future research. Performance system of employees in a financial institution

This master thesis discusses employees appraisal in a selected organization. The theoretical part is devoted to expressions from the area of staff appraisal. Topics such as performance management, performance appraisal and work performance are clarified. Further the common process of employees appraisal is described, including its meaning and functionality. appraisal criteria are specified and selected appraisal metods are described. A specific chapter is devoted to forced distribution method. In the end of this part characteristics of high quality and effective employees appraisal are listed. In the empirical part a specific organization is presented, including description and analysis of used employees appraisal system. The goal of the research is through the analysis of the employees appraisal system and with the help of interviews with its employees determine the characteristics of a quality employees appraisal system. Based on the results of the conducted research recommendations leading to higher efectivity of employees appraisal are formed. Childhood overweight and obesity at primary school age

Childhood overweight and obesity is a present issue not only in the Czech Republic, but worldwide. The main objective of this thesis was to assess overweight and obesity in children of primary school age in towns and in villages in the Central Bohemian Region and to focus on their alimentation habits and physical activity. The thesis consists of two parts. The first theoretical part describes the characteristics of primary school age, introduction to the topic, epidemiology, causes of the origin, types of obesity, diagnostics, problems of obese children, nutrition of children and adolescents and their alimentation habits, prevention, medical treatment, specific programs for the prevention and treatment of childhood overweight and obesity and an overview of the myths about childhood obesity. The second practical part of the thesis includes questionnaire study implemented within the 4th grade of chosen urban and non-urban schools in the Central Bohemian Region and semi-structured interview with mother of a child suffering from overweight since early age. The survey included 155 pupils of the 4th grade from two urban and five non-urban schools of the Central Bohemian Region. The results of the questionnaire study proved that 23 % of urban and 20 % of non-urban pupils suffer from overweight and obesity. An important factor is their different lifestyle. Non-urban children spend their leisure time in a passive way, they prefer playing PC games to physical activities, attend less extra sport classes and consume more sweets than urban children. More urban pupils consume three to four meals on a daily basis in contrast with non-urban pupils who tend to consume five to six portions. Most prefer healthy food to unhealthy food and tend to eat mainly czech dishes at home. 90% of the questioned children expressed positive attitude to sport. More than half of the children suffering from overweight and obesity do not practice any sport or physical activities sufficiently. Due to the importance of regular alimentation and consumption of breakfast and snacks, it is a positive finding that three quarters of the children have breakfast regularly and bring home-made snacks to school. The semi-structured interview with mother of an overweight child proved that overweight is not caused by overeating and by a passive lifestyle but also by genetic predisposition. In the conclusion, the author presents the outcome of the work and a general primary prevention at family level, school level and community level.
