Modernization of Brno - Přerov railway line in Vyškov area for speed 200 km/h

The objective of this master´s thesis is a design of modernization of railway line no. 300 Brno – Přerov in Vyškov area and its vicinity for speed 200 km/h. The track section is located between railway station Luleč and Ivanovice na Hané. The track is designed in two variants. The first variant is proposed keeping railway line through Vyškov railway station including necessary modification of the railway station. The second variant suggests keeping track outside Vyškov and a connection with the railway station by bridging is designed.

Assessment of the economic efficiency of the development project

This bachelor thesis critically discusses the economic efficiency of a development project which is evaluated by means of a cash flow analysis, specifically by utilizing discounted payback period and net cash flows. The theoretical part uses research to outline the business plan and the fundamental concepts associated with the economic efficiency, followed by outlining the aims and objectives of the business plan, investment planning, and development project cash flows. The second part of the thesis introduces the actual project proposal and applies the methods discussed in part one. In the last part, this bachelor thesis evaluates the gathered data and determines the level of economic efficiency.

Analysis of plant extract of Orthosiphon stamineus by capillary zone electrophoresis

Capillary zone electrophoresis with UV detection at 200 nm was developed for the determination of four phenolic acids and four flavonoids in plant material. The electrophoretic behavior of phenolic compounds was investigated to optimize their separation as a function of buffer pH, buffer concentration. A 50m mM borate buffer, pH 9 was found most favorable. The separation was obtained with fused-silica capillary column (50 cm x 50 μm I.D.) an applied voltage of 25 kV and a temperature of 25řC. For increase injection volume and decrease the detection limits was performed by applying Large-volume sample stacking with polarity switching. The limits of detection were between 0,006 and 0,014 μg/ml. The method was used to determine these compounds in methanolic extract of Orthosiphon stamineus leaves that contained 710,9 μg g-1 pulverized drug of rosmarinic acid and 38,2 μg g-1 of caffeic acid. About a 60- fold enhancement in senzitivity, relative to a conventional injection, was demonstrated in a real sample.
