Lucie Pagáčová Allergic diathesis - the effect of familial burden and environment Diploma thesis Charles University in Prague, Faculty Of Pharmacy in Hradec Králové Pharmacy INTRODUCTION: In our study we tried to demonstrate the various factors that might influence the development of allergic diseases. We focused mainly on the genetic influence on the part of parents and pointed to the influence of the siblings. The enviroment in which probands were characterized by size grew and the size of the family residence. OBJECTIVES: We tried to clarify selected factors that may cause allergic diseases. We brought in this regard the new data that compare with studies carried out abroad. METHODS: The necessary data we obtained through a questionnaire form that was designed for mothers of 12 years old children. We used 1672 questionnaires filled correctly form which we selected a group of 740 children without symptoms of allergies and we have defined a clear 626 allergic children. These two groups we compared with factors for which we have tried to demonstrate an impact on the emergence of asthma, eczema, hay fever, perennial rhinitis, hives and other symptoms. Title of diploma thesis: Bazotests and lymphocyte transformation tests linked to drug intolerance Basophil activation tests and lymphocyte transformation tests are methods that allow drug allergy diagnosis. The right diagnosis is crucial for treatment. Drug allergy is often associated with β-lactam antibiotics hypersensitivity and especially with penicillins. The aim of this diploma thesis is to provide actual statistics based on selected patient medical reports. Those patients were completely clinically and laboratory examined. The main condition for data selection was the execution of skin tests for suspicion of drug allergy caused by penicillin-like antibiotics at the Institute of Clinical Immunology and Allergology of the Faculty Hospital Hradec Králové in 2017. Patients' anamnesis, skin tests and laboratory examination focused on basophil activation tests and lymphocyte transformation was composed. Statistics of 25 patients shows beneficial informations. This therapy is mainly used to treat chronic allergic response to antigenic substances batteries. Allergic diathesis - the effect of nutrition

BACKGROUND: In recent years we could see an apparent increasing number of allergic diseases in the population. In this context, it has been described a number of protective and risk factors that affect the development of allergy. With regarding to extent of this issue we decided to address the topic of allergies in our study. We focused on issues related to maternal and child nutrition. The aim is to compare the effect of some selected factors between groups of children (allergic and non-allergic children). METHODS: We sent a total of 2635 anonymous questionnaires to schools. We focused on children aged 12 - 13 years (seven grades students). Mothers filled the questionnaires in for their children. We returned 1672 completed questionnaires back. Children were divided on allergic and non-allergic by allergic symptoms, doctor visits and potential treatment. We investigated the influence of individual factors on the development of allergies among children. RESULTS: Statistically significant as a risk factor appeared to eat honey during pregnancy and breast-feeding twice or five times a week. Among the protective factors were included exclusive breastfeeding for at least 10 - 12 months and occasional eating of a curd during pregnancy, these factors were statistically significant. The effect of the diseases in infancy on the prevalence of allergies in children

Author: Veronika Kotrchová Title: The effect of the diseases in infancy on the prevelance of allergies in children Diploma work Charles University in Prague, Faculty of Pharmacy in Hradec Králové, study program Pharmacy Background: To find out if diseases in infancy are protective or risk factors for prevelance of allergy. We interested mainly in infectious diseases. We compared our results with so called Hygiene hypothesis, which claims that contact with microbes in early childhood boosts immature immune system due to activation of Treg lymphocytes. Methods: We sent 2793 questionnaires to 69 elementary schools all over the Czech republic. 1697 of them returned to us. We defined two groups of respondents allergics and non-allergics. For categorization we used specific criterions. According to these criterions 420 respondents were named as allergic and 774 respondents as non-alergic. For better identification if concrete disease played role as protective or risk factor for development of allergy, we chose another specific criterions. Results: We didn't prove any statistical significance between diseases in infancy and the later prevelance of allergies except for bronchitis. The use of recombinant allergens for diagnosis of allergic dieseases

Laboratory diagnosis of allergic diseases is nowadays focused on the identity of specific IgE. The detection of sensitization are often used mixed allergens from natural sources with poorly defined composition. The exact specification of the allergens are tests using recombinant allergens. Recombinant allergens are not only used for the determination of specific IgE antibodies, but also to test basophiles activation. Latest test is a microarray using recombinant allergens method. In conclusion of this work are presented case reports and the results of laboratory tests that demonstrate the practical use of recombinant allergens.
Systemic and Local Allergic Insect Sting Reaction From The Epidemiological, Diagnostic a Therapeutic Point of View

Hymenoptera venom allergy threatens the patients with severe, often life threating anaphylactic sting reaction. Proper diagnostics and treatment are therefore crucial for the patient with this diagnosis and should be performed by an experienced allergist. We aimed to provide a comprehensive look at these patients, from the epidemiological, diagnostic and therapeutic point of view. Initially, we focused on predictors of severe systemic sting reactions. Then we followed by evaluation of the utility of available laboratory diagnostic methods and by monitoring of immunological parameters during venom immunotherapy. We met all these goals. We detected higher age, sting localization in the head region and male gender as predictors of severe systemic sting reaction. We also evaluated the clinical utility of all diagnostics methods, and we even suggested the possible way, how to optimize the diagnostic procedure in patients with Hymenoptera venom allergy. We noticed significant changes in immunological parameters during venom immunotherapy. Due to a low number of stings during treatment, we have not been able to evaluate the clinical relevance of these changes yet.
