Development and characterization of light-producing deoxyribozymes

Light-producing protein enzymes such as luciferase play important roles in both applied and basic research. In this study, we used an in vitro selection to isolate deoxyribozymes that catalyze a chemiluminescent reaction by dephosphorylation of the commercial substrate CDP-Star. One of the most active variants, named Supernova, was further improved and characterized using a combination of random mutagenesis, in vitro reselection, high-throughput sequencing, comparative sequence analysis, and optimization of reaction conditions. Supernova produces light up to 6,500-fold more efficiently that the background reaction and folds into an unusual triple-helical structure. Moreover, we characterized in detail the buffer requirements including pH, the effect of various ions, substrate and Supernova concentrations, and the presence of crowding agents. Finally, we showed that Supernova can be turned into an allosteric sensor by rational design. We anticipate that this deoxyribozyme can be used as the signaling component in light-producing allosteric deoxyribozyme sensors that respond to a wide variety of stimuli and will complement existing methods that utilize radioactive, fluorescent, and colorimetric readouts.
Direct Elections of the Czech President in the Magazine Katolický týdeník

The presented bachelor's thesis deals with the issue of the Czech Catholic media. The printed periodical Katolický týdeník occupies a completely privileged position among them. The aim of this bachelor's thesis is to map how Katolický týdeník approached the topic of two direct presidential elections in the Czech Republic. These events can be considered not only as socially significant and highly monitored in the media, but their result is also relevant for the position of the Catholic Church in the Czech society due to the competencies and especially the symbolism of the presidency. The theoretical introduction outlines the position of the Catholic media in the period of presidential elections in the Czech Republic based on the reflection of the situation of the Catholic Church in the Czech Republic and its relationship to the media and the state. Other chapters present the most important Czech Catholic media, introduce Katolický týdeník as a key periodical for the analytical part and provide a brief overview of the course of the two direct presidential elections in the Czech Republic. Analysis of heat shock protein genes expression in spruce bark beetle \kur{Ips typographus} and their importance for survival upon exposure to heat

The aim of this work was to examine the expression of certain Hsp genes upon heat exposure in spruce bark beetle Ips typographus. We determined the level in unstressed and heat-exposed animals, and attempted to assess the importance of Hsp proteins for animals' survival upon heat treatment. We used RNA interference to knock down the expression of these genes, and analyzed the influence on animals exposed to elevated temperatures.
